Researchers have found that maternal obesity causes long-term changes in the brain via the microRNA miR-505-5p, leading offspring to prefer high-fat diets and increasing their risk of obesity. This effect can be mitigated by maternal exercise during pregnancy, according to a new study in PLOS Biology. A study reveals that maternal obesity in mice increases microRNA levels in the hypothalamus in offspring, leading to overeating. Maternal obesity impacts the eating behaviors of offspring via long-term overexpression of the microRNA miR-505-5p. This is according to a study published today (June 4th) in the open-access journal PLOS Biology by Laura Dearden and Susan Ozanne from the MRC Metabolic Diseases Unit, Institute of Metabolic Science, University of Cambridge, UK, and colleagues. Link Between Maternal Obesity and Offspring Health Risks Previous studies in both humans and animal models have shown that the offspring of obese mothers have a higher risk of obesity and type 2 diabetes. While this relationship is likely the result of a complex relationship between genetics and environment, emerging evidence has implicated that maternal obesity can disrupt the hypothalamus—the region of the brain responsible for nutrition sensing and energy homeostasis. In animal models, offspring exposed to overnutrition during key periods of development eat more, but little is known about the molecular mechanisms that lead to these changes in eating behavior. Study Findings on MicroRNA and Eating Behaviors In this study, researchers found that mice born from obese mothers had higher levels of the microRNA miR-505-5p in their hypothalamus—from as early as the fetal stage into adulthood. The researchers found that the mice ate more and showed a preference for high-fat foods. Interestingly, the effect of maternal obesity on miR-505-5p and eating behaviors was mitigated if the mothers exercised during pregnancy. Molecular Mechanisms and Preventative Measures Cell culture experiments showed that miR-505-5p expression could be induced by exposing hypothalamic neurons to long-chain fatty acids and insulin, which are both high in pregnancies complicated by obesity. The researchers identified miR-505-5p as a novel regulator of pathways involved in fatty acid uptake and metabolism, therefore high levels of the miRNA make the offspring brain unable to sense when eating high-fat foods. Several of the genes that miR-505-5p regulates have been associated with high body mass index in human genetic studies. The study is one of the first to demonstrate the molecular mechanism linking nutritional exposure in utero to eating behavior. Conclusion and Implications The authors add, “Our results show that obesity during pregnancy causes changes to the baby’s brain that makes them eat more high-fat food in adulthood and more likely to develop obesity. “Importantly we showed that moderate exercise, without weight loss, during pregnancies complicated by obesity prevented the changes to the baby’s brain. Since its formulation in the 1960s, however, the ‘island rule’ has been severely debated by scientists. In a new publication in Nature Ecology and Evolution on April 15, 2021, researchers solved this debate by analyzing thousands of vertebrate species. They show that the island rule effects are widespread in mammals, birds, and reptiles, but less evident in amphibians. Dwarf hippos and elephants in the Mediterranean islands are examples of large species that exhibited dwarfism. On the other hand, small mainland species may have evolved into giants after colonizing islands, giving rise to such oddities as the St Kilda field mouse (twice the size of its mainland ancestor), the infamous dodo of Mauritius (a giant pigeon), and the Komodo dragon. In 1973, Leigh van Valen was the first that formulated the theory, based on the study by mammologist J. Bristol Foster in 1964, that animal species follow an evolutionary pattern when it comes to their body sizes. Species on islands have the tendency to become either giants or dwarfs in comparison to mainland relatives. “Species are limited to the environment on an island. The level of threat from predatory animals is much lower or non-existent,” says Ana Benítez-Lopez, who carried out the research at Radboud University, now a researcher at Doñana Biological Station (EBD-CSIC, Spain). “But also limited resources are available.” However, until now, many studies showed conflicting results which led to severe debate about this theory: is it really a pattern, or just an evolutionary coincidence? Island rule confirmed The team of scientists at Radboud University, Doñana Biological Station, National Museum of Natural Sciences and Imperial College London has revisited the island rule, aiming to solve this debate by performing a meta-analysis of over a thousand vertebrate species. They show that island rule effects are widespread in mammals, birds and reptiles, but