A new study by the Salk Institute presents a groundbreaking non-hormonal and reversible male contraceptive method using HDAC inhibitors to block sperm production without affecting libido. This method, targeting the regulation of gene expression in sperm production, promises fewer side effects and fully reversible fertility, indicating a significant advance in the development of male contraceptives. Administering an HDAC inhibitor orally stopped sperm production and fertility in mice without impacting their libido. Surveys show most men in the United States are interested in using male contraceptives. However, their choices are currently restricted to the less reliable condoms or the more intrusive vasectomies. Efforts to create medications that inhibit sperm production, development, or the ability to fertilize have so far achieved modest success, often resulting in partial efficacy or significant adverse effects. The complexity of sperm development poses a significant challenge for scientists attempting to innovate in the field of male contraception, as finding aspects of the process that can be altered safely and effectively remains difficult. Now, scientists at the Salk Institute have found a new method of interrupting sperm production, which is both non-hormonal and reversible. The study, published in Proceedings of the National Academy of Sciences (PNAS) on February 20, 2024, implicates a new protein complex in regulating gene expression during sperm production. The researchers demonstrate that treating male mice with an existing class of drugs, called HDAC (histone deacetylase) inhibitors, can interrupt the function of this protein complex and block fertility without affecting libido. A Novel Approach “Most experimental male birth control drugs use a hammer approach to blocking sperm production, but ours is much more subtle,” says senior author Ronald Evans, professor, director of the Gene Expression Laboratory, and March of Dimes Chair in Molecular and Developmental Biology at Salk. “This makes it a promising therapeutic approach, which we hope to see in development for human clinical trials soon.” Sperm, pictured inside the cross-sectioned tube of the epididymis, were not generated while mice took the HDAC inhibitor drug (top right), but after 60 days off the drug, spermatogenesis was recovered (bottom right). To do this, sperm stem cells in the testes continuously make more of themselves, until a signal tells them it’s time to turn into sperm—a process called spermatogenesis. This signal comes in the form of retinoic acid, a product of vitamin A. Pulses of retinoic acid bind to retinoic acid receptors in the cells, and when the system is aligned just right, this initiates a complex genetic program that turns the stem cells into mature sperm. Salk scientists found that for this to work, retinoic acid receptors must bind with a protein called SMRT (silencing mediator of retinoid and thyroid hormone receptors). SMRT then recruits HDACs, and this complex of proteins goes on to synchronize the expression of genes that produce sperm. Targeted Intervention without Side Effects Previous groups have tried to stop sperm production by directly blocking retinoic acid or its receptor. But retinoic acid is important to multiple organ systems, so interrupting it throughout the body can lead to various side effects—a reason many previous studies and trials have failed to produce a viable drug. Evans and his colleagues instead asked whether they could modulate one of the molecules downstream of retinoic acid to produce a more targeted effect. The researchers first looked at a line of genetically engineered mice that had previously been developed in the lab, in which the SMRT protein was mutated and could no longer bind to retinoic acid receptors. Without this SMRT-retinoic acid receptor interaction, the mice were not able to produce mature sperm. However, they displayed normal testosterone levels and mounting behavior, indicating that their desire to mate was not affected. Credit: Salk Institute To see whether they could replicate these genetic results with pharmacological intervention, the researchers treated normal mice with MS-275, an oral HDAC inhibitor with FDA breakthrough status. By blocking the activity of the SMRT-retinoic acid receptor-HDAC complex, the drug successfully stopped sperm production without producing obvious side effects. Another remarkable thing also happened once the treatment was stopped: Within 60 days of going off the pill, the animals’ fertility was completely restored, and all subsequent offspring were developmentally healthy. Reversible Effects and Future Implications The authors say their strategy of inhibiting molecules downstream of retinoic acid is key to achieving this reversibility. Think of retinoic acid and the sperm-producing genes as two dancers in a waltz. Their rhythm and steps need to be coordinated with each other for the dance to work. But if you throw something in that makes the genes miss a step, the two are suddenly out of sync and the dance falls apart. In this case, the HDAC inhibitor causes the genes’ misstep, halting the dance of sperm production. However, if the dancer can find its footing and get back in step with its partner, the waltz can resume. In the same way, the authors say that removing the HDAC inhibitor allows the sperm-producing genes to get back in sync with the pulses of retinoic acid, turning sperm production back on as desired. “It’s all about timing,” says co-author Michael Downes, a senior staff scientist in Evans’ lab. “When we add the drug, the stem cells fall out of sync with the pulses of retinoic acid, and sperm production is halted, but as soon as we take the drug away, the stem cells can reestablish their coordination with retinoic acid and sperm production will start up again.” The authors say the drug doesn’t damage the sperm stem cells or their genomic integrity. While the drug was present, the sperm stem cells simply continued regenerating as stem cells, and when the drug was later removed, the cells could regain their ability to differentiate into mature sperm. “We weren’t necessarily looking to develop male contraceptives when we discovered SMRT and generated this mouse line, but when we saw that their fertility was interrupted, we were able to follow the science and discover a potential therapeutic,” says first author Suk-Hyun Hong, a staff researcher in Evans’ lab. “It’s a great example of how Salk’s foundational biological research can lead to major translational impact.” Reference: “Targeting nuclear receptor corepressors for reversible male contraception” by Suk-Hyun Hong, Glenda Castro, Dan Wang, Russell Nofsinger, Maureen Kane, Alexandra Folias, Annette