A study of 88 bird species found that some birds with high blood sugar levels resist glycation, challenging assumptions about metabolism and aging. While smaller birds had the highest glucose levels, lifespan did not consistently correlate with glycation. Unexpectedly, carnivorous birds showed higher glycation rates than omnivores despite similar glucose levels, suggesting other metabolic influences. A comprehensive comparative study indicates that certain birds have developed mechanisms to resist the detrimental effects of high blood sugar, offering valuable insights into metabolism and aging. A large-scale study of 88 bird species has found that some birds with high blood sugar levels exhibit resistance to protein glycation, indicating they may have evolved mechanisms to mitigate the harmful effects commonly associated with glucose metabolism. This study represents the first comprehensive comparative analysis of how blood sugar levels, glycation rates, diet, and life history interact in birds. Published as a revised Reviewed Preprint in eLife, the research provides strong evidence of a positive correlation between blood sugar levels and glycation. While the editors acknowledge some data collection limitations, they suggest these constraints likely make the statistically significant findings more conservative rather than overstated. Understanding how different species regulate blood sugar is crucial for exploring the connections between metabolism, aging, and disease. High blood sugar is generally linked to accelerated aging and health issues, primarily due to protein glycation—a process in which sugars react with proteins, causing cellular damage. The pace-of-life syndrome hypothesis proposes that an organism’s metabolic rate, lifespan, reproductive strategies, and behavior evolve in predictable ways. Under this framework, species with fast metabolisms, short lifespans, and high reproductive rates are expected to have higher blood sugar and glycation levels. Conversely, those with longer lifespans and slower developmental times should have lower blood sugar levels and greater resistance to glycation. However, it is unclear how glycation has coevolved with other traits across species, and so it is undetermined whether glycation fits into the framework of the pace-of-life hypothesis. Birds as a Unique Case Study “Birds are particularly relevant in this context, given their relatively high blood sugar levels – on average almost twice as high as similarly sized mammals,” says lead author Adrián Moreno-Borrallo, (PhD) at the Institut Pluridisciplinaire Hubert Curien, University of Strasbourg, CNRS, Strasbourg, France. “This is thought to be an adaptation allowing flight, providing birds with the fuel needed to power intense bursts of aerobic exercise. But it is also paradoxical. Despite their higher blood sugar levels, birds show remarkable longevity compared to their mammalian counterparts, living up to three times longer.” To investigate this apparent paradox, Moreno-Borrallo and colleagues conducted an analysis of 484 individual birds from 88 different species. They compared blood sugar levels and glycation rates in relation to the birds’ life history traits – lifespan, body mass, developmental time, reproductive investment (measured by clutch mass – the total mass of their eggs), and diet. They sought to identify whether these species of birds followed the framework set by the pace-of-life syndrome hypothesis. Their results revealed substantial variation in blood sugar levels across species. Smaller birds, such as swifts and passerines, had the highest blood sugar levels, while larger species, such as flamingos and geese, had the lowest. Glycation rates followed a similar trend, with smaller birds showing higher levels and larger birds displaying lower levels. However, the relationship between blood sugar levels and lifespan was more complex. While longer-lived birds generally had higher blood sugar levels, this increase plateaued beyond a certain point. This suggests that some species have evolved mechanisms to prevent glycation-related damage, rather than avoiding high blood sugar levels altogether. Another unexpected finding was that terrestrial carnivorous birds had significantly higher glycation rates than omnivorous birds, despite having similar blood sugar levels. This indicates that factors beyond blood sugar levels, such as differences in protein metabolism, antioxidant defenses, or dietary components including fiber and polyunsaturated fatty acids, may influence how different species are able to mitigate glycation-related damage. Challenging the Pace-of-Life Hypothesis The results challenge some predictions of the pace-of-life syndrome hypothesis. While body size showed a strong relation to blood sugar levels, reproductive investment showed no significant relationship with glycation, and glucose levels increased with lifespan instead of decreasing. “Our results are only in minor agreement with predictions from the pace-of-life syndrome hypothesis. It holds true for body mass, but not for the other life-history variables tested,” says co-senior author Fabrice Bertile, a researcher at the Institut Pluridisciplinaire Hubert Curien, University of Strasbourg, CNRS, and the National Proteomics Infrastructure, ProFi, Strasbourg, France. “The link between blood sugar levels and lifespan may depend on species-specific factors that influence how glucose affects aging. Further research should target this area, as well as exploring other groups of birds that are underrepresented in our study due to logistical constraints.” “These results increase our knowledge about the diversity of blood sugar levels and glycation patterns across birds, challenging conventional wisdom about metabolism, aging, and disease,” concludes co-senior author François Criscuolo, a researcher at the Institut Pluridisciplinaire Hubert Curien, University of Strasbourg, CNRS. “There is considerable variation in blood sugar levels and glycation rates between species, with those with the highest glucose levels showing greater resistance to glycation, hinting at an evolved mechanism for this resistance. University of Queensland researchers have unveiled how Haemophilus influenzae manipulates immune responses to foster chronic respiratory infections. By deactivating immune defenses, this bacterium avoids detection and exacerbates conditions like asthma and COPD. The findings suggest potential for new treatments that boost the immune system’s ability to counteract this bacterium. Scientists at The University of Queensland have identified how a common bacterium can manipulate the human immune system during respiratory infections and cause persistent illness. The