According to the researchers, bacterial cellulose enables microbial life on Mars. A team of researchers from Göttingen University investigates kombucha cultivation in extraterrestrial environments. The possibility of kombucha cultures surviving under Martian-like environments has been studied by an international research team that includes the University of Göttingen. Kombucha, also known as tea fungus or mushroom tea, is a popular beverage that is made by fermenting sugared tea using kombucha cultures, a symbiotic culture of bacteria and yeast. Surprisingly, a bacterial species that produces cellulose persisted despite the simulated Martian atmosphere destroying the kombucha cultures’ microbial ecology. The findings were published in the journal Frontiers in Microbiology. Kombucha Cultures in Space In 2014, with the help of the European Space Agency, the researchers working on the “Biology and Mars Experiment” (BIOMEX) project launched kombucha cultures to the International Space Station (ISS). Credit: European Space Agency (ESA) The objective was to get a better understanding of cellulose’s resilience as a biomarker, kombucha’s genomic structure, and its extraterrestrial survival behavior. The samples were reactivated on Earth and cultured for another two and a half years after one and a half years under simulated Martian conditions outside the ISS. Working alongside researchers from the University of Minas Gerais in Brazil, Professor Bertram Brenig, director of the University of Göttingen’s Institute of Veterinary Medicine, was in charge of sequencing and bioinformatic analysis of the metagenomes of the reactivated cultures and individual kombucha cultures. “Based on our metagenomic analysis, we found that the simulated Martian environment drastically disrupted the microbial ecology of kombucha cultures. Credit: European Space Agency (ESA) Cellulose as a Biomarker for Extraterrestrial Life The results suggest that the cellulose produced by the bacteria is probably responsible for their survival in extraterrestrial conditions. This also provides the first evidence that bacterial cellulose could be a biomarker for extraterrestrial life and cellulose-based membranes or films could be a good biomaterial for protecting life and producing consumer goods in extraterrestrial settlements. Another interesting aspect of these experiments could be the development of novel drug delivery systems, for example, the development of medicine suitable for use in space. Another focus was on investigations into changes in antibiotic resistance: the research team was able to show that the total number of antibiotic and metal resistance genes – meaning that these microorganisms might survive despite antibiotics or metals in the environment – were enriched in the exposed cultures. “This result shows that the difficulties associated with antibiotic resistance in medicine in space should be given special attention in the future,” the scientists said. Genetically modifying mosquitoes to express antimalarial genes and pass them on to their offspring is being tested as a new strategy to eliminate malaria. Altering a mosquito’s gut genes to make them spread antimalarial genes to the next generation of their species shows promise as an approach to curb malaria, suggests a preliminary study published today (April 13, 2021) in eLife. The study is the latest in a series of steps toward using CRISPR-Cas9 gene-editing technology to make changes in mosquito genes that could reduce their ability to spread malaria. If further studies support this approach, it could provide a new way to reduce illnesses and deaths caused by malaria. Growing mosquito resistance to pesticides, as well as malaria parasite resistance to antimalarial drugs, has created an urgent need for new ways to fight the disease. Gene drives are being tested as a new approach. They work by creating genetically modified mosquitoes that, when released into the environment, would spread genes that either reduce mosquito populations or make the insects less likely to spread the malaria parasite. But scientists must prove that this approach is safe and effective before releasing genetically modified mosquitoes into the wild. “Gene drives are promising tools for malaria control,” says first author Astrid Hoermann, Research Associate at Imperial College London, UK. “But we wanted a clear pathway for safely testing such tools in countries where the disease most commonly occurs.” In the study, Hoermann and colleagues genetically modified the malaria-transmitting mosquito Anopheles gambiae. They used the CRISPR-Cas9 technology to insert a gene that encodes an antimalarial protein amidst genes that are turned on after the mosquito eats a blood meal. The team did this in a manner that allowed the whole section of DNA to also function as a gene drive that could be passed on to most of the mosquitoes’ offspring. They initially inserted the gene along with a fluorescent marker to help them track it in three different spots in the DNA, and then later removed the marker, leaving only a minor genetic modification behind. Next, the team bred the mosquitoes to see if they were able to successfully reproduce and remain healthy. They also tested how well the malaria parasite developed in the mosquitoes’ guts. Their experiments provide preliminary evidence that this approach to genetic modifications could create successful gene drives. “These genetic modifications are passive, and could be tested in the field and undergo a stringent regulatory process to ensure they are safe and effective in blocking the parasite without raising concerns of accidental spread in the environment,” explains senior author Nikolai Windbichler, Senior Lecturer at the Department of Life Sciences, Imperial College London. “However, once we combine them with other mosquitoes with an active gene drive, they turn into gene drives themselves without the need for any further changes. The research, by the University of Exeter, suggests that the removal of old male elephants, which are often the targets of trophy hunting, could lead to increased human-wildlife conflict. The study, in collaboration with Elephants for Africa, examined the behavior of 281 male elephants in an all-male area in Makgadikgadi Pans National Park, Botswana, over a period of three years. The elephants were divided into four age groups, adolescents (10-15 and 16-20 years), and adults (21-25 and 26+ years). The results found that with fewer old bull elephants around, elephants were more likely to be aggressive towards non-elephant targets such as vehicles, livestock, and other species. Male elephants. Credit: Connie Allen The adolescent elephants, in particular, were more aggressive and fearful to non-elephant targets when they were alone compared to with other males. This indicates that socially isolated adolescents may also be an increased threat to people. “Our research draws attention to what is often a rather overlooked area in animal behavior; that of the complex relationships and connections that occur between males in non-breeding all-male societies,” said lead author Connie Allen of Exeter’s Centre for Research in Animal Behaviour. “It appears the presence of more knowledgeable, older elephants in groups may play a key role in keeping the younger, less experienced males calm and lowering their perception of their current threat level, which means there’s less risk of aggression towards humans and other species. “Alternatively, older bulls may police other males aggression directed toward non-elephant targets.” Male elephants. Credit: Connie Allen Professor Darren Croft, of the University of Exeter, said understanding the causes of aggression in male elephants is essential for reducing human-elephant conflict. “Old male bull elephants are often thought of as redundant and are targeted for trophy hunting,” he said. “These new results highlight the important role that old male elephants can play in shaping the behavior of younger males, which are more aggressive in the absence of old bulls — including towards vehicles. “These findings provide an important message for wildlife managers and suggest that the removal of old male elephants from populations could lead to an increase in human-wildlife conflict.” Associate Professor Lauren Brent, of the University of Exeter, said: “Elephants are highly social animals.
