Credit: OIST The Weevil Possesses Distinctive Characteristics and Is Sensitive to Human Disturbance A new weevil species has been found in the pristine subtropical forests of Ishigaki Island and Yanbaru National Park in Okinawa, Japan. The Ryukyu Islands, a chain of subtropical islands stretching between mainland Japan and Taiwan, are celebrated for their impressive biodiversity. With a unique evolutionary past due to their relative isolation, these islands house an array of intriguing insect species. Researchers at the Okinawan Institute of Science and Technology (OIST) have been placing net traps to monitor insects on Okinawa Island since 2015, and have captured a wide range of insects, including beetles, flies, wasps, and bees, which are preserved in ethanol, dried and stored in the OIST insect collection. The newly discovered beetle species, Acicnemis ryukyuana, was successfully identified through microscope analysis and dissection by OIST entomologist Jake H. Lewis, who works as Collection Manager in the OIST Environmental Science and Informatics Section. Environmental Science and Informatics Section (OIST) staff member Jake Lewis is working on the taxonomy and evolution of weevils. Currently, he is using a wide variety of techniques like microscopy, dissections, DNA analysis, and X-ray microtomography to find and describe new species across Asia. Credit: “When I arrived at OIST in 2022, I dove headfirst into the OIST weevil collection. As I closely examined them, this species immediately caught my eye. It clearly belonged to the genus Acicnemis, but was unlike anything else described from East Asia,” recounts Lewis. “Its elongated scales and unique coloration set this species apart from other known Japanese species.” The genus Acicnemis contains over 180 species, so confirming the discovery of a new species within this genus requires a thorough examination of the existing literature and museum collections. As the “type specimens” (the original specimens used for species description) in the genus Acicnemis are housed in European and Japanese museums, Lewis had to reach out to several institutions, including the Kyushu University Museum (Japan), the Natural History Museum in London (UK), and the Senckenberg German Entomological Institute (Germany) in order to authenticate the status of Acicnemis ryukyuana as a novel species. 3D models created using X-ray microtomography show the differences in leg shape between the newly discovered Acicnemis ryukyuana and Acicnemis postica. Credit: OIST Based on the current knowledge, Acicnemis ryukyuana is endemic to the Ryukyus. The entomologist named this species ryukyuana [from Ryukyu] and リュウキュウカレキゾウムシ [pronunciation: Ryuku-kareki-zoumushi, translation: “Ryukyu dead-tree weevil”] in Japanese to emphasize that it is an endemic element of the Ryukyu biodiversity. Weevils form one of the most diverse animal groups on the planet and generally feed on plants. Some weevil species are highly specialized and have a narrow range of plants they can feed on. The host plant(s) for this new species remain unknown, and Lewis hopes to conduct further field studies investigating this. The Distinctive Features of Acicnemis Ryukyuana Acicnemis ryukyuana can be immediately recognized by the yellow bands on its shoulders, and distinct pattern of grey, black, and yellow scales on its tough forewings. Other unique features visible under the microscope include the long scales (hairs) on the back and the shape of the last segment of the leg. Credit: OIST “Based on the unique set of features observed in this new species, A. ryukyuana appears to be closely related to some other species in southeast Asia, however, DNA analyses will be required to confirm this,” explains Lewis. “I was drawn to Okinawa as there are numerous undescribed weevil species in the region, unlike in Canada, my home country, where weevils have been much more thoroughly studied. Living in Okinawa and having Yanbaru National Park as a backyard is very exciting as it is home to many undescribed, endemic species.” enthuses Lewis. “The Ryukyu Islands offer an irresistible playground for taxonomists, rich in species which you only find here”. The Beetles’ Sensitivity to Human Presence Although OIST researchers distributed insect traps widely across Okinawa Island including heavily populated and disturbed areas, A. ryukyuana was only captured in a pristine, specially protected part of Yanbaru National Park. The new species was also collected in well-preserved subtropical forest areas on Ishigaki Island; and located by Lewis in the Kyushu University Museum collection. “Based on these collection locations, this weevil species appears to be very sensitive to human disturbance compared to other