Vestigial auricular muscles still activate when we strain to hear, especially in noisy environments. Researchers found that their activity increases with listening difficulty, hinting at a possible role in measuring auditory effort. The auricular muscles, which enabled our distant ancestors to move their ears for better hearing, activate when people try to focus on competing sounds. If you can wiggle your ears, you’re using muscles that once helped our distant ancestors listen more attentively. These auricular muscles altered the shape of the pinna—the outer shell of the ear—funneling sound toward the eardrums. Millions of years ago, our ancestors stopped relying on them, leaving humans with only vestigial auricular muscles. However, scientists studying these muscles have recently discovered that they still activate when we try to focus on competing sounds. “There are three large muscles which connect the auricle to the skull and scalp and are important for ear wiggling,” explained Andreas Schröer of Saarland University, first author of the study in Frontiers in Neuroscience. “These muscles, particularly the superior auricular muscle, exhibit increased activity during effortful listening tasks. This suggests that these muscles are engaged not merely as a reflex but potentially as part of an attentional effort mechanism, especially in challenging auditory environments.” Straining your ears It’s difficult to test how hard someone is listening without self-reported measures. But electromyography, which measures electrical activity in a muscle, can help identify activity in the auricular muscles linked to listening closely. Similar research has already shown that the largest muscles, posterior and superior auricular muscles, react during attentive listening. Because they pull the ears up and back, they are considered likely to have been involved in moving the pinna to capture sounds. “The exact reason these became vestigial is difficult to tell, as our ancestors lost this ability about 25 million years ago,” Schröer said. “One possible explanation could be that the evolutionary pressure to move the ears ceased because we became much more proficient with our visual and vocal systems.” To test whether these muscles are more active during more difficult listening tasks, the scientists recruited 20 people without hearing problems. They applied electrodes to participants’ auricular muscles and then played them an audiobook and distracting podcasts from speakers in front of or behind them. Each participant underwent 12 five-minute trials, covering three different levels of difficulty. On easy mode, the podcast was quieter than the audiobook, and the speaker’s voice was a stronger contrast to the audiobook. To create two more difficult modes, the scientists added a podcast that sounded more like the audiobook and made the distractors louder. However, the researchers were careful to make even the most difficult condition achievable: if the participants gave up, no physiological effort would register. Then, the scientists asked the participants to rate their effort levels and to estimate how often they lost the thread of the audiobook in each trial. They also quizzed the participants on the audiobook’s content. Hearsay The scientists found that the two auricular muscles reacted differently to the different conditions. The posterior auricular muscles reacted to changes in direction, while the superior auricular muscles reacted to the difficulty level of the task. Participants’ self-reported measures of effort and how often they lost track of the audiobook rose in line with the difficulty of the task, and the accuracy of their responses to questions about the audiobook dropped noticeably between the medium and the difficult mode. This correlated with the superior auricular muscles’ activity levels: they didn’t activate more during the medium mode than during the easy mode, but were very active during the difficult mode. This suggests that the activity of the superior auricular muscles could provide an objective measure of listening effort, although it’s unclear if the muscle activity helps people hear. “The ear movements that could be generated by the signals we have recorded are so minuscule that there is probably no perceivable benefit,” said Schröer. “However, the auricle itself does contribute to our ability to localize sounds. So, our auriculomotor system probably tries its best after being vestigial for 25 million years, but does not achieve much.” The scientists pointed out that more work will be necessary to confirm these results and develop practical applications. Like many hearing studies, their sample was relatively small and composed of young people without hearing problems. Larger, more diverse participant groups, in more realistic conditions, are needed. “Investigating the possible effects of muscle strain itself or the ear’s minuscule movements on the transmission of sound is something we want to do in the future,” said Schröer. “The effect of these factors in people with hearing impairments would also be interesting to investigate.” Reference: “Electromyographic correlates of effortful listening in the vestigial auriculomotor system” by Andreas Schroeer, Farah I. Scientists Have Developed a Method To Identify Aging Muscle Stem Cells The issue of aging and the fight against it has long been prevalent in both classic and contemporary literature throughout human history. From the ill-fated Qin Shi Huang’s expedition to the sea in pursuit of eternal life to the fame of Count Dracula in the West, aging has caught the world’s fascination for thousands of years and remains unsolved. In an exciting breakthrough, scientists have developed a way to identify aged muscle stem cells (MuSCs) based on their chromatin signature. MuSCs play an important role in muscle repair. The research team was from the Hong Kong University of Science and Technology (HKUST) and was led by Professor Tom Cheung, an associate professor of life sciences. In contrast to their younger counterparts, aging MuSCs have decreased stemness (the ability to become new stem cells or turn into specialized cells to replace damaged tissues). If the chromatin signature of an aged cell can be restored to that of a young cell, the process of cellular aging—and, in this example, skeletal muscle tissue aging—may be slowed or even reversed. The findings were recently published in the journal iScience. Chromatin Signature and Stem Cell Aging “The regulation of chromatin accessibility is critical for cell fate decisions,” said Professor Cheung. “Changes in the chromatin state can lead to dysregulation of gene expression. In our study, we were able to identify the chronically activated chromatin state as a hallmark of stem cell aging, which could be a target for developing anti-aging