University of Maryland biologists have discovered that geckos use a part of their inner ear, the saccule, to detect low-frequency vibrations, offering a new understanding of reptilian hearing. This finding may extend to other reptiles and changes how scientists view animal communication and perception. It also hints at possible connections between hearing and balance in humans. Credit: Duncan Leitch Researchers at the University of Maryland have found that geckos utilize a mechanism typically linked to balance to detect vibrations, providing fresh insights into how animals perceive auditory and tactile stimuli. Biologists at the University of Maryland have discovered a previously unknown talent in geckos that’s shaking up what we thought we knew about animal hearing. In a new study published in Current Biology on October 4, 2024, the researchers revealed that geckos use the saccule—a part of their inner ear traditionally associated with maintaining balance and body positioning—to detect low-frequency vibrations. According to the researchers, this special “sixth sense” also plays a complementary role to the geckos’ normal hearing and the way they sense the world around them. The team believes that this previously unrecognized hearing mechanism may be present in other reptilian species as well, challenging existing ideas about how animal sensory systems evolved and diverged over time. A New Role for the Saccule in Gecko Hearing “The ear, as we know it, hears airborne sound. But this ancient inner pathway, which is typically linked to balance, helps geckos detect vibrations that travel through mediums like the ground or water,” said study co-author Catherine Carr, a Distinguished University Professor of Biology at UMD. “This pathway exists in amphibians and fish, and now it’s proven to be preserved in lizards as well. Our findings shed light on how the auditory system evolved from what you see in fish to what you see in land animals including humans.” The saccule can detect faint vibrations ranging from 50 and 200 Hz, a spectrum well below what geckos can usually hear through their ears. Researchers say this indicates that the saccule serves a distinct yet complementary function to the geckos’ regular auditory system. While geckos can hear airborne sound, many other reptiles do not have that ability. The study’s lead author Dawei Han, a postdoctoral researcher and former graduate student at UMD, says that the discovery of the saccule’s role in gecko hearing may lead to a better understanding of communication and behavior in other animals previously considered to have limited auditory capabilities. Broader Implications for Reptiles and Evolution “A lot of snakes and lizards were thought to be ‘mute’ or ‘deaf’ in the sense that they do not vocalize sounds or hear sounds well,” Han explained. “But it turns out they could potentially be communicating via vibrational signals using this sensory pathway instead, which really changes the way scientists have thought about animal perception overall.” The existence of this shared sensory pathway in modern reptiles offers a unique window into the evolutionary history of vertebrate sensory systems, suggesting that the transition from aquatic to terrestrial environments likely involved more complex and gradual changes in hearing mechanisms than previously thought. Although these findings are not directly connected to how humans hear, the researchers believe that there’s always more than meets the eye—or in this case, ear. “Think about when you’re at a live rock concert,” Carr said. “It’s so loud that you can feel your whole head and body vibrate in the sound field. You can feel the music, rather than just hearing it. That feeling suggests that the human vestibular system may be stimulated during those loud concerts, meaning our sense of hearing and balance may also be linked closely.” Carr and Han hope their findings will prompt more investigations into mammalian hearing, especially in the context of this sensory pathway. UCLA-led research confirms and expands on protein’s role in blood vessel permeability and steps leading to white matter injury and cognitive decline. UCLA researchers have identified placental growth factor (PlGF) as a potential blood biomarker for early detection of cognitive impairment and dementia. High PlGF levels correlate with increased vascular permeability, suggesting its role in the development of cerebral small vessel disease. This finding could enable earlier identification and intervention for at-risk individuals compared to current MRI-based diagnostics. Vascular Changes and Dementia Researchers and clinicians typically use MRI scans to track brain blood vessel changes linked to cognitive impairment and dementia. These scans detect “downstream” biological markers — signs that appear later in the disease process. However, a multicenter study led by UCLA researchers suggests that a simple blood test could spot earlier changes, potentially identifying at-risk patients sooner and at a lower cost. “We studied a protein in the blood that is critical in the formation of blood vessels but that also appears to play a role in vascular permeability associated with cognitive decline,” explained Jason Hinman, MD, PhD, a vascular neurologist at UCLA Health, Interim Co-Director of the Mary S. Easton Center for Alzheimer’s Research and Care at David Geffen School of Medicine at UCLA and senior author of an article in Alzheimer’s & Dementia: The Journal of the Alzheimer’s Association. “Evaluating data from a large group of patients with a range of vascular risk profiles and cognition ranging from unimpaired to mild dementia, we found that plasma levels of this protein, placental growth factor (PlGF), could potentially be used as a biomarker to screen