These compounds, identified through virtual screening, have shown promising results in stabilizing rhodopsin protein and preventing retinal degeneration in animal models. Breakthroughs in Retinitis Pigmentosa Treatment Researchers have identified two promising compounds that may help treat retinitis pigmentosa, a group of inherited eye disorders that lead to blindness. The discovery, published today (January 14th) in the open-access journal PLOS Biology, was led by Beata Jastrzebska from Case Western Reserve University, USA, and her team. These compounds were uncovered using a virtual screening technique. Retinitis pigmentosa occurs when genetic mutations cause the retinal protein rhodopsin to misfold, resulting in the death of retinal cells and progressive vision loss. Approximately 100,000 people in the United States are affected by this condition, creating an urgent need for treatments that can correct rhodopsin misfolding. Existing experimental therapies often rely on retinoid compounds, such as synthetic vitamin A derivatives. However, these treatments are highly sensitive to light and can be toxic, limiting their effectiveness and safety. Virtual Screening and Compound Discovery In the new study, researchers utilized virtual screening to search for new drug-like molecules that bind to and stabilize the structure of rhodopsin to improve its folding and movement through the cell. Two non-retinoid compounds were identified that met these criteria and had the ability to cross the blood-brain and blood-retina barriers. The team tested the compounds in the lab and showed that they improved cell surface expression of rhodopsin in 36 of 123 genetic subtypes of retinitis pigmentosa, including the most common one. Additionally, they protected against retinal degeneration in mice with retinitis pigmentosa. Promising Results and Future Directions “Importantly, treatment with either compound improved the overall retina health and function in these mice by prolonging the survival of their photoreceptors,” the authors say. However, they note that additional studies of the compounds or related compounds are needed before testing the treatments in humans. The authors add, “Inherited mutations in the rhodopsin gene cause retinitis pigmentosa (RP), a progressive and currently untreatable blinding disease. Researchers, funded by FQxI, are exploring whether biophotons may play a part in plant intelligence and could be useful for early disease diagnosis. Credit: SciTechDaily.com Statistical analyses of biophotons from lentil seeds provide evidence supporting theories that suggest a form of ‘intelligence’ may emerge in plants. All living organisms emit a faint level of light radiation, known as ‘biophotons,’ though their origin and function remain largely unexplained. An international team of physicists, supported by the Foundational Questions Institute (FQxI), has introduced a novel approach to study this phenomenon, using statistical analyses of the emitted light. Their aim is to test whether biophotons can play a role in the transport of information within and between living organisms, and whether monitoring biophotons could contribute to the development of medical techniques for the early diagnosis of various diseases. Their analyses of the measurements of the faint glow emitted by lentil seeds support models for the emergence of a kind of plant ‘intelligence,’ in which the biophotonic emission carries information and may thus be used by plants as a means to communicate. The team reported this and reviewed the history of biophotons in an article in the journal Applied Sciences in June 2024. Around a century ago, Russian biologist Alexander Gurwitsch realized that onions give off a weak electromagnetic field, related to cell growth. “Since then, scientists have found that bacteria, plants, animals, and even humans emit biophotons,” says Catalina Curceanu, a member of FQxI and an experimental nuclear and quantum physicist at the National Institute for Nuclear Physics (INFN), in Frascati, Italy. Credit: Catalina Curceanu (2024) “Some scientists think that these biophotons might be involved in the exchange of information, but so far no single model has been able to explain where they come from and what they are for,” adds Maurizio Benfatto, also at INFN, who led the data analyses. Over the years, scientists have tried to monitor biophoton emission from germinating seeds as a way of measuring their quality to study the effects of pesticides and fertilizers on plants, and also as a means for checking on food quality. Experiments with tissue slices have even shown different biophoton emission rates between tumor cells and non-malignant cells. “People even release more biophotons when they are angry,” says Benfatto. One difficulty with performing definitive experiments is that the biophoton signal is very weak and easily drowned out by surrounding lights and noise. Curceanu and her colleagues measured biophotons emanating from 76 lentil seeds in a germination chamber, housed in a darkened box, using a highly sensitive quantum photon detector. Emerging Plant Intelligence The team monitored the seeds over time windows that ranged from 10 to 60 hours. The pattern of emissions supports the notion that biophoton release is related to the activation of different cell groups, during the germination process. “Each unit can be thought of as a node in a network,” explains Benfatto, “and each node interacts with neighboring nodes before emitting biophotons.” This can be interpreted as the emergence of “cooperation and intelligence” he says, in that the units are sensitive to their nearest neighbors and also to units very far away. This global intelligence helps to determine whether releasing biophotons would increase or decrease global benefits for the plant. The work was partially funded by the Foundational Questions Institute, FQxI, which aims to catalyze research into fundamental science. “Understanding this phenomenon not only sheds new light on the mechanisms used in living matter but also opens the possibility for new ideas in the treatment of human and non-human pathologies,” says Curceanu. “I