strategies.” Chromatin, a complex of DNA that wraps around histones to maintain DNA in its proper architecture, undergoes rapid changes in its structure in response to the extrinsic environment. As a continuation of their previous study, the team pre-fixed muscle stem cells in the mouse to obtain quiescent cells (dormant cells that will activate to repair injured muscle) and obtained their gene and chromatin signatures, in which they then compared the chromatin accessibility over time. Differences in Young and Aged Muscle Stem Cells “We showed that the chromatin environment of young muscle stem cells is very compact during quiescence, becomes highly accessible on early activation, and gradually re-establishes the compact state after long-term regeneration. However, aged muscle stem cells lose their ability to maintain such a compact chromatin environment during quiescence,” said Dr. Anqi Dong, first author of the study and a former member of Professor Cheung’s research group who is now a Postdoctoral Fellow at the Université libre de Bruxelles. Many possibilities are waiting to be unearthed now that scientists have gained a better understanding of what happens to an aging cell, opening a variety of avenues for anti-aging strategies to be pursued further. “Have we solved the mystery of aging? Yes, but not quite,” noted Professor Cheung. “If we can find chromatin-modifying regulators that are downregulated in aged stem cells, these will be potential targets to prevent aging by restoring their expression. As we are able to make a clear comparison between the chromatin states of young and old muscle stem cells, we have also identified target locations that are specifically accessible in young muscle stem cells. If the accessibility of those regions can be maintained during aging, we may be able to find ways to keep cells young and healthy longer.” “Our current study describes the changes in chromatin accessibility during stem cell isolation and activation, but the journey has just begun,” said Professor Cheung. “We look forward to further investigating the mechanisms that alter the chromatin state during muscle stem cell isolation and activation, and it is important we conduct the same study in vivo for more insights.” Reference: “Global chromatin accessibility profiling analysis reveals a chronic activation state in aged muscle stem cells” by Anqi Dong, Jing Liu, Kangning Lin, Wenshu Zeng, Wai-Kin So, Shenyuan Hu and Tom H. Credit: Texas Biomed The development of a new cell culture model for human alveolar macrophages is set to propel research into respiratory illnesses. This innovation will aid in the exploration of a variety of conditions such as COVID-19, tuberculosis, asthma, cystic fibrosis, and chronic obstructive pulmonary disease. Scientists at the Texas Biomedical Research Institute have successfully cultivated the alveolar macrophage, a vital immune cell within the lung, in a laboratory setting. This development of a cell culture model significantly simplifies and reduces the cost for global researchers investigating lung inflammatory disorders and exploring potential new treatments. Macrophages are the “Pac-Man” of the immune system, eating up garbage throughout tissues in the body. Alveolar macrophages, in particular, reside in the lining of the lung’s air sacs where the exchange of air takes place. They are typically the initial immune cells to confront pathogens penetrating the deep sections of the lungs, such as SARS-CoV-2 or the bacteria responsible for TB. “It is critical to study tissue-specific cells to better understand mechanisms of health and disease, and to screen potential new therapies,” says Texas Biomed Professor Larry Schlesinger, MD, and senior author of the paper published in the journal mBio. Postdoctoral fellow Susanta Pahari, Ph.D., pivoted during the COVID-19 pandemic to develop the magic cocktail that generates the alveolar macrophage-like (AML) cells. Credit: Texas Biomed Old vs. New Human alveolar macrophages have been challenging to study because they reside deep in the lungs and are hard to access. Typically, they are collected through time-consuming and expensive lung washes, which involve using a bronchoscope to move through the throat and into the airways to collect fluid samples. This new model starts with a simple blood draw. White blood cells are isolated and placed in Teflon jars with specialized cell culture components. Surfactant is added along with three different cytokine proteins, which are usually found in the alveolar lining fluid. “We call it the magic cocktail,” says Susanta Pahari, Ph.D., a postdoctoral researcher at Texas Biomed and the first author of the paper. “We are mimicking the alveolar environment in cell culture. It makes the cells think they are in the lungs.” The generated alveolar macrophage-like cells (left) closely resemble human alveolar macrophages collected through lung washes (right), without the time, expense, and invasive collection procedure. Credit: Texas Biomed Within six days, the cells differentiate, or transform, into alveolar macrophage-like cells. The generated cells are 94% genetically similar to human alveolar macrophages collected from lung washes. The Texas Biomed team confirmed the model can be used to investigate TB and COVID-19; the cells readily take up the pathogens. “It is very rewarding to develop something that can help the research community,” says Dr. Pahari. “We’ve already received numerous emails across the globe requesting macrophage development protocols. We are now looking into developing a kit that we can provide to make it even easier for others to replicate what we have done.” Pivot & Improve In a way, the advancement is a byproduct of the COVID-19 pandemic. When the pandemic hit, Dr. Pahari could not readily access human alveolar macrophages, and his research came to a grinding halt. So, he pivoted to focus on developing an alternative. It took years of trial and error to identify the most effective combination of components that go into the cocktail, as well as to conduct genetic testing and verification. The model improves upon the standard approach used to create human macrophages in Dr. Schlesinger’s lab for many years. “We’ve been using human monocyte-derived macrophages which themselves are an excellent model but they do not closely resemble the unique alveolar macrophages,” says Dr. Schlesinger. Dr. Schlesinger notes that the approach that ultimately worked is reminiscent of the process to generate adult induced pluripotent stem cells, which place adult stem cells in a specific cocktail to help them revert to a state where they can then differentiate into totally new tissues. “I am excited to see the full potential of the alveolar macrophage-like cells and if they can be integrated into next-generation lung organoids,” Dr. Schlesinger says.
