What are the cellular mechanisms within a single-celled marine algae species responsible for triggering toxic algal blooms? A research group under the direction of microbiologist Prof. Dr. Ralf Rabus from the University of Oldenburg, Germany, has conducted first detailed analyses of the unusual cell biology of Prorocentrum cordatum, a globally widespread species of the dinoflagellates group, using both advanced microscopic and proteomics approaches. As the team reports in the science journal Plant Physiology, the photosynthesis process in these microorganisms is organised in an unusual configuration which may help them to better adapt to the changing light conditions in the oceans. The results of the study could lead to an improved understanding of the incidence of harmful algal blooms, which may be becoming more frequent due to climate change. Dinoflagellates are important organisms in both marine and freshwater ecosystems. These unicellular organisms make up a substantial proportion of free-living phytoplankton, which forms the basis of the food web in oceans and lakes. Credit: University of Oldenburg / General and Molecular Microbiology group “We studied this organism because despite its environmental relevance its cell biology and metabolic physiology are still poorly understood,” said Rabus. In addition to studying photosynthesis in the microalgae, the researchers also examined the structure of their cell nuclei and their response to heat stress in collaboration with teams from the Universities of Hanover, Braunschweig, and Munich and set out the findings in two other recently published papers. Advanced Imaging Techniques Reveal Unique Cellular Structures Using a powerful scanning electron microscope with a focused ion beam at the Ludwig-Maximilians-Universität Munich, the team headed by Rabus and lead author Jana Kalvelage from the Institute of Chemistry and Biology of the Marine Environment (ICBM) was able to reconstruct the three-dimensional architecture of the chloroplasts, where photosynthesis takes place. The scientists were able to generate around 600 image layers of a single algae cell and then combine the sections to create a three-dimensional, high-resolution spatial image of the oval-shaped single-celled organisms, which are generally around 10 to 20 thousandths of a millimeter long. The analysis revealed that Prorocentrum cordatum have only a single barrel-like chloroplast that takes up 40 percent of their cell volume. Proteomic (protein) analyses then revealed marked differences between the photosynthetic apparatus of the microalgae and that of Arabidopsis thaliana, a well-studied model plant in genetics research. In both species, photosynthesis takes place in complex protein structures embedded in the chloroplast’s extensive membrane system. However, in Prorocentrum cordatum the team observed that the conversion of solar energy into biochemical energy takes place in a single large structure consisting of numerous proteins, known as a “megacomplex”, whereas in the chloroplasts of the plant species, the different steps of photosynthesis occur in spatially separated structures. The team also reported that P. cordatum uses a large number of different pigment-binding proteins to efficiently capture solar energy. “This diversity is a special adaptation to the changing light conditions to which the organism is exposed in the oceans,” Rabus explained. Exploring Genetic Complexity and Adaptability Two other studies published last year highlight the microalgae’s unusual biology: in the first a German-Australian team of which the ICBM researchers were also members found that the organisms have a very large genome with twice as many base pairs as in humans. The team also discovered that the algae change their metabolism and their rate of growth decelerates in response to heat stress. In a second publication, the team led by Rabus and Kalvelage described the cell nucleus in greater detail, reporting that P. cordatum has 62 chromosomes, an unusually high number that fills almost the entire cell nucleus. The function of a large proportion of the nuclear proteins that were identified by the researchers is currently unknown, the team observed. “We have investigated how this important microalgae functions at the molecular level. These findings form the basis for a better understanding of its role in the environment,” Rabus stressed. Further investigations could provide answers to questions such as how the organism’s metabolism reacts to other stress factors – and why the species is able to adapt to such a wide range of environmental conditions, from those in the tropics to those in temperate climates, he explained. Goodsell and RCSB PDB Genetic code evolution and Darwin’s evolution theory should consider DNA an “energy code.” Darwin’s theory of evolution should be expanded to include consideration of a DNA stability “energy code” – so-called “molecular Darwinism” – to further account for the long-term survival of species’ characteristics on Earth, according to Rutgers scientists. The iconic genetic code can be viewed as an “energy code” that evolved by following the laws of thermodynamics (flow of