Research shows that bottlenose dolphins use a smiling-like “open mouth” expression to communicate playfully, which their peers reciprocate 33% of the time, highlighting the role of facial expressions in managing social interactions across different species. This expression is mimicked by playmates 33% of the time, suggesting its role in dolphin communication. Dolphins are extremely playful, but little is known about how they—and other marine mammals—communicate during playtime. New research recently published in the Cell Press journal iScience shows that bottlenose dolphins (Tursiops truncates) use the “open mouth” facial expression—analogous to a smile—to communicate during social play. The dolphins almost always use the facial expressions when they are in their playmate’s field of view, and when playmates perceived a “smile,” they responded in kind 33% of the time. “We’ve uncovered the presence of a distinct facial display, the open mouth, in bottlenose dolphins, and we showed that dolphins are also able to mirror others’ facial expressions,” says senior author and evolutionary biologist Elisabetta Palagi of the University of Pisa. “Open-mouth signals and rapid mimicry appear repeatedly across the mammal family tree, which suggests that visual communication has played a crucial role in shaping complex social interactions, not only in dolphins but in many species over time.” Dolphin play can include acrobatics, surfing, playing with objects, chasing, and playfighting, and it’s important that these activities aren’t misinterpreted as aggression. Other mammals use facial expressions to communicate playfulness, but whether marine mammals also use facial expressions to signal playtime hasn’t been previously explored. Evolution of the Open Mouth Gesture “The open mouth gesture likely evolved from the biting action, breaking down the biting sequence to leave only the ‘intention to bite’ without contact,” says Palagi. “The relaxed open mouth, seen in social carnivores, monkeys’ play faces, and even human laughter, is a universal sign of playfulness, helping animals—and us—signal fun and avoid conflict.” To investigate whether dolphins visually communicate playfulness, the researchers recorded captive bottlenose dolphins while they were playing in pairs and while they were playing freely with their human trainers. They showed that dolphins frequently use the open-mouth expression when playing with other dolphins, but they don’t seem to use it when playing with humans or when they’re playing by themselves. While only one open-mouth event was recorded during solitary play, the researchers recorded a total of 1,288 open-mouth events during social play sessions, and 92% of these events occurred during dolphin-dolphin play sessions. Dolphins were also more likely to assume the open-mouth expression when their faces were in the field of view of their playmate—89% of recorded open-mouth expressions were emitted in this context—and when this “smile” was perceived, the playmate smiled back 33% of the time. Mimicry and Future Research “Some may argue that dolphins are merely mimicking each other’s open-mouth expressions by chance, given they’re often involved in the same activity or context, but this doesn’t explain why the probability of mimicking another dolphin’s open mouth within 1 second is 13 times higher when the receiver actually sees the original expression,” says Palagi. “This rate of mimicry in dolphins is consistent with what’s been observed in certain carnivores, such as meerkats and sun bears.” The researchers didn’t record the dolphins’ acoustic signals during playtime, and they say that future studies should investigate the possible role of vocalizations and tactile signals during playful interactions. “Future research should dive into eye-tracking to explore how dolphins see their world and utilize acoustic signals in their multimodal communication during play,” says corresponding author and zoologist Livio Favaro. “Dolphins have developed one of the most intricate vocal systems in the animal world, but sound can also expose them to predators or eavesdroppers. DOI: 10.1016/j.isci.2024.110966 Researchers have identified the genetic pathway between the heart and brain responsible for fainting, revealing a two-way communication that could lead to new treatments for syncope-related disorders. Neurobiologists have discovered sensory neurons that regulate fainting, providing a foundation for targeted treatments for related disorders. Syncope, commonly known as fainting, affects nearly 40 percent of people at least once in their lifetime. These transient losses of consciousness can be precipitated by various triggers such as pain, fear, heat, or hyperventilation, and they are a substantial cause of emergency room visits. Despite their prevalence, the fundamental mechanisms underlying syncope have largely remained enigmatic. Breakthrough in Genetic Pathways Publishing a new