Tiny ocean diatoms are highly efficient at capturing carbon dioxide (CO2) from the environment, accounting for up to 20 percent of the Earth’s CO2 fixation. Researchers at the University of Basel in Switzerland have now discovered a protein shell within these algae that is essential for their ability to fix CO2 so effectively. This significant finding could inspire new bioengineering strategies to help reduce atmospheric CO2 levels. Diatom Discovery and Carbon Capture Diatoms, though invisible to the naked eye, are among the most productive algae in the ocean and play a crucial role in the global carbon cycle. Through photosynthesis, they absorb large amounts of CO2 from the environment and convert it into nutrients that sustain much of ocean life. Despite their significance, how diatoms perform this process so efficiently has remained a mystery. Now, researchers led by Prof. Ben Engel at the University of Basel’s Biozentrum, along with teams from the University of York, UK, and Kwansei-Gakuin University in Japan, have uncovered a protein shell crucial to diatoms’ CO2 fixation. Using advanced imaging techniques like cryo-electron tomography (cryo-ET), they mapped the molecular structure of the PyShell protein sheath and revealed its function. These findings were recently published in two papers in the journal Cell. Cryo-electron tomography reveals the molecular architecture of the diatom pyrenoid, showing how Rubisco is surrounded by the PyShell, increasing the efficiency of carbon dioxide fixation. Credit: Gröger et al. / Manon Demulder, Biozentrum, Universität Basel PyShell: A Key to Efficient Photosynthesis In plants and algae, photosynthesis takes place in chloroplasts. Inside these chloroplasts, energy from sunlight is harvested by thylakoid membranes and then used to help the enzyme Rubisco fix CO2. However, algae have an advantage: they pack all their Rubisco into small compartments called pyrenoids, where CO2 can be captured more efficiently. “We have now discovered that diatom pyrenoids are encased in a lattice-like protein shell,” says Dr. Manon Demulder, author on both studies. “The PyShell not only gives the pyrenoid its shape, but it helps create a high CO2 concentration in this compartment. This enables Rubisco to efficiently fix CO2 from the ocean and convert it into nutrients.” When the researchers removed the PyShell from the algae, their ability to fix CO2 was significantly impaired. Photosynthesis and cell growth were reduced. “This showed us how important the PyShell is for efficient carbon capture – a process that is crucial for ocean life and the global climate,” says Manon Demulder. Potential Climate Solutions Through Bioengineering The discovery of the PyShell could also open promising avenues for biotechnological research aimed at combatting climate change – one of the most pressing challenges of our time. “First of all, we humans must reduce our CO2 emissions to slow the pace of climate change. This requires immediate action,” says Ben Engel. “The CO2 that we emit now will remain in our atmosphere for thousands of years. We hope that discoveries such as the PyShell can help inspire new biotechnology applications that improve photosynthesis and capture more CO2 from the atmosphere. Germ-free larvae have altered neural connections due to a reduction in pruning by immune cells. Microorganisms are essential for normal social development in zebrafish via their influence on pruning of neural connections in the developing brain, according to a study published in the open access journal PLOS Biology by Joseph Bruckner at the University of Oregon, US, and colleagues. Zebrafish larvae have transparent skin, offering researchers a rare window into neural development. In a series of experiments, the researchers investigated the neuronal and social development of zebrafish larvae reared with or without the presence of their normal microbiota for the first seven days of development. They found that larval social behavior at day 14 was inhibited by the early absence of microbes, despite the larvae’s normal microbiota being restored a week earlier. Compared to siblings with a normal microbiota, the brains of these germ-free larvae had fewer immune cells called microglia in their forebrains, and denser, more complex neural branching patterns. Single-cell RNA sequencing showed that their microglia also had lower levels of complement c1q, a gene involved in pruning neural connections called synapses. A study published in PLOS Biology indicates that microorganisms significantly influence the social development of zebrafish by affecting neural pruning in the developing brain. Restoring Social and Neural Development Genetically reducing microglia without affecting the microbiota produced similar results, increasing neural density and branching relative to controls. Normal neural and social development was restored in germ-free larvae by adding any of several different bacterial groups native to the zebrafish intestine, indicating that microglial pruning activity is sensitive to a feature common across many types of bacteria. The study is the first to demonstrate that microglia are required for pruning of neural connections in zebrafish larvae, and that an intact microbiota is essential for normal pruning and fish social behavior. The results also show that there is a critical developmental window during the first week of larval development, during which the microbiota stimulates microglial localization to the forebrain where they prune neural connections, the authors say. Eisen adds, “Social interactions are critical for animals and humans. By the time whales return to summer feeding grounds, their calves are already weaned. Only two blue whale births have ever been recorded in human history, both occurring decades ago. This remains an extraordinary mystery, given that there were once hundreds of thousands of blue whales before whaling began. Even today, their population is estimated to be between 10,000 and 25,000, and they give birth every two to three years. Not only are births remarkably stealthy, but calves are also rarely sighted—far less often than would be expected based on their pregnancy rates. Calves closely follow their mothers and are typically seen in mother-calf pairs, yet why so few are detected remains unclear. A new University of Washington study, published on Feb. 20 in Endangered Species Research, offers a possible explanation. The research suggests when and where these unseen births are occurring and where blue whale calves spend their earliest months. The findings provide some hope for the health of the population. Credit: Leigh Torres/Geospatial Ecology of Marine Megafauna Laboratory (GEMM) Lab, Marine Mammal Institute, Oregon State University Trevor Branch, a UW professor of aquatic and fishery sciences who studies blue whales, set out to unravel this mystery by looking at a range of hypotheses. He proposes that one in particular is the best explanation: It’s mostly because researchers prefer summertime research on feeding congregations of blue whales, but calves are born in fall and winter, and are weaned before they return to feeding areas. Seasonal Migration and Birth Timing In summer, blue whales migrate to feed in colder regions where krill is plentiful: for example, off California. In winter, when ready to give birth, they return to warmer regions like the Gulf of California and the eastern tropical Pacific. Around seven months after being born, and already at a whopping 52 feet (16 meters) long, the calves are weaned and stop associating with their mothers. But across various blue whale populations, high pregnancy rates of 33-50% annually seem to contradict the average 3.1% rate of sightings of blue whales involving mother-calf pairs. When compared with other hypotheses to explain the mystery of why so few calves are observed, such as low fetal survival, low calf survival, low birth rates, or calf separation from mother, Branch discovered that the timing hypothesis best explained observed patterns. This would explain why researchers – most of whom conduct blue whale field studies in summer months – seldom sight mothers with calves. Supporting Evidence and Future Research Branch compiled data from long-term field studies and combined this with biological information from historical whaling records to come up with this hypothesis, finding higher proportions of calves in winter regions, and lower proportions in summer regions. He is now coordinating a large collaboration to test the idea with field data by month in each region, combined with estimates of the size of calves by month. One concern about low calf sighting rates was that this might be a warning signal of low birth rates or low survival of calves. Instead, the new hypothesis offers up some hope that higher number of calves could be sighted from field studies concentrated in regions that blue whales travel to in winter and spring. “This new idea provides an alternative explanation for why some blue whale populations appear to produce very few calves: It’s not a failure of calf production, it’s because fieldwork in those populations is understandably concentrated in easily accessible summer feeding areas,” Branch said.
