UH Researchers Receive $1.2 Million Grant to Peer into Ribosomes Two University of Houston researchers are developing a type of spectroscopy to help understand how ribosomes make proteins deep within cells, the discovery of which could potentially guide drug design to treat cancers and viral infections. Spectroscopy measures the interaction between light and matter to determine characteristics and volume of cellular matter. Ribosomes in Protein Assembly In cellular biology, ribosomes are work horses, veritable factories inside cells, whose job is to make proteins. The instructions that tell the ribosome how to work come from messenger RNA, which contains codes on making proteins, actually called codons. One mistake in defining an upstream codon will be propagated to the rest of the messenger like the domino effect, which spells disaster to the cell. During protein assembly, the ribosome must be precise in moving from one codon to the next, a process known as translocation. On the other hand, many viruses contain genomic sequences that are designed to slip on certain codons to re-define the protein composition after that codon, in a process called frameshifting. Yuhong Wang, professor of biology and biochemistry and Shoujun Xu, professor of chemistry, are developing a new imaging technique with super-resolution to peer into ribosomes. Credit: UH “We are developing a multiplexed super-resolution force spectroscopy to investigate high-fidelity and frameshifting translocations,” said Yuhong Wang, professor of biology and biochemistry. Wang and Shoujun Xu, professor of chemistry, received a $1.2 million grant from the National Institute of General Medical Sciences to support their research. “We will measure the power strokes from elongation factors (EF-G) and their mutants, which are the enzymes to interact with ribosome during translocation, on normal and viral mRNA sequences and in the presence of antibiotics,” said Xu. “Our research will provide new methodology that can be applied to other biological systems.” Potential Applications in Drug Design Scientifically, the team is building a new model of ribosome translocation with sub-codon steps and providing potential drug targets for related diseases. “For example, by tuning down and up the EF-G’s activity in cancer cells and low-functioning neuron cells, the diseases can be treated, anti-viral drugs can be designed that only target the specific viral frameshifting motifs,” said Wang. A new method to profile gene activity in the living brain offers breakthroughs in epilepsy treatment and other neurological research, combining molecular analysis with brain activity recordings. Researchers have developed a pioneering technique to profile gene activity in the living human brain. Researchers at FutureNeuro, the SFI Research Centre for Translational Brain Science, and RCSI University of Medicine and Health Sciences, in collaboration with international partners, have developed a revolutionary technique to profile gene activity in the living human brain. This innovative approach, published in JCI Insight, opens new avenues for understanding and treating neurological conditions like epilepsy. Studying gene activity in the brain without requiring invasive tissue samples from surgery or post-mortem donation has been a long-standing challenge in neuroscience. By analyzing molecular traces – specifically RNA and DNA – collected from electrodes implanted in the brains of patients with epilepsy and linking these with electrical recordings from the brain, the researchers were able to take a ‘snapshot’ of gene activity in the living brain. Credit: RCSI These electrodes, clinically used to pinpoint seizure activity in patients enabling surgical interventions, provide a unique opportunity to link brain activity to the genes being switched on or off in specific regions. The study demonstrates how integrating molecular data with electrical recordings of seizures can enhance our understanding of the brain’s seizure networks, potentially improving the precision of epilepsy surgeries. Broader research Professor David Henshall, Director of FutureNeuro and Professor of Molecular Physiology and Neuroscience at RCSI, said: “This study represents a significant advancement in epilepsy research, providing a method to detect active genes within the living brain of individuals with epilepsy. This technology has the potential to complement traditional brain imaging and EEG tests that measure electrical activity in the brain, offering valuable insights to guide surgical decision-making in the treatment of those with epilepsy.” Epilepsy affects approximately 40,000 people in Ireland, with one in three people unable to control seizures through medication. For these individuals, surgical intervention is often the best option, but its success hinges on accurately mapping the regions responsible for seizure activity. Beyond epilepsy, the study lays the groundwork for broader applications, including research into Alzheimer’s, Parkinson’s, and schizophrenia, where understanding molecular processes in the living brain is vital. A step forward The research, led by Professor Henshall and Professor Vijay Tiwari, Professor of Genome Biology at the University of Southern Denmark, also involved a global network of collaborators, including experts from Beaumont Hospital, Blackrock Clinic, Queen’s University Belfast, the University of Southern Denmark, and the Danish Institute for Advanced Study. It underscores the value of international collaboration and marks a step forward in understanding how our brains function at the molecular level, offering hope for improved diagnosis and care for those impacted by neurological conditions. Neonicotinoid insecticides disrupt the sleep patterns of bumblebees and fruit flies, possibly contributing to the decline of insect pollinators. Researchers urge UK to keep EU ban on pesticide which has detrimental effect on pollinators. Just like us, many insects need a decent night’s sleep to function properly, but this might not be possible if they have been exposed to neonicotinoid insecticides, the most common form of insecticide used worldwide, suggests research by academics at the University of Bristol. Two studies by scientists at Bristol’s Schools of Physiology, Pharmacology, and Neuroscience and Biological Sciences have shown these insecticides affect the amount of sleep taken by both bumblebees and fruit flies, which may help us understand why insect pollinators are vanishing from the wild. Dr. Kiah Tasman, Teaching Associate in the School of Physiology, Pharmacology and Neuroscience and lead author of the studies, said: “The neonicotinoids we tested had a big effect on the amount of sleep taken by both flies and bees. If an insect was exposed to a similar amount as it might experience on a farm where the pesticide had been applied, it slept less, and its daily behavioral rhythms were knocked out of synch with the normal 24-hour cycle of day and night.” Neonicotinoids Impair Memory and Internal Clocks The fruit fly study published today (January 21, 2021) in Scientific Reports, allowed the researchers to study the impact of the pesticides on the insect brain. As well as finding that typical agricultural concentrations of neonicotinoids ruined the flies’ ability to remember, the researchers also saw changes in the clock in the fly brain which controls its 24-hour cycle of day and night. Dr. James Hodge, Associate Professor in Neuroscience in the School of Physiology, Pharmacology and Neuroscience and senior author for the study, added: “Being able to tell time is important for knowing when to be awake and forage, and it looked like these drugged insects were unable to sleep. We know quality sleep is important for insects, just as it is for humans, for their health and forming lasting memories.” Implications for Pollinator Behavior and Survival Dr. Sean Rands, Senior Lecturer in the School of Biological Sciences and co-author, explained: “Bees and flies have similar structures in their brains, and this suggests one reason why these drugs are so bad for bees is they stop the bees from sleeping properly and then being able to learn where food is in their environment. “Neonicotinoids are currently banned in the EU, and we hope that this continues in the UK as we leave EU legislation.” References: “Neonicotinoids disrupt memory, circadian behaviour and sleep” by Kiah Tasman, Sergio Hidalgo, Bangfu Zhu, Sean A.
