A groundbreaking study on mulberry fruit genetics has uncovered key mechanisms controlling anthocyanin content, crucial for color and nutritional quality. By analyzing two mulberry cultivars, researchers identified genetic variations and the role of MaVHAG3 in anthocyanin accumulation, providing a foundation for enhancing fruit traits. Researchers have uncovered the genetic basis of anthocyanin regulation in mulberries, identifying key genes that could help improve fruit color, nutritional value, and market appeal. A recent study has uncovered the genetic mechanisms behind anthocyanin content in mulberries, shedding light on the factors that control fruit color and nutritional value. By sequencing and comparing the genomes of two different mulberry cultivars, researchers identified crucial genetic variations and highlighted the significant role of MaVHAG3 in anthocyanin production. These findings provide a valuable biotechnological framework for improving mulberry fruit traits. Mulberry fruits are celebrated for their vibrant colors, nutritional value, and potential health benefits. Anthocyanins, the primary pigments in purple mulberries, are known for their antioxidant properties and role in human health. However, understanding the genetic basis of anthocyanin regulation in mulberry fruits has been challenging due to their complex genomes. Based on these challenges, there is a pressing need for in-depth research to better understand and manipulate mulberry fruit pigmentation. Genome Analysis of Mulberry Cultivars On April 23, 2024, researchers from Southwest University in Chongqing, China, published a pivotal study in Horticulture Research. The team generated haplotype-resolved genome assemblies for two mulberry cultivars, ‘Zhongsang5801’ (high anthocyanin) and ‘Zhenzhubai’ (low anthocyanin). Their comprehensive analysis identified key genes and regulatory mechanisms underlying anthocyanin content in mulberries, paving the way for genetic enhancement of mulberry cultivars. Transcriptional and metabolic patterns associated with fruit color. Credit: Horticulture Research The study involved sequencing the genomes of ‘Zhongsang5801’ (ZS5801) and ‘Zhenzhubai’ (ZZB), followed by a detailed comparative analysis of their genomic and transcriptomic data. The researchers identified MaVHAG3, a vacuolar-type H+-ATPase G3 subunit gene, as crucial in anthocyanin accumulation. By comparing gene expression across developmental stages, they found that ZS5801 exhibited significantly higher levels of anthocyanins and flavonoids compared to ZZB. The analysis also revealed expansions and contractions in flavonol synthase (FLS) and dihydroflavonol 4-reductase (DFR) genes, which impact carbon flow in anthocyanin biosynthesis pathways. These findings highlight the intricate genetic and molecular processes that regulate fruit coloration in mulberries. Dr. Aichun Zhao, the corresponding author, commented, “Our research provides a comprehensive understanding of the genetic and molecular mechanisms controlling anthocyanin content in mulberries. These insights are critical for developing new cultivars with enhanced nutritional and aesthetic qualities. The high-quality genome assemblies we generated will serve as a valuable resource for future research and breeding programs.” The findings from this study have significant implications for the agricultural and food industries. By understanding the genetic basis of anthocyanin regulation, breeders can develop mulberry cultivars with higher anthocyanin content, leading to fruits with better nutritional profiles and health benefits. Additionally, the enhanced pigmentation traits can improve the market appeal of mulberry products. This research also sets the stage for further studies on other fruit crops, potentially leading to broader applications in horticulture and biotechnology. DOI: 10.1093/hr/uhae120 Neuroscientists find they can predict how well someone can see based on the unique structure of their primary visual cortex. Visual Cortex Size, Brain Tissue Can Predict How Well We See In many ways, the eye functions as a camera, with the retina acting as the photographic film (or CCD in a digital camera.) However, you don’t actually see anything without your brain, which receives the visual signals from the eye via the optic nerve. The primary cortical region of the brain that receives, integrates, and processes visual information transmitted from the retinas is known as the visual cortex. It is located in the occipital lobe of the primary cerebral cortex, which is in the most posterior region of the brain. The visual cortex is divided into five different areas (V1 to V5) based on function and structure, with V1 being the primary visual cortex. Link Between Brain Structure and Visual Ability The size of our primary visual cortex and the amount of brain tissue we have dedicated to processing visual information at certain locations of visual space can predict how well we can see, a team of neuroscientists has discovered. Its study, which appears today (June 13, 2022) in the journal Nature Communications, reveals a new link between brain structure and behavior. “We have found that we can predict how well someone can see based on the unique structure of their primary visual cortex,” explains lead author Marc Himmelberg, a postdoctoral researcher