Reduced PRDM2 levels amplify fear memories and may link anxiety disorders with alcohol dependence. The findings provide a basis for potential treatments targeting fear memory mechanisms. A biological mechanism has been identified by researchers at Linköping University in Sweden that increases the strength with which fear memories are stored in the brain The research, conducted in rats, was published in the scientific journal Molecular Psychiatry. It provides new insights into the processes behind anxiety-related disorders and identified shared mechanisms of anxiety and alcohol dependence. The ability to feel fear is critical for escaping life-threatening circumstances and learning how to avoid them in the future. However, in certain conditions, such as post-traumatic stress disorder (PTSD) and other disorders linked to anxiety, the fear reactions become excessive and continue even when they are no longer necessary. This causes intense anxiety even if no danger is present, resulting in disability for the individual afflicted. Researchers believe that some people are predisposed to developing pathological fears, which are caused by problems with how the brain processes fearful memories. Researchers at Linköping University, including Riccardo Barchiesi and Estelle Barbier, have discovered a biological mechanism that increases the strength with which fear memories are stored in the brain. Credit: Anna Nilsen/Linköping University Amygdala and Prefrontal Cortex Some brain regions are especially important for processing fear-related memories. When threatened, the amygdala is activated and collaborates with parts of the frontal brain lobes, known as the “prefrontal cortex,” which are critical for emotion regulation. “We know that the network of nerve cells that connects the frontal lobes to the amygdala is involved in fear responses. The connections between these brain structures are altered in people with PTSD and other anxiety disorders,” says Estelle Barbier, assistant professor in the Center for Social and Affective Neuroscience (CSAN), and the Department of Biomedical and Clinical Sciences (BKV) at Linköping University, who led the study. PRDM2: A Key Protein in Fear and Stress Responses However, the molecular mechanisms involved have long remained unknown. The researchers in the current study have investigated a protein known as PRDM2, an epigenetic enzyme that suppresses the expression of many genes. The researchers have previously found that levels of PRDM2 are lower in alcohol dependence, and lead to exaggerated stress responses. In people, it is very common for alcohol dependence and anxiety-related conditions to be present at the same time, and the researchers suspect that this is caused by common mechanisms behind these conditions. In order for new memories to last, they must be stabilized and preserved as long-term memories. This process is known as “consolidation”. The researchers in the current study have investigated the effects of reduced levels of PRDM2 on the way fear memories are processed. “We have identified a mechanism in which increased activity in the network between the frontal lobes and the amygdala increases learned fear reactions. We show that down-regulation of PRDM2 increases the consolidation of fear-related memories,” says Estelle Barbier. The researchers have also identified genes that are affected when the level of PRDM2 is reduced. It became clear that this resulted in an increase in the activity of nerve cells that connect the frontal lobes and the amygdala. “Patients with anxiety disorders may benefit from treatments that weaken or erase fear memories. The biological mechanism that we have identified involves the down-regulation of PRDM2, and we currently do not have any way of increasing it. But the mechanism may be part of the explanation of why some individuals have a greater vulnerability to developing anxiety-related conditions. It may also explain why these conditions and alcohol dependence so often are present together,” says Estelle Barbier. UCLA-led research confirms and expands on protein’s role in blood vessel permeability and steps leading to white matter injury and cognitive decline. UCLA researchers have identified placental growth factor (PlGF) as a potential blood biomarker for early detection of cognitive impairment and dementia. High PlGF levels correlate with increased vascular permeability, suggesting its role in the development of cerebral small vessel disease. This finding could enable earlier identification and intervention for at-risk individuals compared to current MRI-based diagnostics. Vascular Changes and Dementia Researchers and clinicians typically use MRI scans to track brain blood vessel changes linked to cognitive impairment and dementia. These scans detect “downstream” biological markers — signs that appear later in the disease process. However, a multicenter study led by UCLA researchers suggests that a simple