R. Rainbow Boa Boa constrictors breathe with different sections of rib cage when constricting prey and digesting dinner. The later stages of pregnancy can make life difficult as the fetus presses against the diaphragm making it hard to breathe. But snakes that constrict their victims before swallowing them whole have to overcome the challenges of breathing while their lungs are restricted each time they dine. “With no diaphragm, they rely entirely on motions of their ribs,” says John Capano (Brown University, USA), adding that the earliest snake ancestors must have overcome the challenge of breathing while squeezing and digesting dinner. But it wasn’t clear how modern snakes save themselves from suffocating while constricting their victims. One possibility was that the animals adjust which region of the ribcage they use to inhale, depending on whether they are resting, throttling an animal or digesting. But no one had monitored in detail the breathing patterns of snakes in the act of subduing their dinner to check whether the animals can adjust which section of the ribcage they use. Capano and Elizabeth Brainerd (Brown University) secured a blood pressure cuff around the ribs of boa constrictors to restrict their movements and discovered that the sinuous reptiles use different sections of the rib cage to breathe when their ribs are constricted. They publish their discovery that the hind section of the lung works like a bellows, pulling air into the lung when the ribs further forward can no longer move because they are squeezing prey to death, in Journal of Experimental Biology. But first, Capano attached minute metal markers to two ribs in each reptile – one a third of the way down the snake’s body and another halfway along – to visualize how the ribs moved using X-rays. Then he positioned a blood pressure cuff over the ribs in both regions and gradually increasing the pressure to immobilize them. “Either the animals did not mind the cuff or became defensive and hissed to try to get the researcher to leave,” recalls Capano, explaining that the reptiles really fill their lungs when hissing: “this was an opportunity to measure some of the biggest breathes snakes take,” he says. Ribs as Bellows: How Snakes Adapt Their Breathing Reconstructing the boa constrictors’ rib movements, it was clear that the animals were able to control the movements of ribs in different portions of the rib cage independently. When the boa constrictors were gripped by the blood pressure cuff a third of the way along the body, the animals breathed using the ribs further back, swinging the ribs backward while tipping them up to draw air into the lungs. However, when the ribs toward the rear of the lung were constricted, the snakes breathed using the ribs closer to the head. In fact, the ribs at the far end of the lung only moved when the forward ribs were gripped, drawing air deep into the region, even though it has a poor blood supply and does not provide the body with oxygen. The far end of the lung, was behaving like a bellows, pulling air through the front section of the lung when it could no longer breathe for itself. In addition, Capano, Scott Boback and Charles Zwemer (both from Dickinson College, USA), filmed and recorded the nerve signals controlling the rib muscles when constricted by the blood pressure cuff, while Boback also filmed a snake with a GoPro as it dined, revealing that the ribs were not simply being held immobile. There were no nerve signals in the constricted muscles; the snakes had shifted to breathing by activating a different set of ribs further along the body. Evolutionary Implications of Breathing Adaptations in Snakes As subduing and digesting a victim is one of the most energetic things these snakes can do, it was probably essential that they evolved the ability to adjust where they breathe before adopting their new rib-hindering lifestyle, to ensure that they didn’t suffocate themselves. “It would have been difficult for snakes to evolve those behaviors without the ability to breathe,” he concludes. Bhattacharya How to Identify Heat-Stressed Corals Researchers have found a novel way to identify heat-stressed corals, which could help scientists pinpoint the coral species that need protection from warming ocean waters linked to climate change, according to a Rutgers-led study. “This is similar to a blood test to assess human health,” said senior author Debashish Bhattacharya, a Distinguished Professor in the Department of Biochemistry and Microbiology in the School of Environmental and Biological Sciences at Rutgers University–New Brunswick. “We can assess coral health by measuring the metabolites (chemicals created for metabolism) they produce and, ultimately, identify the best interventions to ensure reef health. Coral bleaching from warming waters is an ongoing worldwide ecological disaster. Therefore, we need to develop sensitive diagnostic indicators that can be used to monitor reef health before the visible onset of bleaching to allow time for preemptive conservation efforts.” Coral reefs provide habitat, nursery, and spawning grounds for fish, food for about 500 million people along with their livelihoods, and coastline protection from storms and erosion. But global climate change threatens corals by warming ocean waters, resulting in coral bleaching and disease. Other threats to corals include sea-level rise, a more acidic ocean, unsustainable fishing, damage from vessels, invasive species, marine debris, and tropical cyclones, according to the National Oceanic and Atmospheric Administration. The study, published in the journal Science Advances, examined how Hawaiian stony corals respond to heat stress, with a goal of identifying chemical (metabolite) indicators of stress. Heat stress can lead to the loss of algae that live in symbiosis with corals, resulting in a white appearance (bleaching) and, potentially, the loss of reefs. Scientists subjected the heat-resistant Montipora capitata and heat-sensitive Pocillopora acuta coral species to several weeks of warm seawater in tanks at the Hawaiʻi Institute of Marine Biology. Then they analyzed the metabolites produced and compared them with other corals not subjected to heat stress. “Our work, for the first time, identified a variety of novel and known metabolites that may be used as diagnostic indicators for heat stress in wild coral before or in the early stages of bleaching,” Bhattacharya said. The scientists are validating their coral diagnosis results in a much larger study and the results look promising. The scientists are also developing a “coral hospital” featuring a new lab-on-a-chip device, which could check coral health in the field via metabolite and protein indicators.