report in Nature, University of California San Diego researchers, along with colleagues at The Scripps Research Institute and other institutions, have for the first time identified the genetic pathway between the heart and brain tied to fainting. One of their unique approaches was to think of the heart as a sensory organ rather than the longstanding viewpoint that the brain sends out signals and the heart simply follows directions. School of Biological Sciences Assistant Professor Vineet Augustine, the paper’s senior author, applies a variety of approaches to better understand these neural connections between the heart and brain. Information resulting from the study could be relevant to better understanding and treating various psychiatric and neurological disorders linked with brain-heart connections, the researchers note in their paper. “Our study is the first comprehensive demonstration of a genetically defined cardiac reflex, which faithfully recapitulates characteristics of human syncope at physiological, behavioral, and neural network levels.” Study on the Bezold-Jarisch Reflex Augustine, along with Biological Sciences Staff Research Associate Jonathan Lovelace and Graduate Student Jingrui Ma, the first authors of the paper, and their colleagues studied neural mechanisms related to Bezold-Jarisch reflex (BJR), a cardiac reflex first described in 1867. For decades researchers have hypothesized that the BJR, which features reduced heart rate, blood pressure, and breathing, may be associated with fainting. But information lacked in proving the idea since the neural pathways involved in the reflex were not well known. Researchers at UC San Diego and collaborating institutions have highlighted the immense crosstalk between the heart and the nervous system. Credit: Augustine Lab, UC San Diego The researchers focused on the genetics behind a sensory cluster known as the nodose ganglia, which are part of the vagus nerves that carry signals between the brain and visceral organs, including the heart. Specifically, vagal sensory neurons, or VSNs, project signals to the brainstem and are thought to be associated with BJR and fainting. In their search for a novel neural pathway they discovered that VSNs expressing the neuropeptide Y receptor Y2 (known as NPY2R) are tightly linked to the well-known BJR responses. Optogenetic Studies and Findings Studying this pathway in mice, the researchers were surprised to find that when they proactively triggered NPY2R VSNs using optogenetics, a method of stimulating and controlling neurons, mice that had been freely moving about immediately fainted. During these episodes, they recorded from thousands of neurons in the brains of the mice, as well as heart activity and changes in facial features including pupil diameter and whisking. They also employed machine learning in several ways to analyze the data and pinpoint features of interest. Once NPY2R neurons were activated, they found, mice exhibited rapid pupil dilation and the classic “eye-roll” seen during human fainting, as well as suppressed heart rate, blood pressure, and breathing rate. They also measured reduced blood flow to the brain, an area of collaboration with Professor David Kleinfeld’s laboratory in the UC San Diego Departments of Neurobiology and Physics. “We were blown away when we saw how their eyes rolled back around the same time as brain activity rapidly dropped,” the researchers reported in a paper summary. “Then, after a few seconds, brain activity and movement returned. This was our eureka moment.” Further testing showed that when NPY2R VSNs were removed from mice, the BJR and fainting conditions vanished. Previous studies have shown that fainting is caused by a reduction in brain blood flow, which the new study also found to be true, but the new evidence indicated that brain activity itself could be playing an important role. The findings, therefore, implicate the activation of the newly genetically identified VSNs and their neural pathways not only with BJR, but more centrally in overall animal physiology, certain brain networks, and even behavior. Implications and Future Research Such findings were difficult to tease out previously because neuroscientists study the brain and cardiologists study the heart, but many do so in isolation of the other. “Neuroscientists traditionally think the body just follows the brain, but now it is becoming very clear that the body sends signals to the brain and then the brain changes function,” said Augustine. As a result of their findings, the researchers would like to continue tracking the precise conditions under which vagal sensory neurons are triggered into action. “We also hope to more closely examine cerebral blood flow and neural pathways in the brain during the moment of syncope, to better understand this common but mysterious condition,” they note. They also hope to use their research as a model to develop targeted