Credit: Utpal Pal / University of Maryland Ticks have the ability to detect a cytokine from infected mammalian blood, triggering a defense mechanism against bacteria that cause Lyme disease. A team of researchers from the University of Maryland has uncovered the first inter-species signaling pathway between a host and an arthropod parasite. The pathway involves molecules in the host’s blood triggering the immunity and development of the parasite. The study demonstrates that when ticks feed on blood from mice infected with the Lyme disease-causing bacteria Borrelia burgdorferi, a protein from the mouse immune system activates receptors on tick cells, signaling the tick’s organs to develop more quickly and producing an immune response before the bacteria can infect the tick. The study, which was recently published in the journal Science, identifies a potential target for anti-tick vaccines or therapeutics to prevent the spread of infections like Lyme disease. The findings also provide important new insights into the evolution of biomolecular interdependencies between species, and highlight, for the first time, both the integration of immunity and animal development and the adaptability of an ancient cell signaling system or pathway that all plant and animal cells use for sensing and responding to their environment. “This adaptive flexibility of a conserved cell signaling pathway was surprising,” said Utpal Pal, senior author of the study and a professor in the Virginia-Maryland College of Veterinary Medicine at College Park. “It is remarkable that this pathway that is present in everything from sponges to humans is so flexible it can adapt to accept a ligand [a binding molecule] from another distant species. Credit: Utpal Pal / University of Maryland The finding suggests that other cell signaling pathways may have been adapted for novel uses in other organisms and points to a new area in immunology and molecular biology ripe for future exploration. A Novel Role for the JAK/STAT Pathway in Ticks Pal and his colleagues made their discovery while investigating tick immunity, which is a poorly understood area of tick biology. In their initial study, seeking to understand how tick immune systems recognize the Borrelia bacteria, the researchers fed ticks a blood meal from either a Borrelia-infected mouse or an uninfected mouse. Comparing the two groups, they found that the infected blood meal activated a protein in ticks that normally produces energy inside cells. The protein is associated with a simple signaling pathway called JAK/STAT, which is present in all multicellular organisms. As in all cellular signaling pathways, a specific molecule senses something in the environment and then binds to a receptor on the outside of a cell wall. This sets off a cascade of reactions inside the cell that turns a specific gene on or off and produces a response to whatever outside stimuli was sensed. Assuming that JAK/STAT was triggered by the Borrelia in the infected mouse blood, the researchers isolated the bacteria and injected it directly into ticks to see what molecules were binding with the JAK/STAT receptor. Surprisingly, the bacteria did not activate JAK/STAT. To find out what did, the researchers removed the Borrelia bacteria from the blood of infected mice and fed the “clean” blood to ticks. The JAK/STAT pathway kicked into action. The researchers discovered that a protein in tick digestive systems was serving as the JAK/STAT receptor, and that it had evolved to bind with the cytokine protein interferon, which is produced by the immune systems of mammals infected with a bacteria like Borrelia. Credit: University of Maryland Implications for Tick-Borne Disease Prevention The researchers also found that the JAK/STAT receptor and pathway are important for normal tick development, even if the pathway is not activated by an infected blood meal. When Pal and his colleagues knocked down the expressed gene that produces the receptor for JAK/STAT, the ticks grew deformed legs, mouthparts, and digestive systems, and were unable to feed and complete the developmental cycle to grow further. These results suggest that in ticks, the JAK/STAT signaling pathway, and the protein receptor have evolved to integrate immunity with development. Bacteria will compete with ticks for nutrients in the blood of an infected host, so when a tick gets the signal that a blood meal is infected, growing rapidly is a way to use up those nutrients before the bacteria gets them. Laboratory experiments concur that ticks fed on Borrelia-infected mouse blood developed much quicker than those that fed on uninfected mouse blood. “Understanding that this pathway integrates immunity and development has important implications for potential strategies to prevent tick-borne disease transmission,” Pal said. “Because if you delete the pathway, ticks with malformed mouthparts cannot feed or transmit disease. New research has uncovered the mechanism by which amino acids activate TORC1, a key protein in cell growth and autophagy. The research reveals that cysteine activates TORC1 via the Pib2 protein and highlights the varied influences of all 20 amino acids on TORC1 through two pathways. This discovery offers new insights into cellular processes and potential treatments for diseases linked to TORC1 malfunctions. Researchers at Osaka University uncover the mechanism by which cysteine activates a crucial regulator of cell growth in yeast. Amino acids serve as