am grateful that FQxI gave us the opportunity to uncover potential links between biophotons and a form of plant intelligence.” Curceanu notes that more work is needed to uncover, for instance, where biophotons originate—possibly within mitochondria—and to confirm whether they carry information. If so, “what kind of information?” Curceanu asks. “And can we somehow change the information they carry?” Sensitive Signals Curceanu, Benfatto, and colleagues have also outlined economical ways in which future experiments could be improved to pick up these sensitive signals. This includes using a ‘Fresnel lens’ (a sectioned convex lens with vertical parallel planes forming concentric rings) to potentially improve the number of light photons collected by a factor of more than 10. The team also suggests housing the seeds within a white Teflon sphere, to improve light reflection. “Teflon reflects more than 99% of light, so biophotons will bounce around the sphere before eventually hitting the detector,” says Benfatto. “We have also included other improvements in our paper which we hope will also inspire others to investigate this fascinating natural phenomenon,” says Curceanu. Lennon and his team have been studying a synthetic minimal cell with 45% of the genes eliminated, reducing it to the smallest set of genes required for autonomous life. Despite its reduced genome, Lennon’s team found that this minimal cell evolved as quickly as a regular cell, showing the inherent resilience of life. Scientists discovered that a synthetic cell with a reduced genome could evolve as quickly as a normal cell. Despite losing 45% of its original genes, the cell adapted and demonstrated resilience in a laboratory experiment lasting 300 days, effectively showcasing that evolution occurs even under perceived limitations. “Listen, if there’s one thing the history of evolution has taught us is that life will not be contained. Life breaks free. It expands to new territories, and it crashes through barriers painfully, maybe even dangerously, but . . . life finds a way,” said Ian Malcolm, Jeff Goldblum’s character in Jurassic Park, the 1993 science fiction film about a park with living dinosaurs. You won’t find any Velociraptors lurking around evolutionary biologist Jay T. Lennon’s lab; however, Lennon, a professor in the College of Arts and Sciences Department of Biology at Indiana University Bloomington, and his colleagues have found that life does indeed find a way. Lennon’s research team has been studying a synthetically constructed minimal cell that has been stripped of all but its essential genes. The team found that the streamlined cell can evolve just as fast as a normal cell—demonstrating the capacity for organisms to adapt, even with an unnatural genome that would seemingly provide little flexibility. Credit: Image by Tom Deerinck and Mark Ellisman of the National Center for Imaging and Microscopy Research at the University of California at San Diego “It appears there’s something about life that’s really robust,” says Lennon. “We can simplify it down to just the bare essentials, but that doesn’t stop evolution from going to work.” For their study, Lennon’s team used the synthetic organism, Mycoplasma mycoides JCVI-syn3B—a minimized version of the bacterium M. mycoides commonly found in the guts of goats and similar animals. Over millennia, the parasitic bacterium has naturally lost many of its genes as it evolved to depend on its host for nutrition. Researchers at the J. Craig Venter Institute in California took this one step further. In 2016, they eliminated 45 percent of the 901 genes from the natural M. mycoides genome—reducing it to the smallest set of genes required for autonomous cellular life. At 493 genes, the minimal genome of M. mycoides JCVI-syn3B is the smallest of any known free-living organism. In comparison, many animal and plant genomes contain more than 20,000 genes. Any mutation in such an organism could lethally disrupt one or more cellular functions, placing constraints on evolution. Organisms with streamlined genomes have fewer targets upon which positive selection can act, thus limiting opportunities for adaptation. Jay T. Lennon. Credit: Photo by Indiana University Although M. mycoides JCVI-syn3B could grow and divide in laboratory conditions, Lennon and colleagues wanted to know how a minimal cell would respond to the forces of evolution over time, particularly given the limited raw materials upon which natural selection could operate as well as the uncharacterized input of new mutations. “Every single gene in its genome is essential,” says Lennon in reference to M. mycoides JCVI-syn3B. “One could hypothesize that there is no wiggle room for mutations, which could constrain its potential to evolve.” The researchers established that M. mycoides JCVI-syn3B, in fact, has an exceptionally high mutation rate. They then grew it in the lab where it was allowed to evolve freely for 300 days, equivalent to 2000 bacterial generations or about 40,000 years of human evolution. The next step was to set up experiments to determine how the minimal cells that had evolved for 300 days performed in comparison to the original, non-minimal M. mycoides as well as to a strain of minimal cells that hadn’t evolved for 300 days. In the comparison tests, the researchers put equal amounts of the strains being assessed together in a test tube. The strain better suited to its environment became the more common strain. They found that the non-minimal version of the bacterium easily outcompeted the unevolved minimal version. The minimal bacterium that had evolved for 300 days, however, did much better, effectively recovering all of the fitness that it had lost due to genome streamlining. The researchers identified the genes that changed the most during evolution. Some of these genes were involved in constructing the surface of the cell, while the functions of several others remain unknown. The research done by Lennon and his team demonstrates the power of natural selection to rapidly optimize fitness in the simplest autonomous organism, with implications for the evolution of cellular complexity.