treatments for fainting-associated conditions. A new study reveals a mechanism for generating DNA palindromes that could lead to new microRNA genes, shedding light on gene origins and potentially impacting our understanding of RNA structures. Researchers at the University of Helsinki have uncovered a mechanism that instantaneously generates DNA palindromes, potentially leading to the creation of new microRNA genes from noncoding DNA sequences. This discovery, which was made while studying DNA replication errors and their impact on RNA molecule structures, offers new insights into gene origins. The complexity of living organisms is encoded within their genes, but where do these genes come from? Researchers at the University of Helsinki resolved outstanding questions around the origin of small regulatory genes, and described a mechanism that creates their DNA palindromes. Under suitable circumstances, these palindromes evolve into microRNA genes. Genes and Proteins: The Building Blocks of Life The human genome contains ca. 20,000 genes that are used for the construction of proteins. Actions of these classical genes are coordinated by thousands of regulatory genes, the smallest of which encode microRNA molecules that are 22 base pairs in length. While the number of genes remains relatively constant, occasionally new genes emerge during evolution. Similar to the genesis of biological life, the origin of new genes has continued to fascinate scientists. Solving the Palindromic Puzzle All RNA molecules require palindromic runs of bases that lock the molecule into its functional conformation. Importantly, the chances of random base mutations gradually forming such palindromic runs are extremely small, even for the simple microRNA genes. Hence, the origin of these palindromic sequences has puzzled researchers. Experts at the Institute of Biotechnology, University of Helsinki, Finland resolved this mystery, describing a mechanism that can instantaneously generate complete DNA palindromes and thus create new microRNA genes from previously noncoding DNA sequences. DNA Replication Insights In a project funded by the Academy of Finland, researchers studied errors in DNA replication. Ari Löytynoja, the project leader, compares DNA replication to typing of text. “DNA is copied one base at a time, and typically mutations are erroneous single bases, like mis-punches on a laptop keyboard. We studied a mechanism creating larger errors, like copy-pasting text from another context. We were especially interested in cases that copied the text backward so that it creates a palindrome.” Researchers studied an error mechanism in DNA replication, and noticed that some errors create palindromes that can fold into hairpin structures. Credit: Ari Löytynoja RNA Structures and DNA Errors Researchers recognized that DNA replication errors could sometimes be beneficial. They described these findings to Mikko Frilander, an expert in RNA biology. He immediately saw the connection to the structure of RNA molecules. “In an RNA molecule, the bases of adjacent palindromes can pair and form structures resembling a hairpin. Such structures are crucial for the function of the RNA molecules,” he explains. Researchers decided to focus on microRNA genes due to their simple structure: the genes are very short – just a few tens of bases – and they have to fold into a hairpin structure to function correctly. A central insight was to model the gene history using a custom computer algorithm. According to postdoctoral researcher Heli Mönttinen, this enables the closest inspection of the origin of genes thus far. “The whole genome of tens of primates and mammals is known. A comparison of their genomes reveals which species have the microRNA palindrome pair, and which lack it. With a detailed modeling of the history, we could see that whole palindromes are created by single mutation events,” says Mönttinen. Credit: Ari Löytynoja Implications and Universality By focusing on humans and other primates, researchers in Helsinki demonstrated that the newly found mechanism can explain at least a quarter of the novel microRNA genes. As similar cases were found in other evolutionary lineages, the origin mechanism appears universal. In principle, the rise of microRNA genes is so easy that novel genes could affect human health. Heli Mönttinen sees the significance of the work more broadly, for example in understanding the basic principles of biological life. “The emergence of new genes from nothing has fascinated researchers. We now have an elegant model for the evolution of RNA genes,” she highlights. Although the results are based on small regulatory genes, researchers believe that the findings can be generalized to other RNA genes and molecules. For example, by using the raw materials generated by the newly found mechanism, natural selection may create much more complex RNA structures and functions. The study was published in PNAS.
