Credit: © UNIGE/HAMEL Research teams from UNIGE have discovered that the cytoskeleton of the malaria parasite comprises a vestigial form of an organelle called conoid, initially thought to be absent from this species and which could play a role in host invasion. Plasmodium is the parasite causing malaria, one of the deadliest parasitic diseases. The parasite requires two hosts — the Anopheles mosquito and the human — to complete its life cycle and goes through different forms at each stage of its life cycle. Transitioning from one form to the next involves a massive reorganization of the cytoskeleton. Two teams from the University of Geneva (UNIGE) have shed new light on the cytoskeleton organization in Plasmodium. Their research, published in PLOS Biology, details the organization of the parasite’s skeleton at an unprecedented scale, adapting a recently developed technique called expansion microscopy. Cells are “inflated” before imaging, providing access to more structural details, at a nanometric scale. The study identifies traces of an organelle called “conoid,” which was thought to be lacking in this species despite its crucial role in host invasion of closely related parasites. The cytoskeleton, or cell skeleton, consists of a network of several types of filaments, including actin and tubulin. It confers rigidity to the cell, allows the attachment or movement of organelles and molecules inside the cell, as well as cell deformations. As the parasite transitions between developmental stages, its cytoskeleton undergoes repeated, drastic, reorganizations. In particular, Plasmodium needs a very specific cytoskeleton in order to move and penetrate the membrane barriers of its host cells, two processes that are central to the pathogenesis of malaria-causing parasites. “Due to the very small size of Plasmodium — up to 50 times smaller than a human cell — it is a technical challenge to view its cytoskeleton!” begins Eloïse Bertiaux, a researcher at UNIGE and the first author of the study. “That is why we adapted our expansion microscopy protocol, which consists of inflating the biological sample while keeping its original shape, so it can be observed at a resolution that has never been attained before,” continues Virginie Hamel, a researcher at the Department of Cell Biology of the Faculty of Sciences of UNIGE and co-leading the study. A Vestigial Form of an Organelle The researchers observed the parasite at the ookinete stage, the form responsible for the invasion of the mosquito midgut, an essential step for the dissemination of malaria. A structure made of tubulin was visible at the tip of the parasite. This structure is similar to a conoid, an organelle involved in host cell invasion, in related Apicomplexa parasites. “The structure observed in Plasmodium seems, however, divergent and reduced compared with the well-described conoid of Toxoplasma, the parasite causing toxoplasmosis. We still need to determine whether this remnant conoid is also important for host cell invasion of Plasmodium,” explains Mathieu Brochet, a professor at the Department of Microbiology and Molecular Medicine of the Faculty of Medicine of UNIGE. Cytoskeleton Under the Microscope The discovery of this vestigial conoid highlights the power of expansion microscopy, which can be used to view cytoskeletal structures at the nanoscale without the need for specialised microscopes. Used in combination with electron microscopy and super-resolution microscopy approaches, this method adds molecular details to the available structural information, paving the way for more in-depth studies of the cytoskeleton and its molecular organization. This will allow us to gain a better understanding of how Plasmodium invades its host cells, a process that is essential for the pathogenesis of this parasite. Credit: Anthony Waddle A novel conservation method employing “frog saunas” helps green and golden bell frogs fight off the lethal chytrid fungus by raising their body temperatures. Successful trials in both controlled and natural environments suggest this technique could significantly aid in conserving this species. Impact of Chytrid Fungus on Amphibians As global diversity continues to decline due to a range of factors, amphibians have been particularly hard hit by infectious diseases. A pernicious fungal disease called chytridiomycosis, more commonly shortened to “chytrid,” has led to the extinction of 90 species of amphibians and has pushed another 500 species into decline. New research, though, offers hope in the form of “frog saunas.” Imagine not a traditional wood sauna beloved by Finns and Estonians, but rather a large, sun-heated brick structure with cubby holes where frogs can find refuge from colder weather. During winter, chytrid hits frogs harder, and the researchers speculated that if green and golden bell frogs were given an opportunity to get warmer in artificial saunas, they would take it. It turns out not only would they take the opportunity, raising their body temperature helped them clear the infection as well. Frog Saunas: A Novel Conservation Strategy Experiments were carried out in both lab and natural settings, where the frogs had both shaded and unshaded options and no inducement to access the saunas. But they took to the saunas like Finns in winter. The researchers also found that frogs that were initially infected and then fought off the infection after spending time in the sauna were significantly less likely to be reinfected. The paper, “Hotspot shelters stimulate frog resistance to chytridiomycosis,” was published in Nature. Of the 14 people listed as co-authors, 12 hail from Australia, where the research was conducted. The lone North American co-author is Erin Sauer, a postdoctoral fellow in biological sciences at the University of Arkansas. Development and Impact of Thermal Gradients In fact, the team’s work on frog saunas was inspired by Sauer’s previous research into how amphibian thermoregulatory behavior influences resistance to chytrid. For that study, done in 2018, Sauer designed thermal gradients she could keep frogs in for longer periods of time while measuring their thermal preferences. In practical terms, this meant putting a Plexiglas lid on sections of aluminum gutter that had a heating element on one end and cooling element on the other, providing a range of temperatures in between the two ends. She ultimately found that species that preferred warmer temperatures were able to clear their infections when they hung out on the warmer parts of the thermal gradients. “Anthony Waddle, the lead author on the Nature paper, got in touch with me because he wanted