Older people become more susceptible to cold. Human evolution has provided us a level of protection from the existential threat of cold temperatures with the capacity to produce heat from fat stored in the body. However, with age, people become more susceptible to cold as well as inflammation and metabolic problems which can lead to a host of chronic diseases. Researchers at Yale and the University of California-San Francisco (UCSF) have found one culprit in this process — the same immune cells within fat that are designed to protect us from cold temperatures. In a new study, they find that the fat tissue of older mice loses the immune cell group 2 innate lymphoid cells (ILC2) which restore body heat in the presence of cold temperatures. But in a cautionary tale for those seeking easy treatments for diseases of aging, they also found that stimulating the production of new ILC2 cells in aging mice actually makes them more prone to cold-induced death. “What is good for you when you are young, can become detrimental to you as you age,” said Vishwa Deep Dixit, the Waldemar Von Zedtwitz Professor of Comparative Medicine and of Immunobiology and co-corresponding author of the study. The results are published today (September 1, 2021) in the journal Cell Metabolism. Dixit and his former colleague Emily Goldberg, now an assistant professor at UCSF, were curious about why fat tissue harbors immune system cells, which are usually concentrated in areas often exposed to pathogens like nasal passages, lungs, and skin. When they sequenced genes from cells of old and young mice they found that older animals lacked ILC2 cells, a deficit which limited their ability to burn fat and raise their body temperature in cold conditions. When scientists introduced a molecule that boosts the production of ILC2 in aging mice, the immune system cells were restored but the mice were surprisingly even less tolerant of cold temperatures. “The simple assumption is that if we restore something that is lost, then we are also going to restore life back to normal,” Dixit said. “But that is not what happened. Instead of expanding healthy cells of youth, the growth factor ended up multiplying the bad ILC2 cells that remained in fat of old mice.” But when researchers took ILC2 cells from younger mice and transplanted them into older mice, they found, the older animals’ ability to tolerate cold was restored. “Immune cells play a role beyond just pathogen defense and help maintain normal metabolic functions of life,” Dixit said. “With age, the immune system has already changed and we need to be careful how we manipulate it to restore the health of elderly.” Reference: “IL-33 causes thermogenic failure in aging by expanding dysfunctional adipose ILC2” by Emily L. The trade-off between developmental plasticity and growth rate helps to understand how fungi penetrate surfaces or plant/animal tissues, with important implications for fungal biotechnology, ecology, and studies of disease. Credit: University of Tsukuba University of Tsukuba research team sheds new light on how fungi that cause diseases can penetrate tissues by squeezing through tiny gaps between plant or animal cells. Fungi are a vital part of nature’s recycling system of decay and decomposition. Filamentous fungi spread over and penetrate surfaces by extending fine threads known as hyphae. Fungi that cause disease within living organisms can penetrate the spaces between tightly connected plant or animal cells, but how their hyphae do this, and why the hyphae of other fungal species do not, has been unclear. Now, a team led by Professor Norio Takeshita at University of Tsukuba, with collaborators at Nagoya University and in Mexico, has discovered a key feature that helps explain the differences among species. They compared seven fungi from different taxonomic groups, including some that cause disease in plants. The team tested how the fungi responded when presented with an obstruction that meant they had to pass through very narrow channels. At only 1 micron wide, the channels were narrower than the diameter of fungal hyphae, typically 2-5 microns in different species. Some species grew readily through the narrow channels, maintaining similar growth rates before meeting the channel, while extending through it, and after emerging. In contrast, other species were seriously impeded. The hyphae either stopped growing or grew very slowly through the channel. After emerging, the hyphae sometimes developed a swollen tip and became depolarized so that they did not maintain their previous direction of growth. The tendency to show disrupted growth did not depend on the diameter of the hyphae, or how closely related the fungi were. However, species with faster growth rates and higher pressure within the cell were more prone to disruption. By observing fluorescent dyes in the living fungi, the team found that processes inside the cell became defective in the fungi with disrupted growth. Small packages (vesicles) that supply lipids and proteins (needed for assembling new membranes and cell walls as hypha extend) were no longer properly organized during growth through the channel. “For the first time, we have shown