A team of researchers has discovered a method to activate a bacterial defense system, known as CBASS, to self-destruct and prevent the spread of viruses among bacteria, potentially offering a new way to manage bacterial infections and combat antibiotic resistance. Credit: SciTechDaily.com Researchers unveil how the self-killing activity of bacteria can be harnessed in the fight against antibiotic resistance. Scientists at the Icahn School of Medicine at Mount Sinai have identified a new approach to controlling bacterial infections. The findings were described in the February 6 online issue of Nature Structural & Molecular Biology. The team found a way to turn on a vital bacterial defense mechanism to fight and manage bacterial infections. The defense system, called cyclic oligonucleotide-based antiphage signaling system (CBASS), is a natural mechanism used by certain bacteria to protect themselves from viral attacks. Bacteria self-destruct as a means to prevent the spread of virus to other bacterial cells in the population. CBASS Defense Mechanism Explored “We wanted to see how the bacterial self-killing CBASS system is activated and whether it can be leveraged to limit bacterial infections,” says co-senior author Aneel Aggarwal, PhD, Professor of Pharmacological Sciences at Icahn Mount Sinai. “This is a fresh approach to tackling bacterial infections, a significant concern in hospitals and other settings. It’s essential to find new tools for fighting antibiotic resistance. In the war against superbugs, we need to constantly innovate and expand our toolkit to stay ahead of evolving drug resistance.” According to a 2019 report by the Centers for Disease Control and Prevention, more than 2.8 million antimicrobial-resistant infections occur in the United States each year, with over 35,000 people dying as a result. Icahn Mount Sinai researchers unveil how the self-killing activity of bacteria can be used in the fight against antibiotic resistance. Credit: Rechkoblit et al., Nature Structural & Molecular Biology Innovative Strategies Against Superbugs As part of the experiments, the researchers studied how “Cap5,” or CBASS-associated protein 5, is activated for DNA degradation and how it could be used to control bacterial infections through a combination of structural analysis and various biophysical, biochemical, and cellular assays. Cap5 is a key protein that becomes activated by cyclic nucleotides (small signaling molecules) to destroy the bacterial cell’s own DNA. “In our study, we started by identifying which of the many cyclic nucleotides could activate the effector Cap5 of the CBASS system,” says co-senior author Olga Rechkoblit, PhD, Assistant Professor of Pharmacological Sciences at Icahn Mount Sinai. “Once we figured that out, we looked closely at the structure of Cap5 when it’s bound to these small signaling molecules. Then, with expert help from Daniela Sciaky, PhD, a researcher at Icahn Mount Sinai, we showed that by adding these special molecules to the bacteria’s environment, these molecules could potentially be used to eliminate the bacteria.” Overcoming Technical Challenges The researchers found that determining the structure of Cap5 with cyclic nucleotides posed a technical challenge, requiring expert help from Dale F. Kreitler, PhD, AMX Beamline Scientist at Brookhaven National Laboratory. It was achieved by using micro-focused synchrotron X-ray radiation at the same facility. Micro-focused synchrotron X-ray radiation is a type of X-ray radiation that is not only produced using a specific type of particle accelerator (synchrotron) but is also carefully concentrated or focused on a tiny area for more detailed imaging or analysis. Future Directions Next, the researchers will explore how their discoveries apply to other types of bacteria and assess whether their method can be used to manage infections caused by various harmful bacteria. Hydrothermal vents are located globally at the boundaries of shifting tectonic plates, with many fields yet to be discovered. In a 2022 expedition aboard the MARIA S. MERIAN, researchers identified the first hydrothermal vent field along the 500-kilometer Knipovich Ridge near Svalbard. Led by Prof. Dr. Gerhard Bohrmann from MARUM – Center for Marine Environmental Sciences and the University of Bremen’s Geosciences department, the international team, including scientists from Bremen and Norway, detailed their findings in the journal Scientific Reports. Hydrothermal vents are seeps on the sea floor from which hot liquids escape. “Water penetrates into the ocean floor where it is heated by magma. The overheated water then rises back to the sea floor through cracks and fissures. On its way up the fluid becomes enriched in minerals and materials dissolved out of the oceanic crustal rocks. These fluids often seep out again at the sea