University of Oregon researchers discovered a 480-million-year-old sex chromosome in the California two-spot octopus, making it one of the oldest known animal sex chromosomes. This finding confirms that cephalopods use chromosomes to determine sex. The chromosome was identified in female octopuses but not males, and further research found it in other cephalopods, suggesting an ancient and stable system. New research at the UO reveals that they have been using one that has existed for 480 million years. The octopus has just revealed another secret: what determines its sex. Researchers at the University of Oregon have identified a sex chromosome in the California two-spot octopus. This chromosome has likely existed for 480 million years, predating the evolutionary split between octopuses and nautiluses. That makes it one of the oldest known animal sex chromosomes. The discovery also provides evidence that octopuses and other cephalopods—a group of sea animals that includes squid and nautiluses—use chromosomes to determine their sex, solving a longstanding mystery among biologists. “Cephalopods are already such interesting creatures, and there are so many things we’re still learning about them, especially in neuroscience,” said Gabby Coffing, a doctoral student at the UO working in the lab of biologist Andrew Kern. “This is just showing one more interesting thing about them: They have really ancient sex chromosomes.” Coffing, Kern, and their team described the findings on February 3 in the journal Current Biology. How Animals Determine Sex In humans and most mammals, sex is determined largely by chromosomes. But “there’s a tremendous amount of diversity” in how animals determine their sex, Kern said. So scientists couldn’t assume the same was true for octopuses. In turtles, for instance, sex is determined by the temperature at which the eggs are incubated. Some fish have a gene that determines sex, but not a whole chromosome. Even in humans, the X/Y sex chromosome system isn’t as clear-cut as it might look on paper; gene mutations or inheriting extra sex chromosomes can lead to development that doesn’t neatly fit in a male/female binary. Plus, because cephalopods aren’t standard lab animals, like mice or fruit flies, they haven’t been subject to nearly as much genetic exploration. Scientists have sequenced the genomes of a handful of octopus species, but they can’t link genes to specific traits the way they can in mice or even humans. When UO researchers recently sequenced the DNA of a female California two-spot octopus, they found something unexpected: a chromosome with only half the amount of genetic material. It looked different from all the others, and it hadn’t been found in male octopuses whose DNA was previously sequenced. “This particular chromosome had half the amount of sequencing data, which indicated there was only one copy,” said Coffing. “Then as we explored that more, we reached the conclusion that we must have stumbled upon a sex chromosome.” Tracing the Chromosome’s Evolutionary History To confirm, the researchers sorted through other octopus genomic data previously collected by other researchers. Not all that data was clearly labeled as being from male or female octopuses. But they found another example of the half-sized chromosome in another species of octopus. They also found it in squid, which diverged evolutionarily from octopuses somewhere between 248 and 455 million years ago. And after more digging, they also found evidence for the chromosome in the nautilus, a mollusk that split apart from the octopus approximately 480 million years ago. The fact that these species share this unique chromosome suggests that it’s been around in some form for a very long time. “This indicates that their common ancestor had this similar sex determination system,” Coffing said. That’s somewhat unusual for sex chromosomes, Kern said. Because they directly impact reproductive capabilities, they’re subject to a lot of selective pressure and so tend to undergo rapid evolutionary change. But cephalopods seem to have found what works and have stuck with it. Other ancient sex chromosomes have been discovered in plant groups like mosses and liverworts, which were some of the first plants to evolve. And insect sex chromosomes might be 450 million years old, but they’ve also changed a lot over time. Kern and his colleagues initially thought octopuses might have a sex determination system similar to birds and butterflies, where males are ZZ and females are ZW. (Biologists have given sex determination systems where males have two copies of the same sex chromosome different letters, to avoid confusion with the XX/XY system where females have two copies of the same chromosome.) But the team hasn’t yet found a W chromosome in an octopus. Alternatively, octopuses could use a sex determination system that only involves the Z chromosome — males have a pair, and females just have one. That’s still to be determined, Coffing said. Commercial whaling in the 20th century decimated populations of large whales but also appears to have had a lasting impact on the genetic diversity of today’s surviving whales, new research from Oregon State University shows. Researchers compared DNA from a collection of whale bones found on beaches near abandoned whaling stations on South Georgia Island in the South Atlantic Ocean to DNA from whales in the present-day population and found strong evidence of loss of maternal DNA lineages among blue and humpback whales. Credit: Bob Pittman, Oregon State University “A maternal lineage is often associated with an animal’s cultural memories such as feeding and breeding locations that are passed from one generation to the next,” said the study’s lead author, Angela Sremba, who conducted the research as part of her doctoral studies at Oregon State University’s Marine Mammal Institute. “If a maternal lineage is lost, that knowledge is likely also lost.” The findings were published recently in the Journal of Heredity. Whaling’s Historical Footprint South Georgia is a remote island about 800 miles southeast of the Falkland Islands and home to several whaling stations operating from the turn of the century through the 1960s. In a little over 60 years, more than 2 million whales were killed throughout the Southern Hemisphere, of which 175,000 were killed near South Georgia. Credit: Scott Baker, Marine Mammal Institute Evidence of that slaughter is still present on the island, which is littered with thousands of whale bones – many 100 years old or more – that were discarded during commercial processing. Cold temperatures in the region aided