energy), causing its evolution to culminate in a nearly singular code for all living species, according to the Rutgers co-authored study in the journal Quarterly Reviews of Biophysics. “These revelations matter because they provide entirely new ways of analyzing the human genome and the genome of any living species, the blueprints of life,” said senior author Kenneth J. Breslauer, Linus C. Pauling Distinguished University Professor in the Department of Chemistry and Chemical Biology in the School of Arts and Sciences at Rutgers University–New Brunswick. He is also affiliated with the Rutgers Cancer Institute of New Jersey. “The origins of the evolution of the DNA genetic code and the evolution of all living species are embedded in the different energy profiles of their molecular DNA blueprints. Under the influence of the laws of thermodynamics, this energy code evolved, out of an astronomical number of alternative possibilities, into a nearly singular code across all living species.” Scientists investigated this so-called “universal enigma,” probing the origins of the astounding observation that the genetic code evolved into a nearly uniform blueprint that arose from trillions of possibilities. From Natural Selection to DNA Stability The scientists expanded the underpinnings of the landmark “survival of the fittest” Darwinian evolutionary theory to include “molecular Darwinism.” Darwin’s revolutionary theory is based on the generational persistence of a species’ physical features that allow it to survive in a given environment through “natural selection.” Molecular Darwinism refers to physical characteristics that persist through generations because the regions of the molecular DNA that code for those traits are unusually stable. Different DNA regions can exhibit differential energy signatures that may favor physical structures in organisms that enable specific biological functions, Breslauer said. Next steps include recasting and mapping the human genome chemical sequence into an “energy genome,” so DNA regions with different energy stabilities can be correlated with physical structures and biological functions. That would enable better selection of DNA targets for molecular-based therapeutics. A new study suggests that the weathering of sulfate rocks, not increasing ocean phosphorus levels, was crucial to the oxygenation of Earth’s atmosphere, influencing the late evolution of animal life, and also indicates that potential for complex intelligent life on other planets might require longer incubation times. Scientists identified sulfate as the missing link in Earth’s oxygenation, reshaping our understanding of life’s evolution and the potential for intelligent life on other planets. A recent research study may have discovered a missing link that helps explain Earth’s uniquely oxygen-saturated atmosphere and the corresponding evolution of animal life on our planet. The study, led by a Fellow of the Forrest Research Foundation at The University of Western Australia and recently published in the esteemed journal Nature, may hold the key to understanding why, for almost 90% of Earth’s history, oxygen levels remained too low for animals to breathe. The first major evolutionary event of animal life occurred during an event dubbed the Shuram Excursion – between 570 and 550 million years ago – which is believed to represent a massive release of carbon dioxide and oxygen into the atmosphere and oceans as a result of increasing ocean phosphorus levels. To test the theory, researchers used a newly developed tool to track the abundance of phosphorus in the oceans hundreds of millions of years ago, recorded in six locations in Australia, China, Mexico, and the US. Sulfate: The True Driver of Earth’s Oxygenation? The data and Earth chemistry model revealed increasing ocean phosphorus levels could not have explained the rise of oxygen. The effect was only replicated by the model when large quantities of sulfate rock were weathered, releasing sulfate into the oceans to produce vast amounts of oxygen. Lead author and Forrest Fellow Dr. Matthew Dodd, from the UWA School of Earth Sciences, said the results suggested sulfate, rather than phosphorus, was the main control in the oxygenation of the planet during the first major evolution of complex life. “Our findings may explain the prolonged low levels of oxygen throughout Earth’s history and consequently the late evolution of animal life on Earth,” Dr. Dodd said. “Importantly, we observe that ocean phosphorus was predominantly low when oxygen levels were low throughout the Shuram Excursion. This phenomenon would have locked the early oceans and atmosphere into an oxygen-devoid state.” Data from the study also holds implications about the possibility of intelligent life on other planets. “These results suggest other potentially habitable planets may support complex intelligent life, only if provided with long enough incubation times,” Dr. Dodd said. “This could imply planets around stars larger than the Sun may not develop complex intelligent life due to the relatively short lifetime of large stars.” Reference: “Uncovering the Ediacaran phosphorus cycle” by Matthew S.
