Scientists discovered that mice display strategic behavior in learning tasks by engaging in exploratory actions that initially appear as mistakes. Through experiments, the study showed that mice test hypotheses and adjust their strategies based on the outcomes, challenging the traditional view of animal errors as mere mistakes. This insight into animal cognition not only sheds light on how mice think but also draws parallels with nonverbal human learning, paving the way for further studies on the neural basis of strategic thinking. Credit: SciTechDaily.com Behavior that is “surprisingly strategic” enhances our understanding of animal cognition. Are mice clever enough to be strategic? Kishore Kuchibhotla, a Johns Hopkins University neuroscientist who studies learning in humans and animals, and who has long worked with mice, wondered why rodents often performed poorly in tests when they knew how to perform well. With a simple experiment, and by acting as “a little bit of a mouse psychologist,” he and his team figured it out. “It appears that a big part of this gap between knowledge and performance is that the animal is engaging in a form of exploration—what the animal is doing is very clever,” he said. “It’s hard to say animals are making hypotheses, but our view is that animals, like humans, can make hypotheses and they can test them and may use higher cognitive processes to do it.” The work, which deepens our understanding of animal cognition, and could lead to identifying the neural basis for strategizing, was published in Current Biology. Kuchibhotla’s lab previously found that animals know a lot more about tasks than they demonstrate in tests. The team had two theories about what could be behind this gap. Either the mice were making mistakes because they were stressed, or they were doing something more purposeful: exploring and testing their knowledge. Experiment Insights To figure it out Kuchibhotla and Ziyi Zhu, a graduate student studying neuroscience, came up with a new experiment. Mice heard two sounds. For one sound they were supposed to turn a wheel to the left. For the other sound, they’d turn the wheel to the right. When the mice performed correctly they were rewarded. The researchers observed how upon hearing either sound over consecutive trials, the mice would turn the wheel left for a bit, then switch to turning it right, seemingly making mistakes but actually being purposeful. Credit: Johns Hopkins University Strategic Behavior in Mice “We find that when the animal is exploring, they engage in a really simple strategy, which is, ‘I’m going to go left for a while, figure things out, and then I’m going to switch and go right for a while,’” Kuchibhotla said. “Mice are more strategic than some might believe.” Zhu added, “Errors during animal learning are often considered as mistakes. Our work brings new insight that not all errors are the same.” The team learned even more about the rodents’ actions by taking the reward out of the equation. When a mouse performed correctly and wasn’t rewarded, it immediately doubled down on the correct response when retested. “If the animal has an internal model of the task, the lack of reward should violate its expectation. And if that’s the case, it should affect the behavior on subsequent trials. And that’s exactly what we found. On subsequent trials the animal just does a lot better,” Kuchibhotla said. “The animal is like, ‘Hey, I was expecting to be rewarded, I wasn’t, so let me test my knowledge, let me use the knowledge I have and see if it’s correct.’” If the animal didn’t have an internal model of the task, there would be no expectations to violate and the mice would keep performing poorly. “At a very early time in learning the animal has an expectation and when we violate it, it changes its strategy,” Kuchibhotla said. “It was surprisingly strategic.” This mouse strategizing is comparable to how nonverbal human babies learn. Both are highly exploratory and both may test hypotheses in various ways, Kuchibhotla said. During the experiments Kuchibhotla said he became “a little bit of a mouse psychologist” to interpret their behavior. Like working with a nonverbal infant, he and Zhu had to infer the underlying mental processes from the behavior alone. “That’s what was really fun in this project, trying to figure out what the mouse is thinking,” he said. “You have to think about it from the perspective of the animal.” Next the team hopes to determine the neural basis for strategic thinking, and how those strategies might compare across different animals. Credit: Lauren Brent A new study explores how Hurricane Maria’s devastation led to significant changes in the social behavior of rhesus macaques on Puerto Rico’s Monkey Island. By destroying much of their habitat, the hurricane created a situation where macaques that were more tolerant and less aggressive had a survival advantage due to their increased ability to share scarce resources like shade. New research reveals that a devastating hurricane transformed a monkey society by shifting the balance of advantages and disadvantages in social interactions. Hurricane Maria hit Puerto Rico in 2017, killing more than 3,000 people. It also destroyed 63% of vegetation on Cayo Santiago (also known as Monkey Island), which is home to a population of rhesus macaques. Even now, tree cover remains far below pre-hurricane levels and – in this hot part of the world – that makes shade a scarce and precious resource for the macaques. The new study, led by the universities of Pennsylvania and Exeter and published in the journal Science, shows the storm damage altered the evolutionary benefits of tolerating others and sharing shade – giving a survival advantage to tolerant macaques. With many ecosystems changing rapidly due to human activity, this study is an example of a species adapting its social strategy to survive. Impact on Macaque Behavior “In response to the drastic changes caused by the hurricane, macaques persistently increased tolerance and decreased aggression towards each other,” said Dr Camille Testard, from the University of Pennsylvania. “This enabled more macaques to access scarce shade, which is critical for survival.” Dr Testard added: “We examined 10 years of data on the strength and number of macaques’ social ties, before and after the hurricane. Before the hurricane, tolerating others had no impact on the risk of death. Afterward, macaques that displayed more than average social tolerance – and were, therefore, better able to share shade – were 42% less likely to die than those that were less tolerant.” This