to build my thermal gradients in Australia and replicate my study with green and golden bell frogs,” Sauer explained. “So, I helped him design his setup and experiment in 2020-2021, and then helped with the data analysis for the lab study. “Very similar to my 2018 study, we found that the green and golden bell frogs preferred warm temperatures and were able to better resist their chytrid infections at those warm temperatures. Anthony and his colleagues in Australia then applied the findings from the lab experiment to build the frog saunas and run the mesocosm experiment.” Limitations and Applications of Amphibian Thermal Therapy While the findings suggest that the general public can play a role in saving green and golden bell frogs from extinction by building low-cost saunas in gardens or wetlands, Sauer cautions that this will not help all species of frogs. “My research has shown that while warm-adapted species have better disease outcomes at warm temperatures, cold-adapted species actually have worse outcomes at warm temperatures,” Sauer said. “My colleagues and I have termed this phenomenon the ‘thermal mismatch hypothesis’ and did a global study testing our hypothesis across systems a few years ago that was published in Science. DOI: 10.1038/s41586-024-07582-y A new study at the University of Maryland School of Medicine (UMSOM) has shed light on the frustrating experience of forgetting a person’s face after only speaking with them hours earlier. This type of memory decline is common with age, but the reason for it is still not fully understood. The study, published in Aging Cell, offers some valuable insights into this dysfunction. Research identifies target for potentially developing new therapies to treat age-related cognitive decline. One of the most upsetting aspects of age-related memory decline is not being able to remember the face that accompanies the name of a person you just talked with hours earlier. While researchers don’t understand why this dysfunction occurs, a new study conducted at the University of Maryland School of Medicine (UMSOM) has provided some important new clues. The study was published recently in the journal Aging Cell. Using aging mice, researchers have identified a new mechanism in neurons that causes memories associated with these social interactions to decline with age. In addition, they were able to reverse this memory loss in the lab. The researchers report that their findings identified a specific target in the brain that may one day be used to develop therapies that could prevent or reverse memory loss due to typical aging. Aging memory problems are distinct from those caused by diseases like Alzheimer’s or dementia. Credit: University of Maryland School of Medicine “If an older adult attends a cocktail party, afterward they would most likely recognize the names or the faces of the other attendees, but they might struggle with remembering which name went with which face,” said the study leader Michy Kelly, PhD, Associate Professor of Anatomy and Neurobiology at UMSOM. These kinds of memories that associate multiple pieces of information within a personal interaction, so-called social associative memories, require an enzyme, known as PDE11A, in a part of the brain responsible for memory involving life experiences. Last year, Dr. Kelly published research on PDE11A demonstrating that mice with genetically similar versions of the PDE11 enzyme were more likely to interact than those mice with a different type of PDE11A. In this new study, Dr. Kelly and her team sought to determine PDE11A’s role in social associative memory in the aging brain and whether manipulating this enzyme could be used to prevent this memory loss. Researchers can study mouse “social interactions” with their neighbors by seeing whether they will be willing to try a new food, based on their memories of encountering that food on the breath of another mouse. Mice do not like to eat new foods to avoid getting sick or even dying from them. When they smell food on another mouse’s breath, mice make an association between the food odor and the smell of the other mouse’s pheromones, the memory of which serves as a safety signal that any food with that odor is safe to eat in the future. PDE11A Levels Increase with Age Dr. Kelly and her colleagues found that although old mice could recognize both food odors and social odors separately, they were not able to remember the association between the two, similar to the cognitive decline in older people. They also discovered that levels of PDE11A increased with age in both people and mice, specifically in a brain region responsible for many types of learning and memory known as the hippocampus. This extra PDE11A in the hippocampus was not simply found where it was normally located in young mice; instead, it preferentially accumulated as little filaments in compartments of neurons. Genetic Deletion of PDE11A Restores Memory The researchers wondered if having too much PDE11A in these filaments was why the older mice forgot their social associative memories and would no longer eat the safe food they smelled on another mouse’s breath. To answer this question, they prevented these age-related increases in PDE11A by genetically deleting the PDE11A gene in mice. Without PDE11A, the older mice no longer forgot the social associative memory, meaning they ate the safe food they smelled on another mouse’s breath. When the researchers added the PDE11A back into the hippocampus of these old mice, the mice once again forgot the social associative memory and would no longer eat the safe food. One potential pathway to drug development to prevent this memory loss in people lies in an additional finding: The researchers learned that the concentrated filaments of PDE11A had an extra chemical modification in a specific place on the enzyme that the other PDE11 diffused throughout the neuron did not have. When they prevented this chemical modification, it reduced PDE11 levels and also prevented it from accumulating as filaments. “PDE11 is involved in more things than just memory, including preferences for who you prefer to be around. So, if we are to develop a therapy to help with cognitive decline, we would not want to get rid of it entirely or it could cause other negative side effects,” said Dr. Kelly. She and her colleagues joke that any drug that eliminated PDE11 would ensure you would remember your friends and family, but you may no longer like them. “Thus, our goal is to figure out a way to target the bad form of PDE11A specifically, in order to not interfere with the normal, healthy function of the enzyme.” Dean Mark T.
