New researchers reveals that lonely individuals have distinct and varied brain processing patterns when compared to those who aren’t lonely, which may contribute to their feelings of isolation. Despite their number of friends, individuals with high loneliness levels exhibited these unique brain responses, suggesting that it’s not about the quantity of social connections, but the quality and shared understanding. A researcher from USC Dornsife in psychology comparing brain images has found significant differences in the brain processing patterns of lonely individuals when compared to those who aren’t lonely. The Russian writer and philosopher Leo Tolstoy may have been onto something when he wrote the opening line of Anna Karenina: “Happy families are all alike; every unhappy family is unhappy in its own way.” A recent study published in Psychological Science and led by a scholar now at the USC Dornsife College of Letters, Arts, and Sciences, suggests that while individuals who aren’t experiencing loneliness exhibit similar patterns in brain information processing, those who are lonely seem to interpret the world in a manner that is distinctly unique to each individual. Copious research shows that loneliness is detrimental to well-being and is often accompanied by self-reported feelings of not being understood by others. A recent report from the United States Surgeon General’s office referred to loneliness as a public health crisis in reaction to the growing number of adults suffering from this condition. Even before the onset of the COVID-19 pandemic, approximately half of U.S. adults reported experiencing measurable levels of loneliness. Loneliness Is Idiosyncratic While she was a postdoctoral fellow at UCLA, Elisa Baek, assistant professor of psychology at USC Dornsife, sought to better understand what contributes to such feelings of disconnection and being misunderstood. Baek and her team used a neuroimaging technique called functional magnetic resonance imaging (fMRI) to examine the brains of 66 first-year college students while they watched a series of video clips. The videos ranged in topic from sentimental music videos to party scenes and sporting events, providing a diverse array of scenarios for analysis. Before being scanned, the participants, who ranged in age from 18 to 21, were asked to complete the UCLA Loneliness Scale, a survey that measures a person’s subjective feelings of loneliness and feelings of social isolation. Based on the survey results, the researchers separated the participants into two groups: lonely and “nonlonely” (those not experiencing loneliness). They then scanned each participant’s brain using fMRI as the participant watched the videos. Comparing the brain imaging data between the two groups, the researchers discovered that lonelier individuals exhibited more dissimilar and idiosyncratic brain processing patterns than their non-lonely counterparts. This finding is significant because it reveals that neural similarity, which refers to how similar the brain activity patterns of different individuals are, is linked to a shared understanding of the world. This shared understanding is important for establishing social connections. People who suffer from loneliness are not only less similar to society’s norm of processing the world, but each lonely person differs in unique ways, as well. That uniqueness may further impact the feelings of isolation and lacking social connections. Baek said, “It was surprising to find that lonely people were even less similar to each other.” The fact that they don’t find commonality with lonely or nonlonely people makes achieving social connection even more difficult for them. “The ‘Anna Karenina principle’ is a fitting description of lonely people, as they experience loneliness in an idiosyncratic way, not in a universally relatable way,” she added. Loneliness Isn’t About Having or Not Having Friends So, does idiosyncratic processing in lonely individuals cause loneliness, or is it a result of loneliness? The researchers observed that individuals with high levels of loneliness — regardless of how many friends or social connections they had — were more likely to have idiosyncratic brain responses. This raised the possibility that being surrounded by people who see the world differently from oneself may be a risk factor for loneliness, even if one socializes regularly with them. The study also suggests that because social connections or disconnections fluctuate over time, it may influence the extent to which an individual processes the world idiosyncratically. Looking forward, Baek said she is interested in examining people who have friends and are socially active but still feel lonely. In addition, the researchers are looking at what particular situations lonely individuals process differently. By examining RNA in hundreds of thousands of individual brain cells, Scripps Research scientists further support that alcohol use disorder could accelerate Alzheimer’s disease progression, paving the way for future targeted treatments. New research from Scripps Research links Alzheimer’s disease and alcohol use disorder, showing similar gene expression changes in the brain that impact inflammation and cell function. Using single-cell transcriptomics, the study indicates that alcohol use may exacerbate Alzheimer’s disease by affecting shared molecular pathways. Larger datasets are anticipated to provide further insights. Gene Expression Links Between Alzheimer’s and Alcohol Use Nearly 7 million Americans have Alzheimer’s disease, and this number is predicted to double by 2060. While aging and genetic predisposition are the most important risk factors for Alzheimer’s, epidemiological studies suggest that lifestyle factors including alcohol use could also impact disease onset and progression. Now, Scripps Research scientists have shown that Alzheimer’s and alcohol use disorder (AUD) are associated with similarly altered gene expression patterns in the brain, supporting the idea that alcohol use may promote Alzheimer’s disease progression. The study, published in eNeuro on September 19, 2024, could inform future preventative and treatment strategies. Shared Molecular Mechanisms in Alzheimer’s and Alcohol Use Disorder “We found several cell-type-specific genes and pathways that are dysregulated in both Alzheimer’s disease and alcohol, which supports the hypothesis that alcohol use