in their preservation. South Atlantic whale populations have begun to recover since commercial whaling was halted, but whale sightings around South Georgia remained low. This suggests that local populations may have been extirpated – a term to describe a type of localized extinction, said study co-author Scott Baker, associate director of OSU’s Marine Mammal Institute. Baker last visited South Georgia Island as part of a research voyage in early 2020. “For 60 years, the whales have been absent from the South Georgia feeding grounds, suggesting that cultural memory was lost,” said Baker, who was Sremba’s Ph.D. advisor. “The numbers of whales returning to this region today are still not large, but there is a sense that they may be rediscovering this habitat.” Genetic Insights and the Future To better understand how whaling may have impacted the genetic diversity of today’s population, Sremba analyzed DNA extracted from bones found on South Georgia Island and compared the genetic information with previously published data from living whales in the contemporary post-whaling population. Credit: Marine Mammal Institute, Oregon State University She and colleagues identified bones of humpback, blue, and fin whales, and found that while genetic diversity among the whales remains high, there are indications of a loss of maternal DNA lineages in the blue and humpback populations. Researchers were not able to detect differences in diversity between pre- and post-whaling DNA samples from Southern Hemisphere fin whales, likely due to the limited availability of post-whaling samples. Because some whale species can live up to 100 years, there is also a chance that some of today’s whales were alive during the whaling era, said Sremba, who is now a researcher with the Cooperative Institute for Marine Ecosystem and Resources Studies at OSU’s Hatfield Marine Science Center in Newport. As these whales die, that could result in further loss of additional maternal lineages. This underscores the importance of preserving genetic information from the whales now, she said. “It’s remarkable these species survived. In another 100 years, we don’t know what might change, and we can’t measure any change now if we don’t have a good understanding of the past,” Sremba said. “This work provides an opportunity to reconstruct the history of these whale populations and help us understand what was truly lost due to whaling activities.” Rising temperatures due to climate change could also lead to deterioration of the DNA in the bones on South Georgia Island, Baker said. “This work is a way to preserve this history indefinitely,” he said. A group of scientists led by the American Museum of Natural History and Bat Conservation International have discovered a new species of a striking orange and black bat in a mountain range in West Africa. The species, which the researchers expect is likely critically endangered, underscores the importance of sub-Saharan “sky islands” to bat diversity. The species is described today in the journal American Museum Novitates. “In an age of extinction, a discovery like this offers a glimmer of hope,” said Winifred Frick, chief scientist at Bat Conservation International and an associate research professor at the University of California, Santa Cruz. “It’s a spectacular animal. It has this bright-orange fur, and because it was so distinct, that led us to realize it was not described before. Discovering a new mammal is rare. Credit: Fiona Reid Fieldwork in the Nimba Mountains In 2018, Frick and her colleagues at Bat Conservation International and the University of Maroua in Cameroon were in the Nimba Mountains in Guinea conducting field surveys in natural caves and mining tunnels, known as adits, that were built in the 1970s and 1980s and have since been colonized by bats. In collaboration with the local mining company, Société des Mines de Fer de Guinée (SMFG), the scientists are trying to understand which bat species use which adits and at what times of the year. Of particular interest is Lamotte’s roundleaf bat, Hipposideros lamottei, which is listed by the International Union for Conservation of Nature (IUCN) as critically endangered and has only ever been recorded in the Nimba Mountains. Much of its known population lives in the adits, which are in different states of collapse and will disappear in time. While surveying for this bat, the researchers found something peculiar — a bat that looked nothing like Lamotte’s roundleaf bat and did not match the descriptions of any other species that they knew occurred in the area. Later that night, they called on American Museum of Natural History Curator Nancy Simmons, a bat expert and chair of the Museum’s Department of Mammalogy, for help. “As soon as I looked at it, I agreed that it was something new,” said Simmons, the lead author of the paper and Bat Conservation International Board member. “Then began the long path of documentation and gathering all the data needed to show that it’s indeed unlike any other known species.” A chain of “African sky islands,” the Nimba Mountains in Guinea have peaks rising between 1,600-1,750 meters (about 1 mile) above sea level and are surrounded by drastically different lowland habitats. Credit: © Bat Conservation International Through morphological, mor¬phometric, echolocation, and genetic data, including comparative data from collections at the Museum, the Smithsonian National Museum of Natural History, and the British Museum, the scientists described the new species, which they named Myotis nimbaensis (“from Nimba”) in recognition of the mountain range in which it is found. A chain of “African sky islands,” the Nimba Mountains have peaks rising between 1,600-1,750 meters (about 1 mile) above sea level and are surrounded by drastically different lowland habitats. As such, they are home to exceptional biodiversity, including bats. The Nimba Mountains: A Hotspot for Unique Biodiversity “In addition to the Lamotte’s roundleaf bat, it’s possible Myotis nimbaensis could be the second bat species found only in this particular mountain range,” said Jon Flanders, Bat Conservation International’s director of endangered species interventions. This study is part of an ongoing effort critical in helping the Nimba Mountain bats survive. Bat Conservation International and SMFG have already started working together to build new tunnels, reinforced to last for centuries and in habitat away from the mining project, for the Lamotte’s roundleaf bat. And although little is known yet about the population and range of Myotis nimbaensis, efforts like this will likely help it as well.
