This research could help understand CALHM1’s role in Alzheimer’s disease and taste perception, providing a basis for potential drug development. Researchers at Cold Spring Harbor Laboratory have generated detailed images of a neuronal channel known as Calcium Homeostasis Modulator Protein 1 (CALHM1), which plays significant roles in various physiological processes, including taste perception and possibly Alzheimer’s disease regulation. The research highlights the structure and functionality of the CALHM1 channel and emphasizes the role of phospholipids in stabilizing the channel. The findings offer a foundation for further exploration into CALHM1’s role in human health and its potential as a drug target. The Role of CALHM1 Protein in Neurons The human body’s neurons are speckled with tiny pores that enable the passage of essential molecules into and out of our cells. These channels are vital for neurons to transmit signals that facilitate our movement, cognition, and perception of the world. Recently, structural biologists at Cold Spring Harbor Laboratory (CSHL) have acquired unprecedented images of one of the most sizable pores present in human neurons, known as the Calcium Homeostasis Modulator Protein 1, or CALHM1. CALHM1 and Alzheimer’s Disease Past studies suggest that mutations in the Cahlm1 gene might elevate the risk for Alzheimer’s disease. The latest research from CSHL offers new insights into how the CALHM1 channel operates in humans and the ways in which it can become obstructed. Functions of CALHM1 CSHL Professor Hiro Furukawa and postdoctoral researcher Johanna Syrjänen have devoted several years to the study of CALHM1, which appears to be implicated in a broad array of physiological processes. In our tongues, CALHM1 contributes to our perception of tastes like sweet, sour, or umami. In our brains, it may play a part in regulating the accumulation of a plaque-forming protein linked to Alzheimer’s. Structure and Regulation of CALHM1 Furukawa, Syrjänen, and their team employed a technique known as cryo-electron microscopy to produce detailed three-dimensional images of the human CALHM1 channel. These images illustrate how eight copies of the CALHM1 protein come together to form the circular channel. Each protein features a flexible appendage that extends into the pore, potentially managing its opening and closing, a characteristic that Syrjänen equates to “octopus tentacles.” Cryo-electron microscopy reveals that the human CALHM1 channel has an eight-protein assembly pattern, similar to that found in chickens. These important fats are abundant in eggs, cereal, lean meats, and seafood. Additionally, the team demonstrated how a chemical commonly used by researchers to block CALHM1 can become lodged in the channel, a finding that could prove beneficial for potential drug development targeting CALHM1. Future Implications of CALHM1 Research Syrjänen says: “If you are thinking way down the line, ‘Can we control taste perception or influence this protein?’ we now know one of the places where you could block the protein activity.” Syrjänen notes that the human CALHM1 channel looks a lot like the version she and Furukawa studied in chickens in 2020. However, determining the structure of the human protein proved more technically challenging. But, researchers agree, it’s crucial to gain a deeper understanding of the channel’s impact on human health. “There are numerous unanswered questions surrounding CALHM1,” Furukawa says. For example, how does the energy-carrying molecule, ATP, escape from cells via this channel? The study was designed to take great care to avoid any discomfort the participants could experience when targeting such a sensitive body region. The somatosensory cortex represented the genitals next to the hips, matching the body’s anatomy. However, the precise location varied from woman to woman. The thickness of the genital field varied with the frequency of sexual intercourse, suggesting the region’s structure alters in relation to its use. These results allow for future studies examining the role of the genital field in healthy sexual function, sexual dysfunction, and especially in the long-term consequences of sexual abuse. Based on this precise mapping, future work can now potentially target the genital representation for treatment of clinical conditions. Researchers found that a gene mutation linked to blindness can also increase intelligence. When genes mutate, it can result in severe diseases of the human nervous system. Neuroscientists at Leipzig University and the University of Würzburg have now used fruit flies to demonstrate how, apart from the negative effect, the mutation of a neuronal gene can have a positive effect – namely higher IQ in humans. They have published their findings in the prestigious journal Brain. Synapses are the contact points in the brain via which nerve cells ‘talk’ to one another. Disruptions in this communication lead to nervous system diseases, since altered synaptic proteins, for example, can impair this complex molecular mechanism. This can cause mild symptoms, but also very severe disabilities in those affected. Mutation Linked to Above-Average Intelligence The interest of the two neurobiologists Professor Tobias Langenhan and Professor Manfred Heckmann, from Leipzig and Würzburg respectively, was aroused when they read in a scientific publication about a mutation that damages a synaptic protein. At first, the affected patients attracted scientists’ attention because the mutation caused them to go blind. However, doctors then noticed that the patients were also of above-average intelligence. “It’s very rare for a mutation to lead to improvement rather than loss of function,” says Langenhan, professor and holder of a chair at the Rudolf Schönheimer Institute of Biochemistry at the Faculty of Medicine. Research on fruit flies helps to better understand diseases of the human nervous system. Credit: Swen Reichhold/Leipzig University The two neurobiologists from Leipzig and Würzburg have been using fruit flies to analyze synaptic functions for many years. “Our research project was designed to insert the patients’ mutation into the corresponding gene in the fly and use techniques such as electrophysiology to test what then happens to the synapses. It was our assumption that the mutation makes patients so clever because it improves communication between the neurons which involve the injured protein,” explains Langenhan. “Of course, you can’t conduct these measurements on the synapses in the brains of human patients. You have to use animal models for that.” “75 percent of genes that cause diseases in humans also exist in fruit flies” First, the scientists, together with researchers from Oxford, showed that the fly protein called RIM looks molecularly identical to that of humans. This was essential in order to be able to study the changes in the human brain in the fly. In the next step, the neurobiologists inserted mutations into the fly genome that looked exactly as they did in the diseased people. They then took electrophysiological measurements of synaptic activity. “We actually observed that the animals with the mutation showed a much increased transmission of information at the synapses. This amazing effect on the fly synapses is probably found in the same or a similar way in human patients, and could explain their increased cognitive performance, but also their blindness,” concludes Professor Langenhan. Credit: Swen Reichhold Molecular Insights into Enhanced Synaptic Transmission The scientists also found out how the increased transmission at the synapses occurs: the molecular components in the transmitting nerve cell that trigger the synaptic impulses move closer together as a result of the mutation effect and lead to increased release of neurotransmitters. A novel method, super-resolution microscopy, was one of the techniques used in the study. “This gives us a tool to look at and even count individual molecules and confirms that the molecules in the firing cell are closer together than they normally are,” says Professor Langenhan, who was also assisted in the study by Professor Hartmut Schmidt’s research group from the Carl Ludwig Institute in Leipzig. “The project beautifully demonstrates how an extraordinary model animal like the fruit fly can be used to gain a very deep understanding of human brain disease. The animals are genetically highly similar to humans. It is estimated that 75 percent of the genes involving disease in humans are also found in the fruit fly,” explains Professor Langenhan, pointing to further research on the topic at the Faculty of Medicine: “We have started several joint projects with human geneticists, pathologists and the team of the Integrated Research and Treatment Center (IFB) AdiposityDiseases; based at Leipzig University Hospital, they are studying developmental brain disorders, the development of malignant tumors and obesity.
