UC San Diego research identifies critical metabolic pathways in early autism development, offering new avenues for early detection and pharmacological treatments. Findings suggest new possibilities for early autism detection. Researchers at the University of California San Diego School of Medicine have shed new light on the changes in metabolism that occur between birth and the presentation of autism spectrum disorder (ASD) later in childhood. The researchers discovered that a small number of biochemical pathways are responsible for the majority of these changes, which could help inform new early detection and prevention strategies for autism. “At birth, the physical appearance and behavior of a child who will develop autism over the next few years are indistinguishable from that of a neurotypical child. Indeed, in most cases the fate of the child with regard to autism is not set at birth,” said Robert Naviaux, M.D., Ph.D., professor in the Departments of Medicine, Pediatrics and Pathology at UC San Diego School of Medicine. “We’re starting to learn about the governing dynamics that regulate the transition from risk to the actual appearance of the first symptoms of ASD. Early diagnosis opens the possibility of early intervention and optimal outcomes.” Autism: A Complex Interplay of Factors ASD is a developmental disorder characterized by difficulties in socializing and communication, as well as repetitive and/or restrictive behaviors. For the majority of people with ASD, the condition is a significant disability, with only 10-20 percent of children diagnosed before 5 years of age able to live independently as adults. While autism is known to have strong genetic risk factors, there are also environmental risk factors that play a role in the development and severity of ASD. Naviaux and other researchers are discovering that the development of autism is governed by the real-time interaction of these varied factors. By studying the developmental biology of metabolism and how it differs in autism, new insights are emerging in ASD and other complex developmental disorders. “Behavior and metabolism are linked – you cannot separate them,” added Naviaux. To learn more about the early metabolic changes that occur in children with autism, researchers studied two cohorts of children. One cohort consisted of newborn children, in whom autism can’t be detected. The second cohort consisted of 5-year-old children, some of whom had been diagnosed with autism. When comparing the metabolic profiles of children in the cohort who were eventually diagnosed with autism to those who developed neurotypically, they found striking differences. Of the 50 different biochemical pathways the researchers investigated, just 14 were responsible for 80 percent of the metabolic impact of autism. New Insights Into Autism’s Biochemical Processes The pathways that were most changed are related to the cell danger response, a natural and universal cellular reaction to injury or metabolic stress. The body has biochemical safeguards in place that can shut down the cell danger response once the threat has passed, and Naviaux hypothesizes that autism occurs when these safeguards fail to develop normally. The result is heightened sensitivity to environmental stimuli, and this effect contributes to sensory sensitivities and other symptoms associated with autism. “Metabolism is the language that the brain, gut, and immune system use to communicate, and autism occurs when the communication between these systems is changed,” added Naviaux. The cell danger response is primarily regulated by adenosine triphosphate (ATP) the body’s chemical energy currency. While these ATP-signaling pathways do not develop normally in autism, they may be partially restorable with existing pharmaceutical drugs. In 2017, Naviaux and his team completed early clinical testing for suramin, the only drug approved in humans that can target ATP signaling and which is normally used to treat African sleeping sickness. Now, the researchers hope that by revealing the specific ATP-related pathways that are altered in autism, their work will help scientists develop more drugs that target these pathways to manage the symptoms of ASD. “Suramin is just one drug that targets the cell danger response,” he said. “Now that we’re closely interrogating how metabolism changes in ASD, we could be at the beginning of a drug renaissance that will create new options for treatment that never existed before.” Reference: “Metabolic network analysis of pre-ASD newborns and 5-year-old children with autism spectrum disorder” by Sai Sachin Lingampelly, Jane C. Pink sea fan. Credit: Jamie Stevens An iconic coral species found in UK waters could expand its range due to climate change, according to a new study. The pink sea fan is a soft coral that lives in shallow waters from the western Mediterranean (southern range) to northwest Ireland and the southwest of England and Wales (northern range). The species is classified as “vulnerable” worldwide and it is listed as a species of principal importance in England and Wales under the NERC Act 2006. The new research, by the University of Exeter, found that the species is likely to spread northwards – including around the British coast – as global temperatures rise. The findings could be used to identify priority areas to protect pink sea fan populations. “We built models to predict the current and future (2081-2100) habitat of pink sea fans across an area covering the Bay of Biscay, the British Isles, and southern Norway,” said Dr. Tom Jenkins, from the University of Exeter. “The model predictions revealed current areas of suitable habitat beyond the current northern range limits of the pink sea fan, in areas where colonies have not yet been observed. Potential Barriers to Northward Spread “It’s not clear why pink sea fans have not yet colonized these areas. Possible barriers include insufficient dispersal of their larvae and high competition between species for space and resources. “Our future predictions, using a high-emissions global warming scenario called RCP 8.5, revealed an increase in suitable habitat for pink sea fans to the north of its current range – so the species could spread northwards by 2100. “We also found that existing habitat across south-west Britain, the Channel Islands, and north-west France