The largest-ever study of brain genetics has identified over 4,000 genetic variants associated with brain structure. The study, which utilized MRI scans from adults and children, revealed that different sets of genes contribute to the folding and size of the cortex, and found that many genes linked to brain size in the general population overlap with genes implicated in cephalic conditions, shedding light on the genetic basis of brain development and its implications for neurological and psychiatric conditions. Largest Genetic Study of the Brain Reveals 4,000 Influencing Factors The largest-ever study of the genetics of the brain which analyzed around 36,000 brain scans, has pinpointed over 4,000 genetic factors associated with brain structure. The research, led by the University of Cambridge team, was recently published in the journal Nature Genetics. Our brains are intricate and highly complex organs, displaying significant variation between individuals in aspects such as overall brain volume, the folding patterns of the brain, and the thickness of these folds. Little is known about how our genetic makeup shapes the development of the brain. To answer this question, a team led by researchers at the Autism Research Centre, University of Cambridge, accessed MRI scans from over 32,000 adults from the UK Biobank cohort and over 4,000 children from the US-based ABCD study. From these scans, the researchers measured multiple properties of the outermost layer of the brain called the cortex. These included measures of the area and volume of the cortex as well as how the cortex is folded. They then linked these properties, measured both across the entire cortex as well as in 180 individual regions of the cortex, to genetic information across the genome. The team identified over 4,000 genetic variants linked to brain structure. Distinct Genetic Contributions to Brain Size and Folding These findings have allowed researchers to confirm and, in some cases, identify, how different properties of the brain are genetically linked to each other. Dr. Varun Warrier from the Autism Research Centre, who co-led the study, said: “One question that has interested us for a while is if the same genes that are linked to how big the cortex is – measured as both volume and area – are also linked to how the cortex is folded. By measuring these different properties of the brain and linking them to genetics, we found that different sets of genes contribute to folding and size of the cortex.” Overlapping Genes in Neurological Conditions The team also checked whether the same genes that are linked to variation in brain size in the general population overlap with genes linked to clinical conditions where head sizes are much larger or smaller than the general population, known as cephalic conditions. Dr. Richard Bethlehem, also from the Autism Research Centre and a co-lead of the study, said: “Many of the genes linked with differences in the brain sizes in the general population overlapped with genes implicated in cephalic conditions. However, we still do not know how exactly these genes lead to changes in brain size.” Dr. Warrier added: “This work shows that how our brain develops is partly genetic. Wayne State University School of Medicine researchers have discovered a unique approach for measuring the age of male sperm has the potential to predict pregnancy success and timing. A New Sperm Age Measurement Could Predict Pregnancy Success Pregnancy, often known as gestation, is the period of time when one or more fetuses grow within a woman’s womb. A multiple pregnancy births more than one offspring, such as twins. Pregnancy is normally caused through sexual intercourse, however, it can also be caused via assisted reproductive technology procedures. Pregnancy may result in a live birth, spontaneous miscarriage, induced abortion, or stillbirth. Childbirth usually happens approximately 40 weeks after the last menstrual cycle begins. According to a recent study conducted by scientists at the Wayne State University School of Medicine, a novel technique for measuring the age of male sperm has the potential to predict pregnancy success and time. The research, which was published on May 13th, 2022 in the journal Human Reproduction, discovered that sperm epigenetic aging clocks may operate as a potential biomarker to estimate couples’ time to conception. Credit: Wayne State University Biological vs. Chronological Age of Sperm “Chronological age is a significant determinant of reproductive capacity and success among couples attempting pregnancy, but chronological age does not encapsulate the cumulative genetic and external – environmental conditions – factors, and thus it serves as a proxy measure of the ‘true’ biological age of cells,” said J. Richard Pilsner, Ph.D., lead author of the study. Dr. Pilsner is the Robert J. Sokol, M.D., Endowed Chair of Molecular Obstetrics and Gynecology, and Director of Molecular Genetics and Infertility at WSU’s C.S. Mott Center for Human Growth and Development. “Semen quality outcomes utilizing World Health Organization guidelines have been used to assess male infertility for decades, but they remain poor predictors of reproductive outcomes. Thus, the ability to capture the biological age of sperm may provide a novel platform to better assess the male contribution to reproductive success, especially among infertile couples.” Sperm Epigenetic Aging and Conception Probability Sperm epigenetic aging is the biological aging of sperm rather than chronological aging. The research discovered a 17% lower cumulative probability of conception after 12 months for couples with male partners in older sperm epigenetic aging categories compared to younger sperm epigenetic aging categories. The research comprised 379 male partners of couples who had stopped using contraception in order to become pregnant. The research also discovered that males who smoked had higher epigenetic age of their sperm. The results, Dr. Pilsner said, indicate that higher sperm epigenetic aging is associated with a longer time to become pregnant in couples not assisted by fertility treatment, and among couples that achieved pregnancy, with shorter gestation. Implications for Pregnancy and Child Health The strong association between sperm epigenetic aging and pregnancy probability and its slowing or reversal through lifestyle choices and/or pharmacological interventions warrants further investigation. In addition, because older fathers have an increased risk of children with adverse neurological outcomes, it is important to understand the potential relation of sperm epigenetic aging on children’s health and development. “There is a critical need for new measures of male fecundity for assessing overall reproductive success among couples in the general population,” Dr. Pilsner said. “These data show that our sperm epigenetic clocks may fulfill this need as a novel biomarker that predicts pregnancy success among couples not seeking fertility treatment. While the chronological age of both partners remains a significant predictor of reproductive success, our clocks likely recapitulate both external and internal factors that drive the biological aging of sperm. Such a summary measure of sperm biological age is of clinical importance, as it allows couples in the general population to realize their probability of achieving pregnancy during natural intercourse, thereby informing and expediting potential infertility treatment decisions.” Dr. Pilsner advised that because those studied were largely Caucasian, greater and more diverse cohorts are necessary to confirm the association between sperm epigenetic aging and couple pregnancy success in other races and ethnicities. Less widely known is the fact that, like kidneys in other animals, fish gills control the pH and salt balance of their blood. This lesser-known gill function referred to as “ion regulation,” has historically been assumed to have evolved in tandem with breathing. However, a surprising new study published in Nature is introducing a new, early chapter to the evolutionary history of gills. “Our work suggests that the early, simplified gills of our worm-like ancestors played an important role in ion regulation. And that role might have originated as early as the very inception of gills, well before they played any role in breathing,” says Dr. Michael Sackville, a zoologist who led the study while with the University of British Columbia (UBC). “This really does flip the script on our understanding of how gills and gill function evolved.” Researchers at the University of British Columbia used three representative animals as a foundation to learn when and how gills acquired their functions. Prior to this research, it was believed that gills were initially utilized for breathing and ion control near the beginning of vertebrate life. These two functions changed from the skin to the gills in tandem in this traditional timeline, helping vertebrates in their transformation from tiny, worm-like organisms to larger, active fishes. This transformation from “small and wormy” to “big and fishy” is a defining moment in vertebrate evolution. The research compared three species that are still alive today but belong to distinct lineages: lampreys, which are vertebrates, and amphioxus and acorn worms, which are close relatives of vertebrates. The researchers reasoned that any gill functions shared by the animals were acquired from a common ancestor, which is thought to have existed well over 500 million years ago. “We found that gills were used for breathing in only our vertebrate representative, and only with increasing body size and activity,” says Dr. Colin Brauner, a UBC zoologist and senior author on the paper. “But we found ion-regulating cells in the gills of all three of our animals. This allowed us to trace the origin of ion regulation at gills all the way back to early deuterostome animals when very simple gill structures are thought to have first evolved.
