Melatonin is used as a dietary supplement to promote sleep and get over jet lag, but nobody really understands how it works in the brain. Now, researchers at UConn Health show that melatonin helps worms sleep, too, and they suspect they’ve identified what it does in us. Our bodies produce melatonin in darkness. It’s technically a hormone, but you can readily buy melatonin as a supplement in pharmacies, nutrition stores, and other retail shops. It’s widely used by adults and often in children as well. Melatonin binds to melatonin receptors in the brain to produce its sleep-promoting effects. Think of a receptor as a keyhole, and melatonin as the key. The two keyholes for melatonin are called MT1 and MT2 in human brain cells. But scientists didn’t really know what happens when the keyhole is unlocked. Now UConn Health School of Medicine neuroscientists Zhao-Wen Wang and Bojun Chen and their colleagues have identified that process through their work with C. elegans worms, as reported in Proceedings of the National Academy of Sciences. When melatonin fits into the MT1 receptor in the worm’s brain, it opens a potassium channel known as the BK channel. A major function of the BK channel in neurons is to limit the release of neurotransmitters, which are chemical substances used by neurons to talk to each other. In their search for factors related to the BK channel, the Wang and Chen labs found that a melatonin receptor is needed for the BK channel to limit neurotransmitter release. They subsequently found that melatonin promotes sleep in worms by activating the BK channel through the melatonin receptor. Worms that lack either melatonin secretion, the melatonin receptor, or the BK channel spend less time in sleep. But Wait — Worms Sleep? Indeed they do, says Chen. There’s actually been quite a lot of research on worm sleep, and researchers found that sleep is similar between worms and mammals like humans and mice. Wang and Chen next plan to see if the melatonin-MT1-BK relationship holds in mice. The BK channel is involved in all kinds of bodily happenings, from epilepsy to high blood pressure. By learning more about the relationships between the BK channel, sleep, and behavioral changes, the researchers hope both to understand melatonin better and also help people who suffer from other diseases related to the BK channel. DOI: 10.1073/pnas.2010928117 Researchers have identified a biological mechanism involving the molecule KIBRA that explains the long-term stability of memories, shedding light on potential treatments for memory-related disorders. Pioneering study reveals a “molecular glue” critical for memory formation and stabilization. New research identifies the molecule KIBRA as a critical “glue” in stabilizing long-term memories by maintaining synaptic strength, offering insights into memory persistence despite ongoing cellular changes. Whether it’s a first-time visit to a zoo or when we learned to ride a bicycle, we have memories from our childhoods kept well into adult years. But what explains how these memories last nearly an entire lifetime? A new study in the journal Science Advances, conducted by a team of international researchers, has uncovered a biological explanation for long-term memories. It centers on the discovery of the role of a molecule, KIBRA, that serves as a “glue” to other molecules, thereby solidifying memory formation. “Previous efforts to understand how molecules store long-term memory focused on the individual actions of single molecules,” explains André Fenton, a professor of neural science at New York University and one of the study’s principal investigators. “Our study shows how they work together to ensure perpetual memory storage.” “A firmer understanding of how we keep our memories will help guide efforts to illuminate and address memory-related afflictions in the future,” adds Todd Sacktor, a professor at SUNY Downstate Health Sciences University and one of the study’s principal investigators. The Challenge of Synaptic Stability It’s been long-established that neurons store information in memory as the pattern of strong synapses and weak synapses, which determines the connectivity and function of neural networks. However, the molecules in synapses are unstable, continually moving around in the neurons, and wearing out and being replaced in hours to days, thereby raising the question: How, then, can memories be stable for years to decades? Memories are stored by the interaction of two proteins: a structural protein, KIBRA (green), that acts as a persistent synaptic tag, and a synapse-strengthening enzyme, protein kinase Mzeta (red). Drugs that disrupt the memory-perpetuating interaction (other colors) erase pre-established long-term and remote memories. Credit: Changchi Hsieh, Ph.D. In a study using laboratory mice, the scientists focused on the role of KIBRA, or kidney and brain expressed protein, the human genetic variants of which are associated with both good and poor memory. They focused on KIBRA’s interactions with other molecules crucial to memory formation—in this case, protein kinase Mzeta (PKMzeta). This enzyme is the most crucial molecule for strengthening normal mammalian synapses that is known, but it degrades after a few days. Their experiments reveal that KIBRA is the “missing link” in long-term memories, serving as a “persistent synaptic tag,” or glue, that sticks to strong synapses and to PKMzeta while also avoiding weak synapses. Mechanisms of Memory Retention “During memory formation the synapses involved in the formation are activated—and KIBRA is selectively positioned in these synapses,” explains Sacktor, a professor of physiology, pharmacology, anesthesiology, and neurology at SUNY Downstate. “PKMzeta then attaches to the KIBRA-synaptic-tag and keeps those synapses strong. This allows the synapses to stick to newly made KIBRA, attracting more newly made PKMzeta.” More specifically, their experiments in the Science Advances paper show that breaking the KIBRA-PKMzeta bond erases old memory. Previous work had shown that randomly increasing PKMzeta in the brain enhances weak or faded memories, which was mysterious because it should have done the opposite by acting at random locations, but the persistent synaptic tagging by KIBRA explains why the additional PKMzeta was memory enhancing, by only acting at the KIBRA tagged sites. “The persistent synaptic tagging mechanism for the first time explains these results that are clinically relevant to neurological and psychiatric disorders of memory,” observes Fenton, who is also on the faculty at NYU Langone Medical Center’s Neuroscience Institute. The paper’s authors note that the research affirms a concept introduced in 1984 by Francis Crick. Sacktor and Fenton point out that his proposed hypothesis to explain the brain’s role in memory storage despite constant cellular and molecular changes is a Theseus’s Ship mechanism—borrowed from a philosophical argument stemming from Greek mythology in which new planks replace old ones to maintain Theseus’s Ship for years. “The persistent synaptic tagging mechanism we found is analogous to how new planks replace old planks to maintain Theseus’s Ship for generations, and allows memories to last for years even as the proteins maintaining the memory are replaced,” says Sacktor. “Francis Crick intuited this Theseus’s Ship mechanism, even predicting the role for a protein kinase. Sklar Fund. Flower sex is an important factor when breeding for quality cultivars. Wines and table grapes exist thanks to a genetic exchange so rare that it’s only happened twice in nature in the last 6 million years. And since the domestication of the grapevine 8,000 years ago, breeding has continued to be a gamble. When today’s growers cultivate new varieties – trying to produce better-tasting and more disease-resistant grapes – it takes two to four years for breeders to learn whether they have the genetic ingredients for the perfect flower. Females set fruit, but produce sterile pollen. Males have stamens for pollen, but lack fruit. The perfect flower, however, carries both sex genes and can self-pollinate. These hermaphroditic varieties generally yield bigger and better-tasting berry clusters, and they’re the ones researchers use for additional cross-breeding. Now, Cornell scientists have worked with the University of California, Davis, to identify the DNA markers that determine grape flower sex. In the process, they also pinpointed the genetic origins of the perfect flower. Their paper, “Multiple Independent Recombinations Led to Hermaphroditism in Grapevine,” published on April 13, 2021, in the Proceedings of the National Academy of Science. “This is the first genomic evidence that grapevine flower sex has multiple independent origins,” said Jason Londo, corresponding author on the paper and a research geneticist in the USDA-Agricultural Research Service (USDA-ARS) Grape Genetics Unit, located at Cornell AgriTech. Londo is also an adjunct associate professor of horticulture in the School of Integrative Plant Science (SIPS), part of the College of Agriculture and Life Sciences. “This study is important to breeding and production because we designed genetic markers to tell you what exact flower sex signature every vine has,” Londo said, “so breeders can choose to keep only the combinations they want for the future.” Today, most cultivated grapevines are hermaphroditic, whereas all wild members of the Vitis genus have only male or female flowers. As breeders try to incorporate disease-resistance genes from wild species into new breeding lines, the ability to screen seedlings for flower sex has become increasingly important. And since grape sex can’t be determined from seeds alone, breeders spend a lot of time and resources raising vines, only to discard them several years down the line upon learning they’re single-sex varieties. In the study, the team examined the DNA sequences of hundreds of wild and domesticated grapevine genomes to identify the unique sex-determining regions for male, female and hermaphroditic species. They traced the existing hermaphroditic DNA back to two separate recombination events, occurring somewhere between 6 million and 8,000 years ago. Londo theorizes that ancient viticulturists stumbled upon these high-yielding vines and collected seeds or cuttings for their own needs – freezing the hermaphroditic flower trait in domesticated grapevines that are used today. Many wine grapes can be traced back to either the first or second event gene pool. Cultivars such as cabernet franc, cabernet sauvignon, merlot, and Thompson seedless are all from the first gene pool. The pinot family, sauvignon blanc, and gamay noir originate from the second gene pool. What makes chardonnay and riesling unique is that they carry genes from both events. Londo said this indicates that ancient viticulturalists crossed grapes between the two gene pools, which created some of today’s most important cultivars.
