Researchers have discovered the oldest known fragment of fossilized skin, over 21 million years older than previous findings, from an early Paleozoic reptile. The study highlights the skin’s resemblance to modern reptile skin and its importance in understanding the evolutionary transition to land. (Artist’s concept.) Credit: SciTechDaily.com A groundbreaking discovery reveals the oldest fossilized reptile skin, showing significant evolutionary features and offering a unique perspective on early terrestrial animal life. Preserved in an Oklahoma cave system, this finding sheds light on the development of skin adaptations in vertebrates. Researchers have identified a 3D fragment of fossilized skin that is at least 21 million years older than previously described skin fossils. The skin, which belonged to an early species of Paleozoic reptile, has a pebbled surface and most closely resembles crocodile skin. It’s the oldest example of preserved epidermis, the outermost layer of skin in terrestrial reptiles, birds, and mammals, which was an important evolutionary adaptation in the transition to life on land. The fossil was described recently in the journal Current Biology along with several other specimens that were collected from the Richards Spur limestone cave system in Oklahoma. A Glimpse Into Ancient Life “Every now and then we get an exceptional opportunity to glimpse back into deep time,” says first author Ethan Mooney, a paleontology graduate student at the University of Toronto who worked on the project as an undergraduate with paleontologist Robert Reisz at the University of Toronto. “These types of discoveries can really enrich our understanding and perception of these pioneering animals.” Skin and other soft tissues are rarely fossilized, but the researchers think that skin preservation was possible in this case because of the cave system’s unique features, which included fine clay sediments that slowed decomposition, oil seepage, and a cave environment that was likely an oxygenless environment. Credit: Current Biology/Mooney et al. “Animals would have fallen into this cave system during the early Permian and been buried in very fine clay sediments that delayed the decay process,” says Mooney. “But the kicker is that this cave system was also an active oil seepage site during the Permian, and interactions between hydrocarbons in petroleum and tar are likely what allowed this skin to be preserved.” Uncovering the Details The skin fossil is tiny—smaller than a fingernail. Microscopic examination undertaken by coauthor Tea Maho of the University of Toronto Mississauga revealed epidermal tissues, a hallmark of the skin of amniotes, the terrestrial vertebrate group that includes reptiles, birds, and mammals and that evolved from amphibian ancestors during the Carboniferous Period. “We were totally shocked by what we saw because it’s completely unlike anything we would have expected,” says Mooney. “Finding such an old skin fossil is an exceptional opportunity to peer into the past and see what the skin of some of these earliest animals may have looked like.” Reptilian Features and Evolutionary Significance The skin shares features with ancient and extant reptiles, including a pebbled surface similar to crocodile skin, and hinged regions between epidermal scales that resemble skin structures in snakes and worm lizards. However, because the skin fossil is not associated with a skeleton or any other remains, it is not possible to identify what species of animal or body region the skin belonged to. The fact that this ancient skin resembles the skin of reptiles alive today shows how important these structures are for survival in terrestrial environments. “The epidermis was a critical feature for vertebrate survival on land,” says Mooney. “It’s a crucial barrier between the internal body processes and the harsh outer environment.” Potential Ancestral Link and Preservation The researchers say that this skin may represent the ancestral skin structure for terrestrial vertebrates in early amniotes that allowed for the eventual evolution of bird feathers and mammalian hair follicles. The skin fossil and other specimens were collected by lifelong paleontology enthusiasts Bill and Julie May at Richards Spur, a limestone cave system in Oklahoma that is an active quarry. The unique conditions at Richards Spur preserved many of the oldest examples of early terrestrial animals. The specimens are housed at the Royal Ontario Museum. When they saw the back of a lizard in the Southern Western Ghats, a group of scientists from the Thackeray Wildlife Foundation in India were reminded of Van Gogh’s The Starry Night. As soon as they figured out it was a new species, it was only apt to name it in honor of the famous painter. “Cnemaspis vangoghi is named for Dutch painter Vincent Van Gogh (1853–1890) as the striking coloration of the new species is reminiscent of one of his most iconic paintings, The Starry Night,” explains Ishan Agarwal, who took part in the study to describe the new lizard. Expedition Findings and Biodiversity Together with his fellow researchers Akshay Khandekar and Tejas Thackeray, they found the new species during an expedition in April 2022 to the Southern Western Ghats in Tamil Nadu, India. Now, they have published their findings in the peer-reviewed journal ZooKeys. “Tamil Nadu is an exceptionally biodiverse state and we expect to name well over 50 new species of lizards by the time we are done [with our expeditions]!” Ishan Agarwal says. “I also had more than 500 tick bites during that summer trip, with the highest densities in the low-elevation, dry forests of