While a transient over-expression of the gene can permanently turn feet scales into feathers, it is much harder to disrupt feather development itself. Credit: Fabrice Berger & Michel Milinkovitch 2025 (CC-BY 4.0) Chickens treated to inhibit the sonic hedgehog (Shh) pathway during development grow feathers that resemble ancient protofeathers—unbranched and underdeveloped. While some recovery occurs after hatching, these findings reveal how critical Shh signaling is to feather patterning and offer compelling insights into how feathers might have evolved from simpler skin structures. The study also hints at the resilience of biological systems, as disrupted feather follicles can reactivate weeks later. Disrupting a Key Pathway in Feather Formation Blocking the sonic hedgehog (Shh) signaling pathway significantly disrupts feather development in chickens, limiting the growth, folding, and branching of feather buds. This finding comes from a study published on March 20th in PLOS Biology by Rory Cooper and Michel Milinkovitch of the University of Geneva, Switzerland. Feathers are complex structures that vary widely in shape across different bird species, body regions, and life stages. Their intricate forms make them an excellent model for studying how tissues develop in embryos. While the Shh pathway is known to play a key role in feather growth and patterning, direct experimental evidence of its function during feather formation has been scarce, until now. Visualizing Feather Growth with Advanced Imaging To fill this knowledge gap, Cooper and Milinkovitch used light sheet fluorescence microscopy imaging to study the normal patterning of embryonic feathers and how their shape develops. The authors also used precise intravenous injections of sonidegib to pharmacologically inhibit Shh pathway signaling during feather development at embryonic day 9, which precedes feather-bud outgrowth on the wings. This treatment temporarily modified Shh expression to produce striped domains instead of spots on the skin, temporarily stopped feather development, and resulted in unbranched and non-invaginated feather buds, akin to putative proto-feathers,until embryonic day 14. Feather Recovery and Evolutionary Implications Although feather development partially recovered later during development, hatched sonidegib-treated chickens exhibited naked regions of the skin surface with perturbed follicles. Remarkably, these follicles were subsequently reactivated by seven weeks post-hatching, highlighting the robustness of feather patterning as a developmental process. Overall, the study provides comprehensive functional evidence for the role of the Shh pathway in mediating feather development in chickens, supporting the idea that modified Shh signaling has contributed to the evolutionary diversification of feathers and other skin appendages such as feet scales. According to the authors, the study also demonstrates the importance of in-vivo experiments for obtaining a comprehensive understanding of developmental systems. Future Questions in Feather and Scale Evolution The authors add, “Our experiments show that while a transient disturbance in the development of feet scales can permanently turn them into feathers, it is much harder to disrupt feather development itself. DOI: 10.1371/journal.pbio.3003061 Researchers at the University of Illinois Chicago have discovered that a natural peptide from fruit flies, named drosocin, can bind to bacteria’s ribosomes and hinder protein production, thereby forcing bacterial cells to self-destruct. This finding, along with the understanding of its working mechanism, could facilitate the creation of new antibiotics. A New Study Reports on Peptide’s Antibacterial Mechanism Researchers from the University of Illinois Chicago have discovered that a peptide derived from fruit flies may pave the way for the development of novel antibiotics. Published in Nature Chemical Biology, their study reveals that the natural peptide, known as drosocin, safeguards the insect against bacterial infections by binding onto bacterial ribosomes. Once bound, drosocin prevents the ribosome from correctly completing its primary task — making new proteins, which cells need to function. Protein production can be halted by interfering with different stages of translation — the process by which DNA is “translated” into protein molecules. The UIC scientists discovered that drosocin binds to the ribosome and inhibits translation termination when the ribosome reaches the stop signal at the end of the gene. “Drosocin is only the second peptide antibiotic known to stop translation termination,” said Alexander Mankin, study author and Distinguished Professor from the Center for Biomolecular Sciences and the department of pharmaceutical sciences in the College of Pharmacy. The other, called apidaecin and found in honeybees, was first described by UIC scientists in 2017. Harnessing Bacterial Cells to Produce Peptide Mutants The UIC lab, which is co-run by Mankin and Nora Vázquez-Laslop, research professor in the College of Pharmacy, managed to produce the fruit fly peptide and hundreds of its mutants directly in bacterial cells. “Drosocin and its active mutants made inside the bacteria forced bacterial cells to self-destruct,” Mankin said. While the drosocin and apidaecin peptides work the same way, the researchers found that their chemical structures and the ways they bind to the ribosome are different. “By understanding how these peptides work, we hope to leverage the same mechanism for potential new antibiotics. Comparing side-by-side the components of the two peptides facilitates engineering new antibiotics that take the best from each,” Mankin said. Credit: Ecological Cascades Lab Rising Populations of Wild Pigs and Macaques Threaten Southeast Asia’s Forests Rising numbers of wild pigs and macaque monkeys in Southeast Asia pose risks to native forests and could lead to disease spread among livestock and humans, reveals a study led by The University of Queensland. Dr. Matthew Luskin, affiliated with UQ’s School of the Environment, alongside his team, gathered and assessed species population data from various parts of the region, some of it collected with a network of cameras. “Macaques and wild pigs are taking over Southeast Asia’s disturbed forests,” Dr Luskin said. “Humans are largely to blame for this by altering forests with logging and establishing palm oil farms which provide food and ideal breeding conditions for these animals. We saw that wild boar and macaque numbers were 400 percent higher in forests near the plantations than in untouched environments. These animals take full advantage of the farmland, raiding crops and thriving on calorie‐rich foods.” Setting and monitoring the camera traps provided Dr. Luskin with an up-close experience of the exploding numbers. “I encountered huge troops of macaques in Thailand, Malaysia, and Indonesia – they were everywhere in the forest edges, following us and interfering with our equipment,” Dr Luskin said. “At first it was frustrating but then was eerie as we became completely surrounded.” The Public Health Risks of Booming Mammal Populations Dr Luskin said there were significant human health risks in the rising pig and macaque populations. “The wildlife origins of the COVID-19 pandemic show that mammals in human-modified ecosystems often host high pathogen loads and pose serious zoonotic disease risks,” he said. “Both pigs and macaques are recognized as carriers of diseases that can be transmitted to people and they’re the most common species in a region considered to be the global zoonotic disease hotspot.” Collaborator, Professor Carlos Peres from the University of East Anglia (UK), said abnormally high populations of wildlife species that are disease reservoirs often occur in human-modified tropical forests. “This study again shows that densely settled rural areas in Southeast Asia may be a source of future human epidemics,” he said. Environmental Consequences of Unchecked Populations University of East Anglia and Southern University of Science and Technology (China) PhD candidate, Jonathan Moore, said the immediate effects of the population explosions could be seen on native flora in the affected regions. “Both pigs and macaques trigger negative cascading impacts in these pristine ecosystems,” Mr Moore said. “They kill the seeds and seedlings of native plants and eat bird and reptile eggs. The Malaysian pigs alone were found to reduce rainforest tree regeneration by 62 percent.” The researchers say action is needed to minimize population expansions of wild pigs and macaques. “Efforts to manage the populations of these species have failed in the past because of their rapid reproductive capacity and public outcry,” Dr Luskin said. “Nobody favors needless killing of wildlife but the negative social and ecological impacts from hyperabundant pest species do demand ethical and urgent management solutions.” Reference: “The rise of hyperabundant native generalists threatens both humans and nature” by Jonathan H.