blood test could spot earlier changes, potentially identifying at-risk patients sooner and at a lower cost. “We studied a protein in the blood that is critical in the formation of blood vessels but that also appears to play a role in vascular permeability associated with cognitive decline,” explained Jason Hinman, MD, PhD, a vascular neurologist at UCLA Health, Interim Co-Director of the Mary S. Easton Center for Alzheimer’s Research and Care at David Geffen School of Medicine at UCLA and senior author of an article in Alzheimer’s & Dementia: The Journal of the Alzheimer’s Association. “Evaluating data from a large group of patients with a range of vascular risk profiles and cognition ranging from unimpaired to mild dementia, we found that plasma levels of this protein, placental growth factor (PlGF), could potentially be used as a biomarker to screen for and monitor cognitive impairment and dementia.” Cerebral Small Vessel Disease and Biomarkers Dysfunctional cells lining blood vessels in the brain are increasingly recognized as a key driver of processes leading to cerebral small vessel disease (CSVD), a major contributor to cognitive decline and dementia. The leaky vessels are believed to allow fluid and inflammatory molecules to seep into brain tissue. CSVD is typically diagnosed through costly brain MRI, where areas of vascular-mediated brain injury appear as bright spots on clinical MRI sequences – called white matter hyperintensities, or WMH. WMH and other structural changes are late markers of vascular brain injury. Potential of PlGF as a Biomarker The researchers studied possible associations involving several factors: plasma levels of PlGF, a highly sensitive research MRI measure of fluid accumulation in the brain called white matter free water (FW), white matter hyperintensities, and patients’ scores on cognitive assessments. Results were consistent with models suggesting that elevated PlGF increases vascular permeability, leading to accumulation of fluid in the brain’s white matter, development of white matter hyperintensities, and subsequent cognitive impairment. “As a biomarker for cerebral small vessel disease and the vascular contributions to cognitive impairment and dementia (VCID), PlGF could be used as a cost-effective screening tool for identifying patients at risk for vascular brain injury before the insidious onset of cognitive decline,” said first author Kyle Kern, MD, a vascular neurologist at UCLA Health and researcher at David Geffen School of Medicine at UCLA. “As a simple blood test, such a tool would be valuable not only for patients and clinicians, but also for researchers identifying patients for clinical trials,” he said. Study Design and Future Directions The study was conducted by researchers involved in MarkVCID, a multisite consortium established to validate candidate biomarkers for CSVD through recruitment of participants from diverse racial and ethnic backgrounds, with a range of vascular risk factors, and across the spectrum of cognitive impairment. Participants were 55 or older and had undergone brain MRI and blood tests for PlGF levels. The authors said that while the study’s multicenter design and large, diverse sample support the use of PlGF as a biomarker, additional longitudinal studies are needed to reach conclusions about causation and timing in the relationships among PlGF, FW, WMH, and cognition. Ideally, PlGF could be used to screen younger populations for whom currently available treatments and lifestyle modifications may prevent or reverse the deleterious effects of vascular injury before the onset of cognitive dysfunction. Credit: Martin Rücklin, Naturalis Biodiversity Center The origins of a pretty smile have long been sought in the fearsome jaws of living sharks which have been considered living fossils reflecting the ancestral condition for vertebrate tooth development and inference of its evolution. However, this view ignores real fossils which more accurately reflect the nature of ancient ancestors. New research led by the University of Bristol and the Naturalis Biodiversity Center published in Nature Ecology and Evolution reveals that the dentitions of living shark relatives are entirely unrepresentative of the last shared ancestor of jawed vertebrates. The study reveals that while teeth evolved once, complex dentitions have been gained and lost many times in evolutionary history and tooth replacement in living sharks is not the best model in the search for therapeutic solutions to human dental pathologies. Lead author Martin Rücklin from Naturalis Biodiversity Center in The Netherlands said: “We used high energy x-rays at the TOMCAT beamline of the Swiss Light Source at the Paul Scherrer Institut in Switzerland, to study tooth and jaw structure and development among shark ancestors. This organization and pattering of teeth is thought to originate in a universal development mechanism, the dental lamina, seen in sharks.