researchers were led by Professor Ulrike Kappler from UQ’s School of Chemical and Molecular Biosciences. The study investigated the virulence mechanisms of Haemophilus influenzae, a bacterium that plays a significant role in worsening respiratory tract infections. Credit: UQ The Impact on Vulnerable Populations “These bacteria are especially damaging to vulnerable groups, such as those with cystic fibrosis, asthma, the elderly, and Indigenous communities,” Professor Kappler said. “In some conditions, such as asthma and chronic obstructive pulmonary disease, they can drastically worsen symptoms. “Our research shows the bacterium persists by essentially turning off the body’s immune responses, inducing a state of tolerance in human respiratory tissues.” Professor Kappler said the bacterium had a unique ability to ‘talk’ to and deactivate the immune system, convincing it there was no threat. Innovative Research Methods and Findings The researchers prepared human nasal tissue in the lab, growing it to resemble the surfaces of the human respiratory tract, then monitored gene expression changes over a 14-day ‘infection’. They found very limited production of inflammation molecules over time, which normally would be produced within hours of bacteria infecting human cells. “We then applied both live and dead Haemophilus influenzae, showing the dead bacteria caused a fast production of the inflammation makers, while live bacteria prevented this,” Professor Kappler said. “This proved that the bacteria can actively reduce the human immune response.” Co-author and pediatric respiratory physician Emeritus Professor Peter Sly from UQ’s Faculty of Medicine, said the results show how Haemophilus influenzae can cause chronic infections, essentially living in the cells that form the surface of the respiratory tract. Implications for Future Treatment Strategies “This is a rare behavior that many other bacteria don’t possess,” Emeritus Professor Sly said. “If local immunity drops, for example during a viral infection, the bacteria may be able to ‘take over’ and cause a more severe infection.” The findings will lead to future work toward new treatments to prevent these infections by helping the immune system recognize and kill these bacteria. “We’ll look at ways of developing treatments that enhance the immune system’s ability to detect and eliminate the pathogen before it can cause further damage,” Professor Kappler said. What are the cellular mechanisms within a single-celled marine algae species responsible for triggering toxic algal blooms? A research group under the direction of microbiologist Prof. Dr. Ralf Rabus from the University of Oldenburg, Germany, has conducted first detailed analyses of the unusual cell biology of Prorocentrum cordatum, a globally widespread species of the dinoflagellates group, using both advanced microscopic and proteomics approaches. As the team reports in the science journal Plant Physiology, the photosynthesis process in these microorganisms is organised in an unusual configuration which may help them to better adapt to the changing light conditions in the oceans. The results of the study could lead to an improved understanding of the incidence of harmful algal blooms, which may be becoming more frequent due to climate change. Dinoflagellates are important organisms in both marine and freshwater ecosystems. These unicellular organisms make up a substantial proportion of free-living phytoplankton, which forms the basis of the food web in oceans and lakes. Credit: University of Oldenburg / General and Molecular Microbiology group “We studied this organism because despite its environmental relevance its cell biology and metabolic physiology are still poorly understood,” said Rabus. In addition to studying photosynthesis in the microalgae, the researchers also examined the structure of their cell nuclei and their response to heat stress in collaboration with teams from the Universities of Hanover, Braunschweig, and Munich and set out the findings in two other recently published papers. Advanced Imaging Techniques Reveal Unique Cellular Structures Using a powerful scanning electron microscope with a focused ion beam at the Ludwig-Maximilians-Universität Munich, the team headed by Rabus and lead author Jana Kalvelage from the Institute of Chemistry and Biology of the Marine Environment (ICBM) was able to reconstruct the three-dimensional architecture of the chloroplasts, where photosynthesis takes place. The scientists were able to generate around 600 image layers of a single algae cell and then combine the sections to create a three-dimensional, high-resolution spatial image of the oval-shaped single-celled organisms, which are generally around 10 to 20 thousandths of a millimeter long. The analysis revealed that Prorocentrum cordatum have only a single barrel-like chloroplast that takes up 40 percent of their cell volume. Proteomic (protein) analyses then revealed marked differences between the photosynthetic apparatus of the microalgae and that of Arabidopsis thaliana, a well-studied model plant in genetics research. In both species, photosynthesis takes place in complex protein structures embedded in the chloroplast’s extensive membrane system. However, in Prorocentrum cordatum the team observed that the conversion of solar energy into biochemical energy takes place in a single large structure consisting of numerous proteins, known as a “megacomplex”, whereas in the chloroplasts of the plant species, the different steps of photosynthesis occur in spatially separated structures. The team also reported that P. cordatum uses a large number of different pigment-binding proteins to efficiently capture solar energy. “This diversity is a special adaptation to the changing light conditions to which the organism is exposed in the oceans,” Rabus explained. Exploring Genetic Complexity and Adaptability Two other studies published last year highlight the microalgae’s unusual biology: in the first a German-Australian team of which the ICBM researchers were also members found that the organisms have a very large genome with twice as many base pairs as in humans. The team also discovered that the algae change their metabolism and their rate of growth decelerates in response to heat stress. In a second publication, the team led by Rabus and Kalvelage described the cell nucleus in greater detail, reporting that P. cordatum has 62 chromosomes, an unusually high number that fills almost the entire cell nucleus. The function of a large proportion of the nuclear proteins that were identified by the researchers is currently unknown, the team observed. “We have investigated how this important microalgae functions at the molecular level.