represents a sudden change in “selection pressure” – the evolutionary benefits or costs of different traits or behaviors. Social behavior was assessed by recording aggression and how often individuals were seen sitting together. Broader Implications and Conclusion “Macaques aren’t the best at sharing resources – be they food or shade. They are well known to live in an aggressive, highly competitive society,” said Professor Lauren Brent, from the University of Exeter. “But in the heat caused by ecological changes, often around 40C, the macaques had to share space or die.” Dr Testard added: “To access shade, they need to tolerate – and be tolerated by – others, and we found that this tolerance spills over into other daily interactions. Macaques that began sharing shade also spend time together in the mornings, before the day’s heat forces them to seek shade. In effect, the hurricane changed the rules of the game in the monkeys’ society.” Professor Brent added: “For group-living animals, social relationships may allow them to cope with disturbances in the environment, including human-induced climate change. Credit: Johanna Spandl / University of Bonn The Study Offers the First Precise Understanding of Crucial Remodeling Processes in Adipose Tissue Fat cells utilize fat molecules as a means of energy storage. These molecules are comprised of three fatty acids attached to a glycerol backbone, and are commonly referred to as triglycerides. It has been long believed that these molecules undergo constant change during storage, being regularly broken down and reconstructed – a process known as “triglyceride cycling.” But is this assumption true, and if so, what is the purpose of this process? “Until now, there has been no real answer to these questions,” explains Prof. Dr. Christoph Thiele of the LIMES Institute at the University of Bonn. “It’s true that there has been indirect evidence of this permanent reconstruction for the past 50 years. However, direct evidence of this has so far been lacking.” The problem: To prove that triglycerides are broken down, and fatty acids modified and reincorporated into new molecules, one would need to track their transformation as they travel through the body. Yet there are thousands of different forms of triglycerides in each cell. Keeping track of individual fatty acids is therefore extremely difficult. Label Makes Fatty Acids Unmistakable “However, we have developed a method that allows us to attach a special label to fatty acids, making them unmistakable,” says Thiele. His research group labeled various fatty acids in this way and added them in a nutrient medium to mouse fat cells. The mouse cells then incorporated the labeled molecules into triglycerides. “We were able to show that these triglycerides do not remain unchanged, but are continuously degraded and remodeled: Each fatty acid is split off about twice a day and reattached to another fat molecule,” the researcher explains. But why is that? After all, this conversion costs energy, which is released as waste heat – what does the cell get out of it? Until now, it was thought that the cell needed this process to balance energy storage and supply. Or perhaps it is simply a way for the body to generate heat. “Our results now point to a completely different explanation,” Thiele explains. “It’s possible that in the course of this process, the fats are converted to what the body needs.” Poorly utilizable fatty acids would consequently be refined into higher-quality variants and stored in this form until they are needed. Fatty acids consist largely of carbon atoms, which hang one behind the other like the carriages of a train. Their length can be very different: Some consist of only ten carbon atoms, others of 16 or even more. In their study, the researchers produced three different fatty acids and labeled them. One of them was eleven, the second 16, and the third 18 carbon atoms long. “These chain lengths are typically found in food as well,” Thiele explains. Short Fatty Acids Are Eliminated, Long Ones “Improved” Labeling allowed the researchers to track exactly what happens to the fatty acids of different lengths in the cell. This showed that the fatty acids consisting of eleven carbon atoms were initially incorporated into triglycerides. After a short time, however, they were split off again and channeled out of the cell. After two days, they were no longer detectable. “Such shorter fatty acids are poorly usable by cells and can even damage them,” says Thiele, who is also a member of the Cluster of Excellence ImmunoSensation2. “Therefore, they are disposed of quickly.” In contrast, the 16- and 18-atom fatty acids remained in the cell, although not in their original fat molecules. They were also gradually chemically modified, for example by additional carbon atoms being inserted. In the original fatty acids, the carbon atoms were moreover linked with single bonds – roughly like a human chain in which neighbors join hands. Over time, this sometimes developed into double bonds – as if revelers at a party were doing a conga. The fatty acids that are formed in this process are called unsaturated. They are better utilizable for the body. “Overall, in this way the cells produce fatty acids that are more beneficial to the organism than those that we had originally supplied with the nutrient solution,” Thiele emphasizes. In the long term, this results for instance in the formation of oleic acid, a component of high-quality olive oil, from palmitate, such as that contained in palm fat. However, the cell cannot change the fatty acids as long as they are inside the fat molecule. They must first be split off, then modified, and finally tacked back on. Thiele: “Without triglyceride cycling, there is also no fatty acid modification.” Adipose tissue can therefore improve triglycerides. If we eat and store food with unfavorable fatty acids, they do not have to be released in that state again when we are hungry. What we get back contains fewer “short” fatty acids, more oleic acid (instead of palmitate), and more of the important arachidonic acid (instead of linoleic acid). “Nevertheless, we should take care in our diet to consume high-quality dietary fats as much as possible,” the researcher stresses. Because the refinement never works 100 percent. In addition, some of the fatty acids are not stored but used directly in the body. In the next step, the researchers now want to test whether the same processes occur in human adipose tissue as in individual mouse fat cells in the test tube. They also want to find out which enzymes make cycling work.