floor through tube-like chimneys called black smokers, where metal-rich minerals are then precipitated,” explains Prof. Gerhard Bohrmann of MARUM and chief scientist of the MARIA S. MERIAN (MSM 109) expedition. At water depths greater than 3,000 meters, the remote-controlled submersible vehicle MARUM-QUEST took samples from the newly discovered hydrothermal field. Named after Jøtul, a giant in Nordic mythology, the field is located on the 500-kilometer-long Knipovich Ridge. The ridge lies within the triangle formed by Greenland, Norway, and Svalbard on the boundary of the North American and European tectonic plates. Among the numerous hydrothermal mounds of the Jøtul field is the Nidhogg spring, named after a serpent-like dragon in Norse mythology that lives on the world tree Yggdrasil. The fluids with temperatures of 40 to 50 degrees Celsius at Nidhogg lead to the precipitation of barite and amorphous opal, and the numerous amphipods, particularly like these temperatures. The Jøtul Field is located on an extremely slow-spreading ridge with a growth rate of the plates of less than two centimeters per year. Because very little is known about hydrothermal activity on slow-spreading ridges, the expedition focused on obtaining an overview of the escaping fluids, as well as the size and composition of active and inactive smokers in the field. Climate Impact and Methane Emissions “The Jøtul Field is a discovery of scientific interest not only because of its location in the ocean but also due to its climate significance, which was revealed by our detection of very high concentrations of methane in the fluid samples, among other things,” reports Gerhard Bohrmann. Methane emissions from hydrothermal vents indicate a vigorous interaction of magma with sediments. On its journey through the water column, a large proportion of the methane is converted into carbon dioxide, which increases the concentration of CO2 in the ocean and contributes to acidification, but it also has an impact on climate when it interacts with the atmosphere. The most spectacular hydrothermal vent of the MSM109 expedition featured several chimneys and vents, and the outflowing fluid shimmered around it. Credit: MARUM – Center for Marine Environmental Sciences, University of Bremen The amount of methane from the Jøtul Field that eventually escapes directly into the atmosphere, where it then acts as a greenhouse gas, still needs to be studied in more detail. There is also little known about the organisms living chemosynthetically in the Jøtul Field. In the darkness of the deep ocean, where photosynthesis cannot occur, hydrothermal fluids form the basis for chemosynthesis, which is employed by very specific organisms in symbiosis with bacteria. In order to significantly expand on the somewhat sparse information available on the Jøtul Field, a new expedition of the MARIA S. MERIAN will start in late summer of this year under the leadership of Gerhard Bohrmann. The focus of the expedition is the exploration and sampling of as-yet-unknown areas of the Jøtul Field. With extensive data from the Jøtul Field, it will be possible to make comparisons with the few already known hydrothermal fields in the Arctic province, such as the Aurora Field and Loki’s Castle. His work, explored today as part of chronobiology, examined daily and seasonal cycles, contributing to our understanding of nature’s temporal patterns. A thorough examination of Darwin’s writings reveals a profound interest in cyclical events in nature. Rhythmic phenomena, now studied in the field of chronobiology, have been a focus of scientific inquiry since at least the 18th century. In a perspective, Tiago Gomes de Andrade and Andrew D. Beale examined the writings and work of Charles Darwin to explore and share the eminent naturalist’s deep fascination with biological rhythms. Darwin’s work on the “sleep movements” in plants, published in 1880 with his son Francis is well known. This work examined the daily cycle of opening and closing of leaves. But as far back as 1838, Darwin was taking notes on whether plants that close their leaves in response to touch might also show daily rhythms. Observations of Biological Rhythms Throughout His Career Throughout his career, he took note of seasonal and daily biological rhythms, including diurnal and nocturnal habits in animals, seasonal migrations, cyclical changes associated with reproduction, hibernation patterns, and tidal rhythms, among other temporal patterns. Darwin recognized the heritability and adaptive nature of many of these rhythms, from the timing of mating seasons to correspond with peak vigor to the evening timing of plant perfume release to communicate with nocturnal pollinators. According to the authors, Darwin’s observations and experiments show a profound engagement in what is now termed chronobiology.
