Scientists from the Salk Institute have discovered two distinct neural pathways that mice use to encode mechanical and chemical sensations of itch, with a particular population of neurons transmitting mechanical itch information from the spinal cord to the brain. The identification of separate neural pathways governing itch-scratch responses and chronic itch conditions in mice lays the groundwork for the development of novel therapeutic targets. Itch serves as a defensive alert in animals, acting to prevent the introduction of potentially harmful pathogens into their bodies by parasites. For instance, when a mosquito alights on a person’s arm, they become aware of its touch and instinctively scratch the area to dispel it. This sensation of itchiness arising from a physical stimulus like a crawling insect is termed as “mechanical,” differing from the “chemical” itchiness provoked by irritants such as a mosquito’s saliva following a bite. Despite eliciting the same reaction (scratching), recent investigations by scientists at the Salk Institute have discovered that, in mice, there is a specific brain pathway controlling the mechanical sensation which is separate from the neural pathway that is responsible for encoding the chemical sensation. Identified mechanical itch-responsive neuron (blue) located among cell nuclei (green) in the brainstem. Credit: Salk Institute Key Neurons and Neuropeptide Signals Identified Their findings, recently published in the journal Neuron, show that a small population of neurons relay mechanical itch information from the spinal cord to the brain and identify the neuropeptide signals that regulate both itch types. “This study provides fundamental insights into how these two forms of itch are encoded by the brain and opens up new avenues for therapeutic interventions for patients that suffer from a range of chronic itch conditions, including ectopic dermatitis and psoriasis,” says co-corresponding author Martyn Goulding, professor and holder of the Frederick W. and Joanna J. Mitchell Chair. The discovery builds on previous work in Goulding’s lab that had identified the neurons in the spinal cord that control mechanical itch and not chemical itch. Members of Goulding’s lab teamed up with co-corresponding author Sung Han, assistant professor and holder of the Pioneer Fund Developmental Chair, who had previously found that a small region of the brain serves as an alarm center that fields threat signals, both external and internal from within the body. Han’s team had noticed that a specific group of neurons were crucial for encoding threat signals. Goulding’s lab then decided to focus on these neurons and ask if they play a specific role in relaying mechanical itch signals to this alarm center. From left: Martyn Goulding and Sung Han. Credit: Salk Institute The team used genetic approaches combined with wearable miniaturized microscopes that allowed the researchers to see itch-induced activity in single neurons of mice. The scientists discovered that by removing an inhibitory pathway involved in itch, they could activate a mechanical itch. By observing subsequent activity and changes occurring in the brainstem, they saw that different cells were responding to either mechanical or chemical itch. This allowed the team to classify distinctions between a chemical itch pathway and a mechanical itch pathway and clearly identify the molecules important for regulating them. Chronic Itch and Its Pathological Drivers “We found that if you sensitize one pathway, you can stimulate a pathological itch state, and vice versa,” says Han. “This indicates that these two pathways act together to drive chronic itch.” Next, the scientists plan to examine where in the brain these pathways converge, and then explore the parts of the brain that receive signals that determine the decision of whether to scratch an itch. They also want to better understand how the spinal cord and brainstem differentiate between pain and itch. “The prevalence of chronic itch increases as we age. For this reason, we would also like to understand more about what is happening to the neural circuits that relay itch as we get older,” says Goulding. “Given that chronic itch is an intractable problem, our findings should help leverage the development of new therapies for treating it.” Reference: “Identification of an essential spinoparabrachial pathway for mechanical itch” by Xiangyu Ren, Shijia Liu, Amandine Virlogeux, Sukjae J. Since then, these bats have diversified into many different species by changing what they eat. Credit: Stowers Institute for Medical Research, edited Researchers recorded the highest natural blood sugar levels ever observed in a mammal, now they hope it could help them better understand diabetes. A recent study highlights how Neotropical leaf-nosed bats manage exceptionally high blood sugar levels through unique evolutionary adaptations across various species. This research could provide new directions for treating human metabolic disorders. Evolutionary Insights From Bats on Blood Sugar Management Humans must regulate blood sugar concentrations to stay healthy and to fuel our cells. Too little or too much can cause serious health complications, and high blood sugar is a hallmark of the metabolic condition, diabetes. New research from the Stowers Institute for Medical Research may enable potential solutions to metabolic disease by turning to evolution and to bats. Published today (August 28, 2024) in Nature Ecology and Evolution, the study led by co-first authors Postdoctoral Research Associate Jasmin Camacho, Ph.D., and former Stowers researcher Andrea Bernal-Rivera from the lab of Stowers Associate Investigator Nicolas Rohner, Ph.D., reports the highest naturally occurring blood sugar concentrations in mammals ever observed, a finding that suggests bats have evolved strategies to survive, and even thrive, with this extreme trait. The new study reports the highest naturally occurring blood sugar concentrations in mammals ever observed, a finding that suggests bats have evolved strategies to survive, and even thrive, with this extreme trait. Credit: Stowers Institute for Medical Research Unique Dietary Adaptations in Neotropical Leaf-Nosed Bats “Our study reports blood sugar levels that are the highest we have ever seen in nature—what would be lethal, coma-inducing levels for mammals, but not for bats,” said Camacho. “We are seeing a new trait we didn’t know was possible.” Thirty million years ago, the Neotropical leaf-nosed bat survived solely on insects. Since then, these bats have diversified into many different species by changing what they eat. From insects, different lineages now specialize on diets ranging from fruits, nectar, meat, and everything in between—even just blood. “Looking to animals that have existed for millions of years allows us to start to catalog changes that have happened over evolution,” said Camacho. “What makes Neotropical leaf-nosed bats so unique to study is that this group is comprised of many different species with very diverse diets, making it feasible to find