Srivilliputhur, where the new species are found,” he adds. Cnemaspis vangoghi is a small-sized gecko that can reach 3,4 cm in length. It was described as new to science together with another species of its genus, Cnemaspis sathuragiriensis, named for its type locality the Sathuragiri Hills. “The two new species are distributed in low elevation (250–400 m asl.), deciduous forests of Srivilliputhur, and add to the five previously known endemic vertebrates from Srivilliputhur-Megamalai Tiger Reserve, Tamil Nadu, India,” Ishan Agarwal explains. They are diurnal and mainly active during the cool hours of the early morning and evening, found largely on rocks. So far, they have only been found in very restricted localities, “an interesting case of micro-endemism in low-elevation species,” he notes. Credit: The Francis Crick Institute Scientists at the Francis Crick Institute, in collaboration with University of Kent, have used gene editing technology to create female-only and male-only mice litters with 100% efficiency. This proof of principle study, published in Nature Communications today (Friday, December 3rd, 2021), demonstrates how the technology could be used to improve animal welfare in scientific research and perhaps also agriculture. In scientific research and also farming, there is often a need for either male or female animals. For example, laboratory research into male or female reproduction requires only animals of the sex being studied. And in farming, only female animals are required for egg production and in dairy herds. This means it is common practice for animals of the unrequired sex to be culled after birth. The researchers’ new method uses a two-part genetic system to inactivate embryos shortly after fertilization, allowing only the desired sex to develop. Such a genetically-based method to control the sex of offspring could drastically reduce culling in both industries. The embryo selection is based on the fact that there are two elements of CRISPR-Cas9 – the Cas9 enzyme that cuts the DNA, allowing scientists to alter specific regions, and the guide RNA which carries the Cas9 to the right location on the genome. The team placed one element of the system on the father’s X or Y chromosome, meaning that it will only be inherited by female or male embryos respectively. The other element is contributed by the mother, and is inherited by all embryos. They targeted the Top1 gene, which is essential to DNA replication and repair. When an embryo formed from a sperm and egg, each containing one half of CRISPR-Cas9, the gene-editing was triggered in the embryo and it was not able to develop beyond a very early stage of around 16 to 32 cells. Using this method, the researchers were able to control, with 100% effect, the sex of a litter. To produce a male-only litter, the researchers edited the father’s X chromosome, meaning only females inherited the deleterious mutation, and for a female-only litter, they edited the Y chromosome. Surprisingly, the method did not lead to a 50% decrease in the number of offspring produced, rather the litter sizes were between 61% – 72% of the control litters. The researchers suggest this is because animals such as mice produce more eggs than required during each ovarian cycle, allowing for a proportion of them to be lost during early development without reducing litter size. This means that in situations where one sex is needed, fewer breeding animals will be required in order to produce the same number of the desired sex of offspring. As the Top1 gene is well conserved across mammals, these results may also be applicable to other animals. Charlotte Douglas, first author and former PhD student and postdoctoral scientist at the Crick, says: “This method works as we split the genome editing process in half, between a male and female, and it is only when the two halves meet in an embryo through breeding, that it is activated. Embryos with both halves cannot develop beyond very early cell stages. “We’ve also shown this process works successfully in different combinations – introducing either the Cas9 or the guide RNA elements on to the mother’s or father’s chromosomes.” As the offspring which survive only contain half of the CRISPR-Cas9 elements within their genome, this acts as a control preventing the sex-selection being passed down to further generations, unless they are selectively bred with an individual of the opposite sex containing the other half. This is different to genetic engineering through ‘gene-drive’ methods, which seek to spread a genetic mutation widely amongst a population. There are also no harmful effects of the gene edit in the surviving offspring. James Turner, author and group leader of the Sex Chromosome Biology Laboratory at the Crick says: “This work could have immediate and valuable impact in scientific laboratories, as we’ve shown how it is safe and effective in mice, a common mammal used in medical and scientific research. While a lot of research needs both sexes, there are areas of study where only one is needed. For example, when studying the reproductive system, sex-specific diseases, or certain hormones.” Peter Ellis, author and senior lecturer in molecular genetics and reproduction at University of Kent, says: “The implications of this work are potentially far-reaching when it comes to improving animal welfare, but should be considered at ethical and regulatory levels. “In particular, before any potential use in agriculture, there would need to be extensive public conversation and debate, as well as changes to legislation. On the scientific side, there is also much work to be done over a number of years.