in New York University’s Center for Neural Science and Department of Psychology. “By showing that individual variation in the structure of the human visual brain is linked to variation in visual functioning, we can better understand what underlies differences in how people perceive and interact with their visual environment.” Unique Brain Structures Influence Vision Quality As with fingerprints, the bumps and grooves on each person’s brain surface are unique. However, the significance of these differences is not fully understood, especially when it comes to their impact on behavior, such as distinctions in our ability to see. In the study published in Nature Communications, Himmelberg and his co-authors, Jonathan Winawer and Marisa Carrasco, professors in NYU’s Center for Neural Science and Department of Psychology, sought to illuminate the relevance of these brain traits to how we see. The primary visual cortex (V1) is arranged into a map of the image projected from the eye. But like many kinds of maps, it is distorted, with some parts of the image enlarged compared to others. “Think of a subway map of New York City which makes Staten Island look smaller than Manhattan,” explains Winawer. “The map maintains some degree of accuracy, but it enlarges regions likely to be of broader interest. Similarly, V1 enlarges the center of the image we see—that is, where our eyes are fixating—relative to the periphery.” This is because V1 has more tissue dedicated to the center of our field of view. Likewise, V1 also enlarges locations to the left and right of where our eyes are fixating relative to locations above or below, again because of differences in the arrangement of cortical tissue. Mapping the Visual Cortex with fMRI Using functional magnetic resonance imaging (fMRI), the scientists mapped the primary visual cortex (or “V1”) size of more than two dozen humans. The researchers also measured the quantity of V1 tissue these individuals have dedicated to processing visual information from different locations in their field of view—locations to the left, right, above, and below fixation. These participants also undertook a task designed to assess the quality of their vision at the same locations in their field of view as the V1 measurements. The participants discriminated among the orientation of patterns shown on a computer screen, which were used to gauge “contrast sensitivity,” or the ability to make distinctions among images. V1 Structure Predicts Visual Sensitivity Their results showed that differences in V1 surface area could predict measurements of people’s contrast sensitivity. First, people with a large V1 had better overall contrast sensitivity than those with a small V1 (the largest surface area being 1,776 square millimeters [mm2] and the smallest being 832 mm2). Second, people whose V1 had more cortical tissue processing visual information from a specific region in their field of view had higher contrast sensitivity at that region relative to those with less cortical tissue dedicated to the same region. Third, across participants, higher contrast sensitivity at a specific location (e.g., left) than at another location equidistant from fixation (e.g., above) corresponded to regions with more or less cortical tissue, respectively. “In sum, the more local V1 surface area dedicated to encoding a specific location, the better the vision at that location,” concludes Carrasco. “Our findings show differences in visual perception are inextricably linked to differences in the structure of the primary visual cortex in the brain.” Reference: “Linking individual differences in human primary visual cortex to contrast sensitivity around the visual field” by Marc M. Himmelberg, Jonathan Winawer and Marisa Carrasco, 13 June 2022, Nature Communications. DOI: 10.1038/s41467-022-31041-9 The research was supported by a grant from the National Institutes of Health (R01-EY027401). Shiikuwasha have an important commercial value in Okinawa and are used to create many different products. Within Okinawa, Oogimi and Katsuyama are the biggest citrus productive area in Okinawa. This image shows products from Katsuyama. Credit: Katsuyama Shiikuwasha Co., Ltd. Citrus fruits from the mandarin family have important commercial value but how their diversity arose has been something of a mystery Researchers analyzed the genomes of the East Asian varieties and found a second center of diversity in the Ryukyu Islands along with the previously known center in the mountains of southern China They discovered a new citrus species native to Okinawa that arose about two million years ago when the Ryukyu archipelago became disconnected from mainland Asia Other citrus from Okinawa and mainland Japan, including shiikuwasha and tachibana, are hybrids of this newly discovered wild species with different mainland Asian varieties This research may have commercial implications and opens the potential to create additional hybrids with favorable traits Citrus fruits from the mandarin family are popular throughout the world for their tasty and healthy characteristics. Within Japan, the tiny shiikuwasha and the ornamental tachibana are of special cultural and historical importance. However, the origin of these two varieties, and other East Asian citrus, was always something of a mystery, until now. In a new