Researchers in Brazil identified probiotic Lactobacillus strains in traditional cheeses, finding them safe and beneficial. The strains could enhance cheese quality without altering its composition, though more research is needed to confirm their industrial viability. In a new paper, researchers from Brazil discuss the benefits of three Lactobacillus bacteria strains for human health and their applications in the cheese industry. Research conducted at the Center for Dairy Technology (Tecnolat) in Campinas, São Paulo state, Brazil, has identified lactic acid bacteria (LAB) that have probiotic properties and are beneficial to human health in samples of traditional Brazilian cheeses. Tecnolat is part of the Food Technology Institute (ITAL), an arm of the São Paulo State Department of Agriculture and Supply. The researchers analyzed three strains of bacteria belonging to the genus Lactobacillus from Tecnolat’s bacterial strain bank. The bacteria were isolated from traditional Brazilian cheeses, such as Prato and Minas, for example. These and other LAB are recognized for their probiotic potential and are widely used by the food industry in products such as yogurt, kombucha, and kefir, as well as cheese. “We selected these strains after screening the entire strain bank to see which isolates had been found to have the best fermentative, enzymatic, and sensory properties in previous research conducted at Tecnolat,” said Cristian Mauricio Barreto Pinilla, first author of the article and a researcher at ITAL with a PhD in food science and technology. This was followed by in vitro tests to confirm that the bacteria were safe and probiotic. “We analyzed this information, focusing on the technological properties of the isolates, with a view to producing pilot batches of Prato cheese containing each of the three strains. We also studied the changes in physicochemical parameters, bacterial communities, volatile compounds [responsible for aroma and flavor], and fatty acids during cheese ripening,” Barreto explained. The study was supported by FAPESP and reported in an article published in the journal Current Microbiology. The findings show that all three Lactobacillus isolates are safe for consumption, have potential as novel probiotics, and have strong inhibitory effects against some of the pathogens that may be present in dairy produce. Furthermore, none of the three strains significantly affected the physicochemical composition of the cheese containing them in terms of fatty acid and protein profiles. “We also observed a drop in levels of undesirable volatile compounds during longer maturation periods for cheeses containing the cultures, but each strain produced a different profile in this regard,” Barreto said, explaining that this is important for diversification, quality, and shelf life of cheeses with a maturation period of more than 25 days. “We need to find out more about certain aspects, such as a possible reduction in deteriorating and pathogenic microorganisms in cheese due to the action of our Lactobacillus strains.” Industrial and Market Potential The conclusions met the aim of the study, he said, which was to analyze Lactobacillus strains derived from Brazilian biodiversity with probiotic and technological properties suitable for use in cheese production so as to enhance the sensory diversity and microbiological quality of the products, while also benefiting the health of consumers. “This kind of microorganism is easy to produce industrially, favoring studies designed to optimize the use of low-cost nutrients such as serum. Research along these lines is relevant to the needs of the cheese industry, which has significant growth potential but is cramped by the limited market for probiotics, dominated by large multinational companies,” he said. Although microbial culture technology is well-established in the multinationals that supply cultures to Brazil, it is necessary to develop local companies with the capacity to produce cultures for traditional Brazilian products that are competitive and meet the needs of small and medium producers for high-quality products with enhanced sensory properties, he added. The studies conducted by the team to date have had good results, but more research is needed for the industry to be able to use these strains and to ensure their classification as probiotics. “We need to conduct tests in an animal model, followed by clinical trials that enable us to comply with the regulatory requirements. These procedures take time and are costly,” Barreto said. Researchers have discovered that a particular protein can be used as a brain marker to indicate whether emotional memories can be changed or forgotten. This is a study in animals, but the researchers hope that the findings will eventually allow people suffering from Post-Traumatic Stress Disorder (PTSD) to return to leading a more balanced life. This work is presented at the ECNP Conference in Lisbon. Scientists know that long-term memories can broadly be divided into two types: fact-based memory, where we can recall such things as names, places, events, etc., and a sort of instinctive memory where we remember such things as emotions and skills. Scientists have come to believe that these emotional memories can be modified, so perhaps allowing the trauma underlying PTSD to be treated. In 2004 some ground-breaking work by scientists in New York[1] showed that if animals were treated with the beta-blocker propranolol, this allowed them to forget a learned trauma. However, the results have sometimes been difficult to reproduce, leading to doubts about whether the memories were modifiable at all. Now scientists at Cambridge University have shown that the presence of a particular protein – the “shank” protein, which acts as a scaffold for the receptors that determine the strength of connections between neurons – determines whether the memories can be modified in animals treated with propranolol. If this protein is degraded, then memories become modifiable.