is predicted to remain suitable for this species over the next 60-80 years.” Pink sea fans. For this species, future predictions revealed an overall decrease in suitable habitat in the southern portion of the study area and an accompanying increase in the northern portion of the species’ range. Ecological Importance of Pink Sea Fans Pink sea fans, like many octocoral species, are ecologically important because they add complexity to reef systems and support marine biodiversity, especially when they form dense ‘forests’. They can also be used as a broader indicator of ecosystem health because fragmented or diseased colonies may be an indicator of degraded environments. Dr. Jamie Stevens, also from the University of Exeter, said: “This research highlights the complex effects of climate change on marine ecosystems, in which the ranges of some species respond to warming by shifting pole-wards. “In a rapidly changing mosaic of habitats, some species – typically those favoring warmer conditions – may come out as short-term ‘winners’. “How long these species can continue to expand and benefit in the face of accelerated warming remains to be seen.” The paper, published in the journal PeerJ, is entitled: “Predicting habitat suitability and range shifts under projected climate change for two octocorals in the north-east Atlantic.” Reference: “Predicting habitat suitability and range shifts under projected climate change for two octocorals in the north-east Atlantic” by Tom L. Credit: Brock and Sherri Fenton Researchers have developed predictive models to identify bats that may harbor betacoronaviruses. Their study uses machine learning to dynamically update wildlife virus sampling, emphasizing bat conservation to prevent outbreaks. Daniel Becker, an assistant professor of biology in the University of Oklahoma’s Dodge Family College of Arts and Sciences, has been leading a proactive modeling study over the last year and a half to identify bat species that are likely to carry betacoronaviruses, including but not limited to SARS-like viruses. The study “Optimizing predictive models to prioritize viral discovery in zoonotic reservoirs,” which was published by Lancet Microbe, was guided by Becker; Greg Albery, a postdoctoral fellow at Georgetown University’s Bansal Lab; and Colin J. Carlson, an assistant research professor at Georgetown’s Center for Global Health Science and Security. It also included collaborators from the University of Idaho, Louisiana State University, University of California Berkeley, Colorado State University, Pacific Lutheran University, Icahn School of Medicine at Mount Sinai, University of Glasgow, Université de Montréal, University of Toronto, Ghent University, University College Dublin, Cary Institute of Ecosystem Studies, and the American Museum of Natural History. The Verena Consortium’s Role in Viral Forecasting Becker and colleagues’ study is part of the broader efforts of an international research team called the Verena Consortium (viralemergence.org), which works to predict which viruses could infect humans, which animals host them, and where they could emerge. Albery and Carlson were co-founders of the consortium in 2020, with Becker as a founding member. Despite global investments in disease surveillance, it remains difficult to identify and monitor wildlife reservoirs of viruses that could someday infect humans. Statistical models are increasingly being used to prioritize which wildlife species to sample in the field, but the predictions being generated from any one model can be highly uncertain. Scientists also rarely track the success or failure of their predictions after they make them, making it hard to learn and make better models in the future. Together, these limitations mean that there is high uncertainty in which models may be best suited to the task. Methodology of Bat Host Prediction In this study, researchers used bat hosts of betacoronaviruses, a large group of viruses that includes those responsible for SARS and COVID-19, as a case study for how to dynamically use data to compare and validate these predictive models of likely reservoir hosts. The study is the first to prove that machine learning models can optimize wildlife sampling for undiscovered viruses and illustrates how these models are best implemented through a dynamic process of prediction, data collection, validation and updating. In the first quarter of 2020, researchers trained eight different statistical models that predicted which kinds of animals could host betacoronaviruses. Over more than a year, the team then tracked discovery of 40 new bat hosts of betacoronaviruses to validate initial predictions and dynamically update their models. The researchers found that models harnessing data on bat ecology and evolution performed extremely well at predicting new hosts of betacoronaviruses. In contrast, cutting-edge models from network science that used high-level mathematics – but less biological data – performed roughly as well or worse than expected at random. Findings and Implications for Bat Conservation Importantly, their revised models predicted over 400 bat species globally that could be undetected hosts of betacoronaviruses, including not only in southeast Asia but also in sub-Saharan Africa and the Western Hemisphere. Although 21 species of horseshoe bats (in the Rhinolophus genus) are known to be hosts of SARS-like viruses, researchers found at least two-fourths of plausible betacoronavirus reservoirs in this bat genus might still be undetected. “One of the most important things our study gives us is a data-driven shortlist of which bat species should be studied further,” said Becker, who adds that his team is now working with field biologists and museums to put their predictions to use. “After identifying these likely hosts, the next step is then to invest in monitoring to understand where and when betacoronaviruses are likely to spill over.” Becker added that although the origins of SARS-CoV-2 remain uncertain, the spillover of other viruses from bats has been triggered by forms of habitat disturbance, such as agriculture or urbanization. “Bats conservation is therefore an important part of public health, and our study shows that learning more about the ecology of these animals can help us better predict future spillover events,” he said.