Acicnemis species commonly found in the Ryukyu Islands.” “This newly discovered beetle might be considered a vulnerable, endemic element of the Ryukyu fauna, similar to the flight-less bird Okinawa rail, the Yanbaru long-armed scarab beetle, and the Okinawa spiny rat,” says Lewis. “I am sure that taxonomists, conservation biologists, and local naturalists in Okinawa will be interested in knowing that yet another remarkable species has been discovered in the Ryukyu Islands.” Reference: “A New Species of Acicnemis Fairmaire, 1849 (Coleoptera: Curculionidae) from Okinawa and Ishigaki Islands, Okinawa Prefecture, Japan” by Jake H. The raw material for evolution is much more abundant in wild animals than was previously thought, according to new research from The Australian National University (ANU). Darwinian evolution is the process by which natural selection results in genetic changes in traits that favor the survival and reproduction of individuals. The rate at which evolution occurs depends crucially on genetic variations between individuals. An international research team, led by Dr. Timothée Bonnet from ANU, wanted to know how much of this genetic difference, or “fuel of evolution,” exists in wild animal populations. The answer: two to four times more than previously thought. “Being able to see so much potential change came as a surprise to the team.” Dr. Timothée Bonnet According to Dr. Bonnet, the process of evolution that Darwin described was an incredibly slow one. “However, since Darwin, researchers have identified many examples of Darwinian evolution occurring in just a few years,” Dr. Bonnet said. “A common example of fast evolution is the peppered moth, which prior to the industrial revolution in the UK was predominantly white. With pollution leaving black soot on trees and buildings, black moths had a survival advantage because it was harder for birds to spot them. “Because moth color determined survival probability and was due to genetic differences, the populations in England quickly became dominated by black moths.” Large-Scale Evaluation of Evolutionary Speed The research is the first time the speed of evolution has been systematically evaluated on a large scale, rather than on an ad hoc basis. The team of 40 researchers from 27 scientific institutions used studies of 19 populations of wild animals from around the world. These included superb fairy-wrens in Australia, spotted hyenas in Tanzania, song sparrows in Canada, and red deer in Scotland. “We needed to know when each individual was born, who they mated with, how many offspring they had, and when they died. Each of these studies ran for an average of 30 years, providing the team with an incredible 2.6 million hours of field data,” Dr. Bonnet said. “We combined this with genetic information on each animal studied to estimate the extent of genetic differences in their ability to reproduce, in each population.” “This research has shown us that evolution cannot be discounted as a process which allows species to persist in response to environmental change.” Dr. Timothée Bonnet After three years of trawling through reams of data, Dr. Bonnet and the team were able to quantify how much species change occurred due to genetic changes caused by natural selection. “The method gives us a way to measure the potential speed of current evolution in response to natural selection across all traits in a population. This is something we have not been able to do with previous methods, so being able to see so much potential change came as a surprise to the team,” Dr. Bonnet said. Professor Loeske Kruuk, also from ANU and now based at the University of Edinburgh in the United Kingdom, said: “This has been a remarkable team effort that was feasible because researchers from around the world were happy to share their data in a large collaboration. “It also shows the value of long-term studies with detailed monitoring of animal life histories for helping us understand the process of evolution in the wild.” However, the researchers warn it’s too early to tell whether the actual rate of evolution is getting quicker over time. “Whether species are adapting faster than before, we don’t know, because we don’t have a baseline. We just know that the recent potential, the amount of ‘fuel’, has been higher than expected, but not necessarily higher than before,” Dr. Bonnet said. According to the researchers, their findings also have implications for predictions of species’ adaptability to environmental change. Species Adaptability and Environmental Change “This research has shown us that evolution cannot be discounted as a process which allows species to persist in response to environmental change,” Dr. Bonnet said. Dr. Bonnet said that with