[2] However, if this protein is found to be present, then this shows that the memories were not degradable, so explains why propranolol does not always produce amnesia. Lead researcher, Dr. Amy Milton said: “We trained rats to associate a clicker with a mild electric footshock, to create a fear memory, similar to the way Pavlov conditioned dogs more than a hundred years ago. We then reminded the rats of this memory (‘reactivated the memory’) by introducing the clicker on its own, and immediately after this reminder, we gave an injection of the beta-blocker propranolol. However, we did not see the amnesia that had been previously reported in the literature following this intervention. We then used the presence of the shank protein to determine whether the memories had become unstable in the first place, and found that they had not. “This means that the shank protein can be used a biomarker for a malleable memory. We don’t yet know if it’s directly involved in the memory degradation, or if it’s a by-product of a deeper reaction. What it does do is give us a way in, a key to one of the first doors in understanding the biochemistry of memory. “These are really complex mechanisms, and we need to bear in mind that this is animal work; the brains of humans are similar, but much more complex. We don’t see this leading to the sort of situation shown in the movies, like for example “Eternal Sunshine of the Spotless Mind”, where the protagonists can choose which memories to erase. But we hope that over time we will be able to identify the factors that make memories modifiable in animals and translate these to human patients. Ultimately, we hope to lessen the unconscious impact of traumatic emotional memories, the sort of trauma which can ruin the lives of people with PTSD. In ancient Greek legend they spoke of a drug, Nepenthe, which made them forget painful memories. We hope that this is a step on the path to treatment.” Commenting, Dr. Livia de Picker, University of Antwerp, said: “This is interesting work. Unpicking what makes a memory is extremely difficult, and this work takes us a step nearer to understanding how memories are retained and changed. There is a long way to go in this process, and of course, transferring these steps to humans will be difficult. The European College of Neuropsychopharmacology is Europe’s main organization working in applied neuroscience. Mother mouse takes care of its offspring. Watching a mother mouse gather her pups into the family’s nest trains other female mice without pups to perform the same parenting task, a new study shows. Furthermore, these observations lead to the production of oxytocin in the brains of virgin female mice, biochemically shaping their maternal behaviors even before they have pups of their own. Led by researchers at NYU Grossman School of Medicine, the new set of experiments involved round-the-clock filming of female mice interacting with their newborns as well as with virgin mice. Simultaneous electrical readings were made in several brain regions known to produce oxytocin or thought to be responding to the hormone. The research team built on its earlier studies of the so-called pleasure hormone showing that the release of oxytocin is essential not only for the onset of nursing but also for the initiating of other maternal behaviors. Publishing in the journal Nature online today (August 11, 2021), researchers describe what they called a never-before-seen behavior in which new mouse mothers would without prompting shepherd virgin female mice into the family’s nest along with their pups. Within 24 hours, the virgins began mimicking the maternal behavior of gathering the mom’s pups into the nest even if the mother was not there. Almost as quickly, virgin mice would also start to perform the pup-retrieving task without any direct contact with an experienced mouse mother and after having only “viewed” the mother through a clear plastic window. Credit: NYU Langone The research team also measured brain electrical activity in virgin mice during shepherding and later when they became mothers on their own. They found that both the sight and sound of crying pups moving outside of their nest stimulated oxytocin production in a specific region of the brain, the hypothalamic paraventricular nucleus (PVN). By contrast, chemically blocking any of the visual, auditory, or oxytocin-producing PVN nerve pathways prevented virgin mice from learning to take care of pups. “Our study shows that in mice the best way to be a mom is to watch and learn from an experienced mom,” says study senior investigator Robert Froemke, PhD, a professor in the Skirball Institute of Biomolecular Medicine at NYU Langone Health. “Given the evidence, we propose that similar mechanisms operate in human mothers.” Froemke says the study findings in rodents add scientific evidence to the benefits observed from parenting classes in humans. He says the team next plans to examine if the same tutoring relationship exists among dad mice and virgin males. “This work redefines oxytocin’s role in brain function, broadening its impact to include formidable and complex social networking activities that force the brain to pay attention and adapt to its surroundings at the time, whether it’s reacting to the sound of a pup’s cries or feelings of happiness,” says Froemke, who also serves as a professor in the departments of Otolaryngology-Head and Neck Surgery, and Neuroscience and Physiology at NYU Langone. As part of the ongoing study, researchers analyzed nearly 5,000 hours (over six months) of video footage of several dozen mother mice interacting with their pups and with virgin mice.