disorder can accelerate Alzheimer’s disease progression by impinging on some of the same molecular mechanisms that are affected by Alzheimer’s,” says senior author Pietro Paolo Sanna, MD, a professor in the Immunology and Microbiology Department at Scripps Research. “By understanding these dysregulations with this level of molecular detail, we can understand what’s causing these diseases, and we can also identify targets that could be used therapeutically.” This is the first time researchers have used single cell transcriptomics—a method that analyzes gene expression within individual cells by sequencing their RNA—to compare changes associated with Alzheimer’s disease and AUD in different populations of human brain cells. The study builds upon previously published research in the Sanna lab that showed that excessive alcohol consumption accelerates Alzheimer’s progression in mice that are genetically predisposed to the disease. Researchers characterized the gene expression of more than 100,000 individual cells from brains of humans with Alzheimer’s disease and compared the patterns to those in individuals with alcohol use disorder (AUD). Compared to healthy individuals, they showed that there was significant overlap in gene expression changes for those with Alzheimer’s and AUD. Pathways that were downregulated in AUD and at least one AD stage are shown in purple, and pathways that are upregulated in AUD and at least one AD stage are shown in pink. Sanna (Scripps Research) Study Methodology and Comparative Gene Analysis To examine cell-specific gene expression changes, the team analyzed RNA sequencing data from hundreds of thousands of individual brain cells from 75 patients with varying stages of Alzheimer’s disease (early, intermediate or advanced), and 10 patients without Alzheimer’s. Then, they compared this Alzheimer’s gene expression data with previously published RNA sequencing data from individuals with AUD. They showed that both AUD and Alzheimer’s are associated with similar gene expression changes in the brain, including upregulation of inflammatory genes and pathways, disruption to cell signaling and cell-death-related pathways, and changes to blood vessel cells. The Broader Implications of Alcohol Use as a Risk Factor “What we’ve presented here is a differential analysis of two disorders that cause cognitive decline,” says first author Arpita Joshi, PhD, a staff scientist in Sanna’s lab at Scripps Research. “It deepens our understanding of Alzheimer’s disease and what the three clinically defined stages of Alzheimer’s entail, and it underscores the importance of considering alcohol use disorder as a risk factor for Alzheimer’s.” Because the study was based on a small sample size for AUD, in the future, researchers plan to repeat their analysis using larger gene expression databases from individuals with AUD, which they expect to become available in the next year. Bacteria do not sexually reproduce, but that does not stop them from exchanging genetic information as it evolves and adapts. During conjugal transfer, a bacterium can connect to another bacterium to pass along DNA and proteins. Escherichia coli bacteria, commonly called E. coli, can transfer at least one of these gene-containing plasmids to organisms across taxonomic kingdoms, including to fungi and protists. Now, researchers from Hiroshima University have a better understanding of this genetic hat trick, which has potential applications as a tool to promote desired characteristics or suppress harmful ones across genetic hosts. They recently published their results in Frontiers in Microbiology. Plasmids transfer from one bacterium — the donor — to another — the recipient. A particular kind of plasmid, called IncP1, can be hosted by a variety of bacteria and, seemingly as a result of its broad hosts, can transfer DNA to recipients beyond bacteria. The hypothesis is that the plasmid contains genes cultivated from different hosts and donors, resulting in this unique ability. “Although conjugation factors encoded on plasmids have been extensively analyzed, those on the donor chromosome have not,” said paper author Kazuki Moriguchi, associate professor, Program of Basic Biology, Graduate School of Integrated Sciences for Life, Hiroshima University. There have been some studies on the various genes, according to Moriguchi, but the function of the genes was not examined, so it is not clear how they were related to the conjugation mechanism. In this study, the researchers conducted a genome-wide survey on an extensive collection of bacteria mutants as donors to yeast. The mutants were engineered to have specific genes “knocked out” in order to study how the overall system performs without the presence of that specific gene, allowing researchers to infer information about the gene’s function. “We focused on ‘up’ mutants that have the ability to accelerate conjugative transfer to both prokaryotes and eukaryotes as they could be potent donor strains applicable to gene introduction tools,” Moriguchi said, noting how IncP1’s ability to transmit genetic material across kingdoms could be used to develop precise tools to introduce genes capable of changing how the bacteria perform certain functions or react to changes in their environments. Out of 3,884 mutants surveyed, three were identified that could conjugate across E. coli or from E. coli to yeast without accumulating genetic material, indicating that the genes worked together. The researchers analyzed the genes but were unable to elucidate the exact target or targets of conjugation mechanism that allows for cross-kingdom transfer. However, their analysis did reveal how the genes appear to work. Two of the genes work to repress the unknown target in the E. coli donor. Simultaneously, the third gene is inactivated, allowing another unknown target to resume activity. “The results suggest that the unknown target factors of these three genes form a complex in order to activate or repress the conjugation, either directly or indirectly at an identical step or steps of the IncP1 conjugation machinery, although the exact mechanism beyond this phenomenon remains unknown,” Moriguchi said. According to Moriguchi, the data collected in this study can help facilitate the breeding of donor strains from various bacteria, each of which carries a high affinity with target organisms in addition to having a high conjugation ability.