study, published in Nature Communications, scientists from the Okinawa Institute of Science and Technology Graduate University (OIST), and collaborators from other institutes, analyzed 69 genomes from the East Asian mandarin family, alongside their mainland Asian relatives, to reveal a far-ranging story of isolation, long-distance travel, and hybridization. Shiikuwasha have an important commercial value in Okinawa and are used to create many different products. Within Okinawa, Oogimi and Katsuyama are the biggest citrus productive area in Okinawa. This image shows products from Oogimi. Credit: Hidekazu Sumi The story starts in the Hunan Province of southern China, which is the center of wild mandarin diversity and the genetic source of the most well-known mandarins. When the scientists re-analyzed previously published genomic data, they unexpectedly found that wild mandarins of this mountainous region are split into two subspecies. “We found that one of these mandarin subspecies can produce offspring that are genetically identical to the mother,” said Dr. Guohong Albert Wu, a research collaborator at the Lawrence Berkeley National Laboratory in California. “Like many other plants, wild citrus typically reproduces when the pollen of the father combines with the egg of the mother, mixing the genes from both parents in the seed. But we found a subspecies of wild mandarins from Mangshan, in southern China, where the seed contains an identical copy of the mother’s DNA without any input from a father. So, the seed grows to be a clone of the mother tree.” A shiikuwasha flower photographed in Ōgimi, Okinawa. The researchers found that this well-known plant is both a hybrid and a clone. Credit: Dr. Chikatoshi Sugimoto Back in Okinawa, the researchers looked more carefully at a strange shiikuwasha-like citrus that produces small, acidic fruit and had been ignored by local farmers since it has little commercial value. To their surprise, they found that this strange citrus represented a previously undescribed species, which they named the Ryukyu mandarin or, more formally, Citrus ryukyuensis. And in contrast to the well-known shiikuwasha, which reproduces clonally (like the subspecies in Mangshan), the new species always reproduces sexually. Remarkably, the researchers found that all shiikuwasha are hybrids of a very specific type — one parent is from the local Ryukyuan species and the other, from mainland Asia. Surprisingly, all shiikuwasha have the same mainland mandarin parent, meaning that all shiikuwasha are half-siblings. They concluded that tens of thousands of years ago a mainland Asian mandarin was transported, either by people or by natural methods, to the land that would become the Ryukyu Islands. There it mated with the native Ryukyu citrus. The researchers traced the ancestry of this mainland Asian mandarin back to Mangshan, where it acquired its ability to reproduce asexually. This ability was passed on to its children. Thus, all the shiikuwasha varieties found in Okinawa’s markets today are descended from this mating, and reproduce asexually, allowing stable varieties like golden shiikuwasha to be propagated from generation to generation. And what of tachibana and the other East Asian mandarin variations? “They’re all hybrids!” explained Dr. Chikatoshi Sugimoto, Postdoctoral Scholar in OIST’s Molecular Genetics Unit. “The tachibana lineage also seems to have descended from the newly described Ryukyu species and another mandarin from China, but its birthplace was probably what is now mainland Japan.” Once they saw the genetic pattern in shiikuwasha and tachibana, the researchers also recognized another half-sibling family comprising various traditional Ryukyuan types — oto, kabuchii, tarogayo, and other unnamed citrus. This family, which the researchers called ‘yukunibu’ (sour citrus in the native Okinawan language), is much younger than shiikuwasha and tachibana. It arose when the famous kunenbo — also the father of satsuma mandarins — hybridized with the native Ryukyu mandarin. Kunenbo was brought to Okinawa from Indochina around 4-500 years ago by maritime trade. Like the mainland parents of shiikuwasha and tachibana, it was also able to clone itself by seeds, due to its distant Mangshan ancestry, and it passed this trait on to its children. “It’s fascinating to puzzle out the story of mandarin diversification and its relationship to the biogeography of the region,” concluded Prof. Dan Rokhsar, Principal Investigator of OIST’s Molecular Genetics Unit. “But it also could have commercial value. What other possible hybrid types are there? Could we create new hybrids that are more resilient to disease or drought, or have other desirable characteristics? By looking into the past, we can create all sorts of possibilities for the future.” Reference: “Diversification of mandarin citrus by hybrid speciation and apomixis” by Guohong Albert Wu, Chikatoshi Sugimoto, Hideyasu Kinjo, Chika Azama, Fumimasa Mitsube, Manuel Talon, Frederick G. Gmitter Jr. and Daniel S. Rokhsar, 26 July 2021, Nature Communications. DOI: 10.1038/s41467-021-24653-0 To unravel this diversity, the researchers worked closely with industry and individuals in Okinawa, including Okinawa Prefectural Agricultural Research Center, Nago Branch, Katsuyama Shiikuwasha, and local farmer, Hiroshi Kobashigawa.