climate change predicted to increase at an increasing rate, there is no guarantee that these populations will be able to keep up. “But what we can say is that evolution is a much more significant driver than we previously thought in the adaptability of populations to current environmental changes,” he said. Credit: Tomaž Einfalt, University of Basel Researchers at Sanford Burnham Prebys have shown that vesicles transporting between brain cells contain more complete instructions for altering cellular function than previously thought. In a study recently published in Cell Reports, researcher Jerold Chun, M.D., Ph.D. and his team found that the molecular instructions contained in these vesicles varied greatly among postmortem brain samples provided by individuals who had Alzheimer’s disease. Researchers call the tiny brain bubbles under scrutiny in this study small extracellular vesicles (sEVs). These tiny biological water balloons are produced by most cells in the body to ferry a wide variety of proteins, lipids, and byproducts of cellular metabolism, as well as RNA nucleic acid codes used by recipient cells to construct new proteins. Because this biologically active cargo can easily elicit changes in other cells, scientists are interested in brain sEVs as a medium for passing along normal as well as bungled instructions for misfolded proteins that accumulate in the brain as neurodegenerative diseases such as Alzheimer’s disease progress. Credit: The Chun lab To be a potential contributor to the buildup of unwanted proteins, sEVs would have to carry blueprints with sufficient information to enable other cells to produce the problematic proteins. Most previous research had indicated that the messenger RNA (mRNA) carrying plans for proteins were chopped into too many shorter fragments to allow recipient cells to change their construction patterns. “We found quite the opposite to be true in our study,” says Chun, professor in the Center for Genetic Disorders and Aging Research at Sanford Burnham Prebys. “We identified more than 10,000 full-length mRNAs through the use of a relatively newer DNA sequencing technique called PacBio long-read sequencing.” Detailed Findings from the Research The team isolated sEVs from the prefrontal cortex of 12 postmortem brain samples donated from patients diagnosed with Alzheimer’s disease and 12 from donors without Alzheimer’s disease (or any other known neurological disease). Nearly 80% of identified mRNAs were full-length, allowing them to be transcribed by recipient cells into viable proteins. “To corroborate the results of long-read sequencing in the human samples, we also looked at vesicles isolated from mouse cells,” says first author Linnea Ransom, Ph.D., a postdoctoral fellow at Sanford Burnham Prebys. “We found similar averages of between 78% and 86% full-length transcripts in three brain cell types: astrocytes, microglia, and neurons.” Negative-stain transmission electron microscopy was used to confirm the isolation of small extracellular vesicles in mouse neurons (shown above), as well as in mouse astrocytes and microglia. Credit: The Chun lab In addition to analyzing and validating the results regarding the length of mRNAs in brain sEVs, the researchers compared the sequence of genes reflected in the sEV mRNA transcriptome. In Alzheimer’s disease samples, 700 genes showed increased expression whereas nearly 1500 were found to have reduced activity. The scientists determined that the 700 upregulated genes are associated with inflammation and immune system activation, which fits within known patterns of brain inflammation present in neurodegenerative diseases such as Alzheimer’s disease. The researchers also found many genes associated with Alzheimer’s disease in prior genome-wide association studies also were present in Alzheimer’s disease sEVs. “The changes in gene expression contained in these vesicles reveal an inflammatory signature that may serve as a window into disease processes occurring in the brain as Alzheimer’s disease progresses,” says Chun. Following this study, Chun and team will dig deeper into how cells package sEVs and how the enclosed mRNA codes lead to functional changes in other brain cells affected in Alzheimer’s disease. Better understanding of sEVs and their mRNA contents may enable the discovery of biomarkers that could be used to improve early detection of Alzheimer’s disease and potentially other neurological conditions, while identifying new disease mechanisms to provide new therapeutic targets. “Additionally, sEVs naturally occur as a vehicle for transporting biologically active cargo between cells, so it also may be possible to leverage them as a targeted delivery system for future brain therapies,” says Chun.