answers about how diet evolves. The hope is that we can extend this understanding to other mammals, including humans, where there may be ways to learn how to better protect our own health.” “Looking to animals that have existed for millions of years allows us to start to catalog changes that have happened over evolution,” said Camacho. Credit: Stowers Institute for Medical Research Field Research on Bat Glucose Tolerance To uncover how bats diversified their diets, the team traveled to the jungles of Central America, South America, and the Caribbean to conduct fieldwork over several years. These catch-and-release expeditions were focused on performing glucose tolerance tests—measuring the concentration of sugar in blood—on nearly 200 wild-caught bats across 29 species after a single feeding of one of three types of sugars associated with diets of insects, fruits, or nectar. “We saw various ways sugar is assimilated—absorbed, stored, and used in the body—and how this process has become specialized due to different diets,” said Bernal-Rivera. Implications for Human Metabolic Research The mechanism for maintaining blood sugar levels within a narrow, healthy range is called glucose homeostasis, which is typically regulated by the hormone insulin and is what goes awry in diabetes. Different species of leaf-nosed bats reveal a spectrum of adaptations to glucose homeostasis, ranging from changes in intestinal anatomy to genetic alterations for proteins that transport sugar from blood to cells. “Fruit bats have honed their insulin signaling pathway to lower blood sugar,” said Camacho. “On the other extreme, nectar bats can tolerate high blood glucose levels, similar to what is observed in people with unregulated diabetes. They have evolved a different mechanism, and it does not seem to depend on insulin.” Although precisely how nectar bats are managing glucose is still under investigation, the researchers found potential clues for alternative metabolic strategies for glucose regulation. Bats with sugar-rich diets were observed to have longer portions of their intestines and to have intestinal cells with greater surface areas for absorbing nutrients from food, compared to bats with other diets. In addition, nectar bats, unique from fruit bats, have a continual expression of a gene responsible for sugar transport, a trait also observed in a species of hummingbird. “This study establishes extremely important resources for the field,” said Nadav Ahituv, Ph.D., a bioengineering and genetics professor at the University of California, San Francisco. “It provides not only metabolic characteristics of various bat species with different diets, but also their intestinal morphology, and candidate genomic regions and protein structural differences that could be driving dietary adaptations.” “The datasets will fuel future research that aims to differentiate mammalian dietary differences and could progress the development of novel therapeutics for a variety of metabolic diseases in humans,” said Ahituv. The heart (cyan) forms from two distant regions of the embryo (far left). These regions migrate to the embryo midline, where they fuse into a tube to make the first heart structure (far right). Precise alignment and pairing of these cells are crucial for proper heart development. Credit: Thamarailingam Athilingam and Kate McDole Cells in the developing heart must find the perfect match, much like a game of microscopic speed dating. Using filopodia, tiny tentacle-like structures, they probe their environment and latch onto potential partners. If they mismatch, proteins step in to separate them, ensuring precise alignment. Researchers modeled this process in fruit flies, uncovering the delicate balance of adhesive energy and elasticity that guides cell organization. How Developing Heart Cells Find Their Perfect Match In a developing heart, cells move around, jostling and bumping into each other as they search for their correct position. The stakes are high—pairing with the wrong cell could mean the difference between a properly beating heart and one that doesn’t function correctly. A study published today (March 12) in the Biophysical Journal explores this intricate “matchmaking” process. Researchers created a model to track how heart cells move and interact, helping predict how genetic variations might disrupt heart development in fruit flies. In both humans and fruit flies, heart tissue forms from two separate regions in the embryo, starting far apart. As development progresses, these cells migrate toward each other and eventually merge into a tube-like structure that becomes the heart. For this process to work, cells must align precisely and pair up correctly. The Cellular Dance of Finding the Right Partner “As the cells come together, they jiggle and adjust, and somehow always end up pairing with a heart cell of the same type,” says the lead author, Timothy Saunders of the University of Warwick. This observation inspired the team to explore how cells match up in the first place and how they know when they’ve found the right fit. Developing heart cells use thin, tentacle-like structures called filopodia to explore their surroundings and latch onto potential partners. Saunders’ earlier research found that proteins generate waves that push mismatched cells apart, giving them another chance to find the correct match. “It’s basically like cells are speed dating,” says Saunders. “They have just a few moments to determine if they’re a good match, with molecular ‘friends’ ready to pull them apart if they’re not compatible.” The Science of Stability: How Cells Settle in Place The researchers found that heart cells seek stability where they remain closest to stillness—like a rolling ball that eventually comes to a stop, known as energy equilibrium in physics. In developing heart cells, this principle applies when cells find a balance between connection forces and their ability to adjust to strain—also known as adhesive energy and elasticity. Based on this observation, the team developed a model that shows how cells can self-organize. Next, the team tested their model on fruit fly hearts with mutations and misalignments. By calculating the adhesive energy between different cell types and assessing tissue elasticity, the model predicted how cells would match and rearrange. “Although rare, sometimes the heart tube ends up with one cell on one side when it should have two, or two cells when there should be four,” says Saunders. “We could input these imperfections into the model and run it.” The model produced outcomes that closely mirrored what was observed in real embryos. Beyond the Heart: Why This Research Matters The team notes that their model not only enhances our understanding of how cells match and align during heart development but also has broader applications. Similar cell-matching processes are crucial in neuronal connections, wound repair, and facial development, where hiccups can lead to conditions like cleft lip. “Essentially, we’re putting numbers to biological processes to explain what we observe,” Saunders adds.