life’s fundamental components. They are sourced from our diet, and our bodies utilize them to create proteins. These proteins are crucial for growth, development, and various other processes. Yet, prior to utilizing these building blocks, the body must first detect their presence. When amino acids are available, a master regulator protein called TORC1 is switched on, causing proteins to be manufactured and cells to grow. If no amino acids are available, TORC1 is switched off, and cells start to recycle themselves in a process known as autophagy. Until now, it was unclear exactly how amino acids triggered the TORC1 switch in yeast. Discovery in Amino Acid Sensing Now, in a study published in Cell Reports, researchers from Osaka University have revealed how TORC1 is activated: detection of the amino acid cysteine. “We investigated the relationships between amino acids and TORC1 activation in the yeast Saccharomyces cerevisiae,” says the study’s lead author Qingzhong Zeng. “We found that cysteine is sensed by a protein called Pib2 and that the two bind together and activates TORC1. This stimulates the synthesis of proteins and lipids, promoting cell proliferation.” Pib2 senses Cysteine to activate TORC1. Cell Reports The Role of Amino Acids in TORC1 Activation What’s more, cysteine is not the only amino acid that triggers TORC1. All 20 amino acids were found to differently affect TORC1 using two ‘pathways’: Pib2 and Gtr. A pathway can be thought of as a specific chain reaction that leads to certain outcomes in a cell. The team set out to elucidate how each amino acid uses these pathways to affect TORC1. “Some amino acids primarily use the Pib2 pathway, while others primarily use Gtr,” explains senior author Takeshi Noda. “We also identified amino acids that can use either pathway and some that need both. This work excites us because it deepens our understanding of how amino acids control cell growth and autophagy, and how each amino acid is detected.” In humans, faulty TORC1 function has been linked to diseases like cancer, diabetes, and dementia. A fuller understanding of how TORC1 is switched on and off, and how each amino acid is detected, could help researchers find new treatments for these diseases – an exciting prospect indeed. DOI: 10.1016/j.celrep.2023.113599 Using ancient DNA, researchers have traced the diverse origins of workers buried at Machu Picchu, revealing that they hailed from across the vast expanse of the Inca Empire. This study significantly enhances our understanding of the societal composition during the Inca Empire’s zenith. DNA analysis of Machu Picchu’s residents reveals they came from various regions of the Inca Empire, supporting historical and archaeological evidence of a diverse worker population. Who lived at Machu Picchu at its height? New research, recently published in the journal Science Advances, leverages ancient DNA to unveil the origins of workers buried over half a millennium ago within the lost Inca Empire for the first time. A team of researchers, including Jason Nesbitt, an associate professor of archaeology at Tulane University’s School of Liberal Arts, conducted genetic examinations on bodies buried at Machu Picchu. Their objective was to gather more information about the people who lived and worked there. Machu Picchu is a UNESCO World Heritage Site located in the Cusco region of Peru. It is one of the most well-known archaeological sites in the world and attracts hundreds of thousands of visitors every year. It was once part of a royal estate of the Inca Empire. Like other royal estates, Machu Picchu was home not only to royalty and other elite members of Inca society, but also to attendants and workers, many of whom lived in the estate year-round. These residents did not necessarily come from the local area, though it is only in this study that researchers have been able to confirm, with DNA evidence, the diversity of their backgrounds. “It’s telling us, not about elites and royalty, but lower status people,” Nesbitt said. “These were burials of the retainer population.” Genetic Analysis Reveals Widespread Origins This DNA analysis works in much the same way that modern genetic ancestry kits work. The researchers compared the DNA of 34 individuals buried at Machu Picchu to that of individuals from other places around the Inca Empire as well as some modern genomes from South America to see how closely related they might be. The results of the DNA analysis showed that the individuals had come from throughout the Inca Empire, some as far away as Amazonia. Few of them had shared DNA with each other, showing that they had been brought to Machu Picchu as individuals rather than as part of a family or community group. “Now, of course, genetics doesn’t translate into ethnicity or anything like that,” said Nesbitt of the results, “but that shows that they have distinct origins within different parts of the Inca Empire.” Confirming Archaeological and Historical Findings “The study does really reinforce a lot of other types of research that have been done at Machu Picchu and other Inca sites,” Nesbitt said. The DNA analysis supports historical documentation and archaeological studies of the artifacts found associated with the burials. This study is part of a larger movement in archaeology to combine traditional archaeological techniques with new technologies and scientific analyses. This combination of fields leads to a more complete understanding of the discoveries made.