less evident in amphibians, which mostly tend towards gigantism. The study also indicates that the magnitude of insular dwarfism and gigantism is more pronounced in smaller, more remote islands for mammals and reptiles. Size is context-dependent They also found an effect of climate and seasonality on the island rule. Small mammal and bird species grew larger and large species stayed the same size to conserve heat in colder, harsher insular environments. Furthermore, when seasons are present, the availability of resources becomes less predictable for reptiles, leading smaller reptile species to become larger. Credit: Helena Pinheiro Scientists have developed new biosensors that enable real-time tracking of opioid signaling in the brain, offering profound insights into the effects of opioids and enhancing the development of targeted therapeutics. New findings from a researcher at the Max Planck Florida Institute for Neuroscience could revolutionize our understanding of how opioids impact the brain. Despite the extensive discussion surrounding the ongoing opioid crisis, our current knowledge of how opioids operate in the brain remains quite limited. This is primarily due to challenges in observing and measuring opioid effects in the brain in real time. However, a recent technological breakthrough, led by Dr. Lin Tian and her research team and collaborators, recently published in Nature Neuroscience, has overcome these limitations and is set to transform how scientists study opioid signaling in the brain. What do we know about opioid signaling? Pharmaceutical opioids, such as morphine and oxycodone, and illicit opioid drugs like heroin, affect the brain and body by binding to opioid receptors on the surface of cells in the nervous system. These receptors normally respond to naturally produced chemicals that are released in your brain, called endogenous opioids, including endorphins, enkephalins, and dynorphins. Released in response to pleasurable activities such as laughter, sex, and exercise and aversive activities like injury and trauma, these chemicals bind opioid receptors and reduce the ability of neurons to receive and transmit signals. These cellular effects ultimately lead to the cognitive and behavioral effects associated with opioids, including positive feelings, pain relief, and addiction. Challenges in understanding opioid signaling Numerous questions remain about how these behavioral effects are caused by opioids and whether it is possible to harness specific opioid properties such as pain relief without undesirable effects, such as addiction. The opioid scientific literature is extensive and has confirmed that targeting the opioid system is of clinical interest – not only for pain management but also, more recently, for the treatment of mental health disorders such as anxiety and depression. Development of therapeutics that can target these health challenges while preventing the tragedy of the current opioid epidemic requires further understanding of the diverse effects of opioids in the brain. The diversity of opioid effects on the brain is driven by more than 20 different opioid chemicals produced in the brain and more than 500 different synthetic opioids. Most of these different opioids interact with all three types of opioid receptors with different strengths. Their varied effects depend on the concentration of opioids, the specific receptors present and the brain circuits involved. “Efforts are underway to harness various therapeutic properties of opioids by targeting specific receptor actions and brain circuits to develop more effective and safer therapeutics. However, these efforts have been hampered by our inability to measure diverse opioid signaling in real-time in the brain effectively,” said Dr. Tian. New technology opens the door to understanding opioids in brain Through a massive effort developing and testing over 1,000 variants, Dr. Tian’s team has optimized highly sensitive biosensors based on the three opioid receptors. These biosensors, originally developed while Tian was at the University of California, Davis, emit fluorescence upon opioid binding to the sensor and turn off when the opioid is no longer there. The biosensors, therefore, serve as a proxy for opioid binding to specific opioid receptors. Introducing these sensors into the brain of an animal provides a way to visualize opioid signaling across the brain in real time. “The power of this new technology is that we now have the tools to understand the natural opioid system in the brain, including how to distinguish between different opioid effects. We can track endogenous opioid release in real-time, triggered by both reward and aversion, and see the differences in opioid signaling in different brain circuits.” Dr. Tian’s team has already been sharing these new tools widely to accelerate the impact this new technology will have on the understanding of opioids.
