After examining two large databases of traits in spouses, researchers discovered that the tendency for individuals to mate with others who possess similar traits (known as cross-trait assortative mating) is strongly linked to genetic correlation estimates and likely plays a significant role in these estimates. According to researchers, mating patterns may be able to explain many of the relationships between traits that were previously thought to be biological. A new study led by the University of California, Los Angeles suggests that current methods for evaluating the genetic connections between traits often overlook the influence of mating patterns, leading to inflated estimates of the strength of the genetic link between traits and diseases. Scientists have been using powerful genome sequencing technology to try to uncover the genetic connections between traits and disease risk in recent years, hoping that this knowledge could lead to new disease treatments. However, a study conducted by UCLA and published in the journal Science warns against relying too heavily on genetic correlation estimates, as these estimates may be distorted by non-biological factors that have not been fully taken into account. Impact of Cross-Trait Assortative Mating Genetic correlation estimates typically assume that mating is random. But in the real world, partners tend to pair up because of many shared interests and social structures. As a result, some genetic correlations in previous work that have been attributed to shared biology may instead represent incorrect statistical assumptions. For example, previous estimates of genetic overlap between body mass index (BMI) and educational attainment are likely to reflect this type of population structure, induced by “cross-trait assortative mating,” or how individuals of one trait tend to partner with individuals of another trait. The study authors said genetic correlation estimates deserve more scrutiny since these estimates have been used to predict disease risk, glean for clues for potential therapies, inform diagnostic practices, and shape arguments about human behavior and societal issues. The authors said some in the scientific community have placed too much emphasis on genetic correlation estimates based on the idea that studying genes, because they are unalterable, can overcome confounding factors. “If you just look at two traits that are elevated in a group of people, you can’t conclude that they’re there for the same reason,” said lead author Richard Border, a postdoctoral researcher in statistical genetics at UCLA. “But there’s been a kind of assumption that if you can track this back to genes, then you would have the causal story.” Findings on Spousal Traits and Psychiatric Disorders Based on their analysis of two large databases of spousal traits, researchers found that cross-trait assortative mating is strongly associated with genetic correlation estimates and plausibly accounts for a “substantial” portion of genetic correlation estimates. “Cross-trait assortative mating has affected all of our genomes and caused interesting correlations between DNA you inherit from your mother and DNA you inherit from your father across the whole genome,” said study co-author Noah Zaitlen, a professor of computational medicine and neurology at UCLA Health. The researchers also examined genetic correlation estimates of psychiatric disorders, which have sparked debate in the psychiatric community because they appear to show genetic relationships among disorders that seemingly have little similarities, such as attention-deficit hyperactivity disorder and schizophrenia. The researchers found that genetic correlations for a number of unrelated traits could be plausibly attributed to cross-trait assortative mating and imperfect diagnostic practices. On the other hand, their analysis found stronger links for some pairs of traits, like anxiety disorders and major depression, suggesting that there truly is at least some shared biology. “But even when there is a real signal there, we’re still suggesting that we’re overestimating the extent of that sharing,” Border said. A new study highlights the urgent need to stop overfishing and protect the ocean’s fastest shark as it struggles to survive in a changing climate. Shortfin makos are the fastest sharks in the sea, yet they are unable to outpace the relentless overfishing driving them toward extinction. Global demand for their meat and highly valuable fins has placed this predator on the International Union for Conservation of Nature’s (IUCN) endangered list and Appendix II of the Convention on Trade in Endangered Species of Wild Fauna and Flora (CITES). The situation for shortfin mako sharks in the Atlantic Ocean is particularly dire. Populations are currently managed as two assumed separate stocks, with fishery-based assessments indicating that Northern Atlantic mako sharks are overfished. Independent scientific surveys, using data from satellite tags deployed on shortfin makos, suggest that fishing mortality may be up to 10 times higher than previous fisheries models estimated. With extreme pressure from international fisheries, two critical questions arise: Has the genetic health and adaptive potential of shortfin makos been compromised? And is the current fisheries management strategy, based on two separate populations, supported by scientific evidence? In the U.S, fishermen may not land or retain Atlantic shortfin mako sharks Credit: Justin Gilligan | Save Our Seas Foundation Groundbreaking Genetic Research A team of scientists led by Dr Andrea Bernard and Professor Mahmood Shivji from the Save Our Seas Foundation Shark Research Center (SOSF-SRC) and Guy Harvey Institute at Nova Eastern University, USA, has published its answers in a paper in the journal Environmental Applications. The scientists have for the first time sequenced entire genomes for mitochondrial DNA and conducted high-resolution scans across the nuclear genomes of shortfin makos from nearly the entire distribution of this species in the Atlantic Ocean. These genomic assessments have discovered a potential lifeline that should add urgency to curbing overfishing. ‘Despite decades of fishing pressure, shortfin mako sharks in the Atlantic Ocean still show a (relatively) high level of genetic diversity,’ explains Professor Shivji. ‘Genetic diversity in a population is what allows species to adapt to environmental change, or to survive catastrophes.’ While overfishing is the single greatest threat to sharks worldwide, many species remain vulnerable to complex and compounding additional threats like habitat loss, deep-sea mining, pollution, and our changing climate. Credit: Justin Gilligan | Save Our Seas Foundation ‘We were rather surprised, but also pleased, to see that the genetic health of shortfin makos does not appear to have been severely compromised – yet – by the population reductions caused by overfishing,’ says Professor Shivji. ‘That means that if we can prevent further erosion of this genetic diversity in shortfin mako sharks by urgently curbing overfishing, we have more hope for this species to retain the resilience needed for its populations to adapt to our fast-changing climate and survive.’ He goes on to caution, ‘Typically, in most of the exploited shark species we study we see pretty low diversity.’ Such is the case for the critically endangered great hammerhead shark, another species being fished to the edge of existence, but whose vulnerability to being tipped into extinction is higher because it lacks the diversity to adapt to our rapidly changing climate. Credit: Sebastian Staines Genetic Insights into Mako Shark Movement The scientists also hypothesized that nomadic sharks like makos, which have been tracked making extraordinary journeys across oceans, would mix freely, hampered by few genetic barriers. And that is exactly what the research team found from the high-resolution scans made of shortfin mako nuclear DNA. Nuclear DNA is inherited from both parents, and it suggests that male shortfin mako sharks are indeed ranging across the Atlantic and spreading their genes widely. ‘Female mako sharks, which get even larger than males, are quite capable of also making these large-scale journeys,’ says Professor Shivji. ‘But when we look at the mitochondrial DNA – the genetic material inherited only from mothers – we see a contrasting picture.’ The mitochondrial genome sequences show matrilineal genetic structure for northern and southern hemisphere populations. That’s scientific-speak for the populations in each hemisphere being genetically distinct from each other. In fact, the results suggest that although female shortfin makos may well be as wide-ranging as their male counterparts, they return to key sites in one hemisphere to pup. And if we’re to protect this important genetic diversity, the management of two distinct Atlantic populations – the northern Atlantic and southern Atlantic shortfin mako sharks – is now backed by this high-resolution genetic information. Some microbes on Earth could temporarily survive on the surface of Mars, finds a new study by NASA and German Aerospace Center scientists. The researchers tested the endurance of microorganisms to Martian conditions by launching them into the Earth’s stratosphere, as it closely represents key conditions on the Red Planet. Published in Frontiers in Microbiology, this work paves the way for understanding not only the threat of microbes to space missions, but also the opportunities for resource independence from Earth. “We successfully tested a new way of exposing bacteria and fungi to Mars-like conditions by using a scientific balloon to fly our experimental equipment up to Earth’s stratosphere,” reports Marta Filipa Cortesão, joint first author of this study from the German Aerospace Center, Cologne, Germany. “Some microbes, in particular spores from the black mold fungus, were able to survive the trip, even when exposed to very high UV radiation.” Quartz disc with dried Aspergillus niger spores, before being placed in the aluminum sample carriers that went on the Trex-box. When searching for extra-terrestrial life, we need to be sure that anything we discover has not just traveled with us from Earth. “With crewed long-term missions to Mars, we need to know how human-associated microorganisms would survive on the Red Planet, as some may pose a health risk to astronauts,” says joint first author Katharina Siems, also based at the German Aerospace Center. “In addition, some microbes could be invaluable for space exploration. Credit: German Aerospace Center (DLR) Mars in a Box Many key characteristics of the environment at the Martian surface cannot be found or easily replicated at the surface of our planet; however, above the ozone layer in Earth’s middle stratosphere, the conditions are remarkably similar. “We launched the microbes into the stratosphere inside the MARSBOx (Microbes in Atmosphere for Radiation, Survival and Biological Outcomes experiment) payload, which was kept at Martian pressure and filled with artificial Martian atmosphere throughout the mission,” explains Cortesão. “The box carried two sample layers, with the bottom layer shielded from radiation. This allowed us to separate the effects of radiation from the other tested conditions: desiccation, atmosphere, and temperature fluctuation during the flight. The top layer samples were exposed to more than a thousand times more UV radiation than levels that can cause sunburn on our skin.” “While not all the microbes survived the trip, one previously detected on the International Space Station, the black mold Aspergillus niger, could be revived after it returned home,” explains Siems, who highlights the importance of this ongoing research. “Microorganisms are closely connected to us; our body, our food, our environment, so it is impossible to rule them out of space travel.