that there appears to be a trade-off between cell plasticity and growth rate,” says Professor Takeshita. “When a fast-growing hypha passes through a narrow channel, a massive number of vesicles congregate at the point of constriction, rather than passing along to the growing tip. This results in depolarized growth: the tip swells when it exits the channel, and no longer extends. In contrast, a slower growth rate allows hyphae to maintain correct positioning of the cell polarity machinery, permitting growth to continue through the confined space.” As well as helping explain why certain fungi can penetrate surfaces or living tissues, this discovery will also be important for future research into fungal biotechnology and ecology. A new study conducted by an international team of researchers, including faculty from Binghamton University, State University of New York, indicates that Neanderthals underwent a significant decline in genetic diversity over time, a trend that may have contributed to their eventual extinction. Co-authored by Binghamton University Professor of Anthropology Rolf Quam and graduate student Brian Keeling, the study analyzed morphological diversity in the semicircular canals—inner ear structures crucial for balance. The researchers examined fossil specimens from two notable sites, Atapuerca (Spain) and Krapina (Croatia), along with other locations across Europe and western Asia. “The development of the inner ear structures is known to be under very tight genetic control, since they are fully formed at the time of birth,” said Quam. “This makes variation in the semicircular canals an ideal proxy for studying evolutionary relationships between species in the past since any differences between fossil specimens reflect underlying genetic differences. Credit: Allan Henderson, under CC BY 2.0 Neanderthal Evolution and Sample Comparisons The Atapuerca fossils, referred to as “pre-Neanderthals,” date to about 400,000 years ago and represent the earliest fossils that anthropologists consider clear Neanderthal ancestors. The Neanderthals emerged around 250,000 years ago from these populations which inhabited the Eurasian continent between 500,000 and 250,000 years ago. The Croatian site of Krapina represents the most complete collection of early Neanderthals and dates to approximately 130,000 years ago. The researchers calculated the amount of morphological diversity (i.e., disparity) of the semicircular canals of both samples, comparing them with each other and with a sample of “classic” Neanderthals of different ages and geographical origins. “It is exciting to be included in this research project which relies on some of the latest cutting-edge methodologies in our field,” said Keeling. “As a student of human evolution, I am always amazed at research that pushes the boundaries of our knowledge.” Schematic representation of the changes in morphological diversity along the evolutionary history of the Neanderthal clade. Credit: Alessandro Urciuoli, Institut Català de Paleontologia Miquel Crusafont Recent research based on ancient DNA samples extracted from fossils revealed the existence of a drastic loss of genetic diversity between early Neanderthals and the later “classic” Neanderthals. Technically known as a “bottleneck”, this genetic loss is frequently the consequence of a reduction in the number of individuals of a population. The ancient DNA data indicate that the decline in genetic variation took place approximately 110,000 years ago. The new study’s findings reveal that the morphological diversity of the semicircular canals of classic Neanderthals is clearly lower than that of pre-Neanderthals and early Neanderthals, which aligns with the ancient DNA results. Significance of the Findings The study was led by Alessandro Urciuoli (Institut Català de Paleontologia Miquel Crusafont, Universitat Autònoma de Barcelona) and Mercedes Conde-Valverde (director of the Cátedra de Otoacústica Evolutiva de HM Hospitales y la Universidad de Alcalá). Conde-Valverde, co-author of the study, emphasized the importance of the analyzed sample. “By including fossils from a wide geographical and temporal range, we were able to capture a comprehensive picture of Neanderthal evolution,” said Conde-Valverde. “The reduction in diversity observed between the Krapina sample and classic Neanderthals is especially striking and clear, providing strong evidence of a bottleneck event.” On the other hand, the results challenge the previously accepted idea that the origin of the Neanderthal lineage was associated with a significant loss of genetic diversity, prompting the need to propose new explanations for their origin. “We were surprised to find that the pre-Neanderthals from the Sima de los Huesos exhibited a level of morphological diversity similar to that of the early Neanderthals from Krapina,” said Alessandro Urciuoli, lead author of the study. “This challenges the common assumption of a bottleneck event at the origin of the Neanderthal lineage.” Reference: “Semicircular canals shed light on bottleneck events in the evolution of the Neanderthal clade” by Alessandro Urciuoli, Ignacio Martínez, Rolf Quam, Juan Luis Arsuaga, Brian A.