for and monitor cognitive impairment and dementia.” Cerebral Small Vessel Disease and Biomarkers Dysfunctional cells lining blood vessels in the brain are increasingly recognized as a key driver of processes leading to cerebral small vessel disease (CSVD), a major contributor to cognitive decline and dementia. The leaky vessels are believed to allow fluid and inflammatory molecules to seep into brain tissue. CSVD is typically diagnosed through costly brain MRI, where areas of vascular-mediated brain injury appear as bright spots on clinical MRI sequences – called white matter hyperintensities, or WMH. WMH and other structural changes are late markers of vascular brain injury. Potential of PlGF as a Biomarker The researchers studied possible associations involving several factors: plasma levels of PlGF, a highly sensitive research MRI measure of fluid accumulation in the brain called white matter free water (FW), white matter hyperintensities, and patients’ scores on cognitive assessments. Results were consistent with models suggesting that elevated PlGF increases vascular permeability, leading to accumulation of fluid in the brain’s white matter, development of white matter hyperintensities, and subsequent cognitive impairment. “As a biomarker for cerebral small vessel disease and the vascular contributions to cognitive impairment and dementia (VCID), PlGF could be used as a cost-effective screening tool for identifying patients at risk for vascular brain injury before the insidious onset of cognitive decline,” said first author Kyle Kern, MD, a vascular neurologist at UCLA Health and researcher at David Geffen School of Medicine at UCLA. “As a simple blood test, such a tool would be valuable not only for patients and clinicians, but also for researchers identifying patients for clinical trials,” he said. Study Design and Future Directions The study was conducted by researchers involved in MarkVCID, a multisite consortium established to validate candidate biomarkers for CSVD through recruitment of participants from diverse racial and ethnic backgrounds, with a range of vascular risk factors, and across the spectrum of cognitive impairment. Participants were 55 or older and had undergone brain MRI and blood tests for PlGF levels. The authors said that while the study’s multicenter design and large, diverse sample support the use of PlGF as a biomarker, additional longitudinal studies are needed to reach conclusions about causation and timing in the relationships among PlGF, FW, WMH, and cognition. Ideally, PlGF could be used to screen younger populations for whom currently available treatments and lifestyle modifications may prevent or reverse the deleterious effects of vascular injury before the onset of cognitive dysfunction. As they are walking or running, they make a continuous series of noises. Researchers have decoded how wild meerkats utilize two of these sounds. “The first sound, a ‘close call’, is like a call-and-response exchange between the animals. When one meerkat calls, a neighbor is likely to reply,” explains Vlad Demartsev, a postdoctoral researcher from the Cluster of Excellence Collective Behaviour. “Whereas the second call, named a ‘short note’, announces ‘I am here’ but doesn’t necessarily get a direct reply from communication partners.” Meerkats are highly social animals and live in groups of up to 50 members. Credit: Vlad Demartsev Vocal Interactions: Exchange vs. Broadcast Consider an announcement in front of a large crowd where the flow of information is mostly one-way, with no real exchange between the speaker and the audience. “It is impossible to hold a conversation with 20 people, so we normally talk to one partner at a time,” explains Demartsev. Close calls are such an exchange between communication partners and short notes are more like a broadcast or an announcement aimed at the whole group. Credit: Vlad Demartsev Research Methodology Demartsev, along with Ariana Strandburg-Peshkin and collaborators from the University of Zurich, equipped several groups of meerkats with collars at the Kalahari Research Centre in South Africa. The collars recorded continuous audio data and GPS positions were sampled every second. Using these collars, the researchers got a synchronized recording and could see which animal produced which sound at which time and where. Researchers used collars that recorded audio and GPS data to track the origin of each sound, identifying the specific meerkat making the call, their location, and the timing of the sound. Credit: Vlad Demartsev Analysis of Vocal Interactions The researchers prepared a vocal timeline for the entire group and analyzed the data. “We saw that when a close call is given, there is a very high probability that within less than half a second a nearby neighbor will respond. But when we have a short note, we do not have this pattern. All of them are calling nearly at the same time and there is no structure,” says Demartsev. Strandburg-Peshkin adds, “Ultimately, calls are not just single isolated events, but a continuous stream of communication between group members. So, looking at the temporal structure of the interactions can help us to better understand how calls are used and what their function is.” Group cohesion is critical for meerkats because isolation increases their risk of predation or harassment by other groups, highlighting the importance of their communication system. Credit: Vlad Demartsev The Importance of Group Cohesion Staying in a group is crucial for meerkats and they use multiple mechanisms that evolved to avoid getting separated. “When meerkats are by themselves there is a higher chance of predation or harassment by other groups. Generally, meerkats therefore try very, very hard to stay together,” says Demartsev.
